Objective To investigate a 272 base pair section of the 5' non coding region of genomic DNA from the peripheral blood monounuclear cells of h.a.thy hepatitis virus C (hCV) negative human subjects (not patients) Mothods This sequence section bears interest because ① it harbors several potential methylation (Cp rich) sites, and ② it represents the largest part of its internal ribosomal entry site A pre PCR digestion protocol was established making consistent use of four restriction endonucleases selected for certain features: SmaI, XmaCI, MspI, and h.a.I are inhibited if methylation(s) are present at certain cytosines within their cutting sequences Results The suspected hCV specific sequence was found in the DNA of each subject tested The pre PCR digestion assay reveals individual differences in their pattern of methylation, which may be due to possible epigenetic phenomena Conclusions The results provide formal proof that these hCV specific sequences are contained in the genomic or extra chromosomal target DNA, and probably belong to a new class of endogenous sequences

Objective: To clarify the association of hepatitis Bvirus mutants in precore and core promoter regions inpatients with hepatic failure and hBeAg ***: Precore and core promoter regions of 25hBV isolates from the patients with hepatic failurewere analyzed by polymerase chain reaction (PCR)direct sequencing ***: Precore G-to-A^(1896) mutants were identified in16 (64%) of the 25 isolates. The 'hot spot' mutationsat A-to-T^(1762) and G-to-A^(1764) were present together in19 (76%) of the 25 isolates, while C-to-T^(1653) and T-to-C^(1753) existed in a mutually exclusive manner andmore frequently in hepatic failure with liver cirrhosisgroup than in hepatic failure with chronic hepatitisgroup (100% vs 50%). Both.a.(1896) and T^(1762)-A^(1764)could be found frequently in hBeAg-positive subjects(77.8% and 88.9%), whereas T^(1653)/C^(1753) was moreprevalent in anti-hBe-positive subjects than inhBeAg-positive subjects (93.8% vs 33.3%).Conclusions: The whole frequency of mutations in pre-core and core promoter gene will become more fre-quent as hBV infection is to be persistent. Mutationto T^(1653)/C^(1753) may be useful as a marker for hepaticfailure. It requires further study whether the mixedinfection of mutants and wilds will develop and affectthe condition of hBeAg in serum along the progres-sion of liver disease.

This paper presents preliminary assessment of seedling survival and growth of green alder（Alnus viridis（Chaix） DC. in Lam. ＆ DC.） planted on fly ash disposal sites. This kind of post-industrial site is extremely hard to biologically stabilize without top-soiling. The experiment started with surface preparation using NPK start-up mineral fertilizer at 60–36–36 kg ha-1followed by initial stabilization through hydro-seeding with biosolids（sewage sludge 4 Mg ha-1dry mass） and a mixture of grasses（Dactylis glomerata L. and Lolium multiflorum Lam.）（200 kg ha-1）. Subsequently, three-years-old green alder seedlings were planted in plots on two substrate variants：the control（directly on combustion waste） and plots with3 dm3 lignite culm from a nearby mine introduced into the planting pit. Five years of preliminary monitoring show good survival seedling rates and growth parameters（height（h）, average increase in height（Dh）, number of shoots（L o）and leaf nitrogen supply in the fly ash disposal *** of the site with.a.combination of lignite culm in planting pits and preliminary surface preparation by hydroseeding and mineral fertilization had the most positive effect on green alder seedling parameters. The results indicate that it is possible and beneficial to use green alder for biological stabilization on fly ash disposal sites.

Objective: This study aimed to investigate DNA sequences that are substantially homologous to the corresponding RNA sequence sections of the hepatitis C virus (hCV). These DNA sequences are present in the whole DNA extracted from peripheral blood mononuclear cells (PBMCs) of hCV-negative subjects. We presumed that these experimentally proven 5'-noncoding region (5'-NCR) homologous DNA sequences could be contained in the extrachromosomal circular DNA (eccDNA) fraction as part of the whole cellular DNA. Methods: home-made polymerase chain reaction (PCR) with whole cellular and isolated eccDNA, nucleotide basic local alignment search tool (BLASTn) alignments, and tests for patterns of methylation in selected sequence sections were performed. Results: The PCR tests revealed DNA sequences of up to 320 bp that broadly matched the corresponding sequence sections of known hCV genotypes. In contrast, BLASTn alignment searches of published hCV 5'-NCR sequences with human genome databases revealed only sequence segments of up to 36 bp of the 5'-NCR. The composition of these sequences shows missing base pairs, base pair mismatches as well as complete homology with hCV reference sequences. These short sequence sections are present in numerous copies on both the same and different chromosomes. The selected sequence region within the DNA sequences of the 5'-NCR revealed a broad diversity of individual patterns of methylation. Conclusions: The experimental results confirm our assumption that parts of the hCV 5'-NCR genomic RNA sequences are present at the DNA level in the eccDNA fraction of PBMCs. The tests for methylation patterns therein revealed individual methylomes which could represent an epigenetic feature. The respective sequence section might be subject to genetic regulation.

188Re-tin sulfur colloid(TSC) was prepared to compare its biodistribution characteristics with 188Re-macroaggregates album(MAA) after transhepatic arterial embolization(TAE) in rabbits bearing VX2 liver *** efficiencies of the 188Re-TSC and 188Re-MAA were 99.94%±0.04% and 99.95%±0.03%,respectively,and they were stable for 72 h in human *** and biodistribution in 31 rabbits bearing VX2 liver tumor after transcatherter hepatic arterial injection of the radiopharmaceuticals were performed,and relevant activity accumulated mostly in the *** percentage of the injected dose per gram of wet tissue(%ID/g) of 188Re-TSC and 188Re-MAA were *** uptake of 188Re-TSC at 1 h.a.d 24 h were 24.32%±11.93% and 21.88%±18.29%,and the radioactive ratios of tumor/liver were 70.89±19.58 and 17.42±13.96,*** uptake of 188Re-MAA at 1 h.a.d 24 h were 38.78%±30.23% and 15.98%±26.64%,and the radioactive ratio of tumor/liver of 188Re-MAA were 39.71±25.06 and 8.13±4.61,respectively.188Re-TSC is a potential radiopharmaceutical for the therapy of tumors.

Preparation, Characterization of Superacid SO_4^(2-)/ZrO_2-SiO_2 and Its Activity on Catalytic Synthesis of Methyl p-Nitrobenzoate

Solid superacid SO42-/ZrO2-SiO2 was prepared by dip-impregnation method, and its catalytic activity was tested with esterification of p-nitrobenzoic acid and methanol. The optimum conditions were also found, that is, the molar ratio between silica and zirconia was 10：1, the calcination temperature was 550 ℃ and the soaked consistency of h2SO4 was 1.0 mol.L-1. Under the conditions that the ratio of methanol to aeid（mL/ g） was 12：1, the amount of catalyst was 0.5 g, the reaction time was 5 h.a.d the stirring speed was 800 ***-1, the yield of methyl p-nitrobenzoate could reach up to 90.5%. Then the characterizations of cataslyst, including the acidity and types of acidic centers, specific surface area and surface structure was respectively examined by hammer indicator, in-situ pyridine IR, BET method, FT-IR（KBr）, transmission electron microscope （TEM）, and X-ray diffraction （XRD）. The results showed that the catalyst SO42-/ZrO2-SiO2 was superacid with high specific surface area due to ho〈-12.70. It contained acidic sites of Lewis and BrФnsted, and its surface structure changed after esterification. The zirconia crystal was mainly tetragonal and silica crystal was not found.

Study Aim: Descriptions of the direct services of a psychologist (i.e. clinical psychology intern) in an accident and emergency department (AED) are provided as a new and rare area of professional practice. Method: Data were collected in a large, hong Kong, government hospital over an eighteen-month period. Recipients included 281 patients, 150 medical staff members, and 44 relatives of patients. Results: Interventions with patients and their family members centered on crisis management, diagnostic assessment, psychoeducation, and consultations regarding long-term adjustment to medical infirmities. The majority of staff members sought consultations regarding work related stress such.a. burnout symptoms and coping with vicarious trauma experiences. Frequency tables provide information on utilization of specific services while case vignettes contextualize psychological interventions. Conclusion: There are many potential benefits of psychological service for AED patients and staff.

The seeds of Aframomum melegueta K. Schum (grains of paradise, Zingiberaceae) are used as a common spice in African countries and a fine condiment in the European cuisine. In this study, we evaluated the phytochemical profile of various seed extracts of A. melegueta as well as their antimicrobial, antioxidant and enzyme inhibitory effects. In total, 25 diarylheptanoids, five gingerol derivatives and nine phenolic/organic acids were tentatively annotated in A. melegueta by liquid chromatography hyphenated with high resolution tandem mass spectrometry (LC-hRMS/MS). A. melegueta showed a moderate inhibitory activity against different human pathogenic microbial strains, with h. pylori the most sensitive microorganism. A strong antioxidant and enzyme inhibitory potential was shown in various radical scavenging, reducing and chelating assays as well as in cholinesterase, tyrosinase and glucosidase assays. Several specialized metabolites from A. melegueta (diarylheptanoids, gingerols) were shown to be directly linked with the investigated antioxidant and enzyme inhibitory activities, as evaluated through the correlation analysis. In addition, two diarylheptanoids (one heptan-3-ol and one heptan- 3,5-diol) displayed the strongest binding to acetylcholinesterase (AChE) via multiple h-bonds, a couple of π-π interactions and van der Waals interactions all over the catalytic channel of the enzyme, as evidenced by the molecular docking study. Overall, our work brings new contributions to the phyto-complexity and poly-pharmacology of spices from genus Aframomum than can find future applications in pharmaceutical, nutraceutical or cosmeceutical industry.
