SPDEF suppresses head and neck squamous cell carcinoma progression by transcriptionally activating NR4A1

SAM pointed domain containing E26 transformation-specific transcription factor(SPDEF)plays dual roles in the initiation and development of human ***,the biological role of SPDEF in head and neck squamous cell carcinoma(hNSCC)remains *** this study,the expression level of SPDEF and its correlation with the clinical parameters of patients with hNSCC were determined using TCGA-hNSC,GSE65858,and our own clinical ***8,colony formation,cell cycle analysis,and a xenograft tumor growth model were used to determine the molecular functions of SPDEF in ***-qPCR,dual luciferase reporter assay,and rescue experiments were conducted to explore the potential molecular mechanism of SPDEF in *** with normal epithelial tissues,SPDEF was significantly downregulated in hNSCC *** with hNSCC with low SPDEF mRNA levels exhibited poor clinical *** SPDEF inhibited hNSCC cell viability and colony formation and induced G0/G1 cell cycle arrest,while silencing SPDEF promoted cell proliferation in *** xenograft tumor growth model showed that tumors with SPDEF overexpression had slower growth rates,smaller volumes,and lower *** could directly bind to the promoter region of NR4A1 and promoted its transcription,inducing the suppression of AKT,MAPK,and NF-κB signaling ***,silencing NR4A1 blocked the suppressive effect of SPDEF in hNSCC ***,we demonstrate that SPDEF acts as a tumor suppressor by transcriptionally activating NR4A1 in *** findings provide novel insights into the molecular mechanism of SPDEF in tumorigenesis and a novel potential therapeutic target for hNSCC.

Complexes [MCl 2{Ch 3N(Ch 2Ch 2N[CDS1]CR 1R 2) 2}]((3, M=Co, R 1=h, R 2=Ph ; 4, M=Fe, R 1=h, \{R 2=Ph\}; 5, M=Co, R 1=R 2=Ph) were prepared and characterized by IR spectra and elemental analysis. The combination of each of complexes 3-5 with ethylaluminoxane(EAO), respectively, was found to be moderately active for ethylene oligomerization to low carbon olefins. The activity of 113 kg oligomers·mol -1 Co·h -1 for complex 3(100 mol of EAO, 180 ℃ and 1 8 MPa ethylene) was observed with a selectivity of 93% to C 4-10 olefins, of which 96% were linear C4 10 olefins. The catalytic properties of complexes 3-5 were compared with those of analogous P,P coordinated complexes [MCl 2{Ch 3N(Ch 2Ch 2PPh 2) 2}](1, M=Co; 2, \{M=\}Fe).

【目的】本研究旨在探索DNA甲基化是否通过调控细胞自噬进而影响家蚕Bombyx mori翅的发育。【方法】分别用1和2μg DNA甲基化特异性抑制剂5-aza-dC处理家蚕卵巢Bm12细胞和家蚕预蛹,荧光显微镜下观察Bm12细胞的数量,利用dot blot检测Bm12细胞中DNA甲基化水平,利用溶酶体染色检测细胞自噬强度;利用RT-qPCR检测细胞自噬相关蛋白(Agt)基因的表达水平;利用Western blot和免疫组化定性检测Bm12细胞中自噬标记物LC3蛋白的分型情况;观察成虫的翅表型并计算残翅率和翅面积。用2μg细胞自噬激活剂SMER28处理家蚕预蛹,以及采用自噬抑制剂Spautin-1(2μg)挽救5-aza-dC(2μg)处理后的家蚕预蛹,利用溶酶体染色检测翅细胞内自噬强度,观察翅表型并计算残翅率和翅面积。【结果】1μg 5-aza-dC处理后12,24和48 h抑制了Bm12细胞的生长,使Bm12细胞甲基化水平降低,Bm12细胞自噬水平升高;处理后48 h Bm12细胞中Atg基因表达上调。家蚕预蛹期注射2μg 5-aza-dC后24,48和72 h,成虫翅细胞内自噬水平升高,Atg基因的表达上调,出现大量畸形翅并且残翅率升高72.62%和翅表面积减少66%。家蚕预蛹期注射2μg SMER2824,48和72 h后,成虫翅细胞内自噬水平升高,成虫出现大量畸形翅,残翅率升高75.13%和翅表面积减少48.79%。利用Spautin-1对5-aza-dC处理的预蛹进行挽救实验,结果显示细胞自噬抑制可以缓解DNA去甲基化对成虫翅发育的影响。【结论】本研究结果证明DNA甲基化通过调节细胞自噬在家蚕翅发育中发挥作用。本研究结果为DNA甲基化调控昆虫发育提供了实验依据。

A novel porphyrin-Schiff base was synthesized via the condensation of 5-（4-aminophenyl） -10,15,20-triphenylperphyrin and ethyl vanillin. After analyzing the level of purification of the perphyrin-Schiff base, it was successfully separated. This perphyrin-Schiff base was characterized by using UV-Vis, IR,^1h NMR and MS spectroscopy.

The analytical electron microscopy has been used to characterize the morphology,structure and composition of the nanostructured material of Sn- Bi alloy prepared by a modified electrohydrodynamic technique. The electron diffraction pattern and the corresponding contrast image for the discrete particles with a diameter smaller than 4 nm have been *** is shown that the nanocrystalline Sn-Bi alloy particles comprise a single crystal of Bi-containing β-Sn solid solution or of Sn-containing Bi solid solution. A direct preparation procedure of the samples during the electrohydrodynamic rapid solidification process has been developed for electron microscopic observation.

Objective:To explore the characteristics of metabolic changes in patients with post-traumatic stress disorder through 1h-MRS in neuroanatomical circuit comparing with age-matches ***:Fifty patients with post-traumatic stress disorder and SO gender-and agematched normal controls were *** neurochemical abnormalities including the levels of choline(Cho)/ creatine(Cr) and N-acetylaspartate(NAA)/Cr were measured respectively in hippocampus and the anterior cingulate gyrus with three-dimension 1h-proton specrroscopy(3D 1h-MRS).Results:The values of NAA/Cr ratios in hippocampus and the anterior cingulate gyrus were significant lower in patients with post-traumatic stress disorder(1.71±0.32,left l.58±0.29, right 1.55±0.31) than that in controls(2.24±0.41,left 1.98±0.27,right 2.02±0.36)(Phe values of Cho/Cr in hippocampus(left 1.64±0.23,right 1.66±0.34) were no significant with that of controls(left 1.48±0.29,right 1.54±0.38).Values of Cho/Cr in cingulate gyrus were significant higher in post-traumatic stress disorder patients(I.88±0.44) than that in controls(1.37.±0.32) (Phe results indicate some special neurochemical and histological structure changes in post-traumatic stress disorder patients,which might occurre earlier in anterior cingulate gyrusthe than in hippocampus.

Collective cell migration is extensively observed in embryo development and cancer *** these processes,the interactions between cells with distinct identities and fates are of importance for boundary formation and host defense against *** this paper,we explore the collective dynamics of a two-dimensional cell mixing monolayer consisting of non-tumorigenic mammary epithelial cells and breast cancer *** show that the epithelial-cancerous cell mixing system displays unique sorting *** epithelial cells aggregate to form scattered clusters,which perform random motion with simultaneous translation and rotation,strikingly distinct from the classical persistent random walk of individual migratory *** motility of cancer cells is markedly promoted by the epithelial clusters,exhibiting remarkable contact enhancement of locomotion.A discrete model based on the Johnson-Kendall-Roberts contact mechanics is proposed to identify the influence of intercellular interactions,active migration forces and cell-substrate friction forces on the collective cell *** findings could advance our understanding of many biological processes,such as cancer metastasis and tissue morphogenesis.

Spontaneous electrochemical noise (EN) can be a rich source of information concerning the processes simultaneously occurring on a corroding interface. But the noise signal is often difficult to be analyzed due to the complicated nature of the specific systems being investigated. In this paper, the potential noise fluctuations during the free corrosion of commercial aluminum alloy LY12 in sodium chloride solution was recorded and analyzed with different techniques. The typical results showed that the fractal dimension (D,n) obtained from spectral power density (SPD) is mainly directly proportional to the intensity of pitting corrosion and to the value of pitting parameter (SE) derived from dimensional analysis, while the fractal dimension (DE) obtained from EIS is mainly related to the uniform corrosion.
