Rbfox1 controls alternative splicing of focal adhesion genes in cardiac muscle cells

Alternative splicing is one of the major cellular processes that determine the tissue-specific expression of protein ***,it remains challenging to identify physiologically relevant and tissue-selective proteins that are generated by alternative ***,we investigated the target spectrum of the splicing factor Rbfox1 in the cardiac muscle context in more *** using a combination of in silico target prediction and in-cell validation,we identified several focal adhesion proteins as alternative splicing targets of *** focused on the alternative splicing patterns of vinculin(metavinculin isoform)and paxillin(extended paxillin isoform)and identified both as potential Rbfox1 *** analyses suggested that both isoforms are promoted by Rbfox1 due to binding in the *** adhesions play an important role in the cardiac muscle context,since they mainly influence cell shape,cytoskeletal organization,and cell–matrix *** data confirmed that depletion of Rbfox1 changed cardiomyoblast morphology,cytoskeletal organization,and multinuclearity after differentiation,which might be due to changes in alternative splicing of focal adhesion ***,our results indicate that Rbfox1 promotes alternative splicing of focal adhesion genes in cardiac muscle cells,which might contribute to heart disease progression,where downregulation of Rbfox1 is frequently observed.

Kaapvaal lamproites(aka orangeites)are a group of volatile-rich(h_(2)O,CO_(2)),micaceous,ultrapotassic igneous rocks that are unique to the Kaapvaal craton in southern ***,the composition of the melts that give rise to these rocks remains poorly understood due to overprinting effects of contamination by mantle and crustal material,volatile exsolution,fractional crystallisation and post-magmatic ***,this lack of reliable data on the initial composition of the Kaapvaal lamproite melts hampers our understanding of their source,petrogenesis and ascent *** is a common mineral comprising the Kaapvaal lamproites that has both xenocrystic(i.e.,mantle)and magmatic *** inclusions(melt/fluid)entrapped within olivine have been consistently demonstrated as an effective tool for gaining fundamental insights into the composition and evolution of melts that produce both kimberlites and lamproites,prior to processes,such as eruption,devolatization and syn-/post-magmatic *** this study,multiphase inclusions of both secondary and pseudosecondary origin hosted in olivine from a Kaapvaal lamproite(silvery home,south Africa)provide novel insights into the composition of the melt(s)that initially transported olivine to the surface and then crystallised after emplacement to form the lamproite *** inclusions in our study contain daughter mineral assemblages consisting of diverse Ca-Mg carbonates,including K-,Na-,Ba-,and sr-bearing varieties,moderate K-rich silicates(phlogopite,tetraferriphlogopite),and subordinate oxides,phosphates,sulphides,sulphates,and *** on these daughter mineral assemblages,we suggest that the composition of the melt entrapped by olivine was siO_(2)-poor,Ca-Mg carbonate-rich and contained elevated concentrations of K,Na,Ba,sr,P and *** mineral and reconstructed melt compositions are in stark contrast to the mineral association of the groundmass and the bulk-rock composition

Methane dominated gas is one of the cleanest energy resources;however, there is no direct method to determine its source rock. Natural gases produced from the eastern sichuan Basin together with seismic data were studied for their sources and secondary alteration by thermochemical sulfate reduction(TsR). Our results demonstrate that Upper Permian to Lower Triassic(P_(3)ch-T_(1)f) gases in the surrounding of the Kaijiang-Liangping area show volatile organic sulfur compounds(VOsCs) δ^(34)s values close to those of the associated h_(2)s, and may have been altered by methane-dominated TsR, resulting in positive shift in methane δ^(13)C_(1)values with increasing TsR extents. Other(or group 2) gases produced from the P_(3)ch-T_(1)f reservoirs from the southern area and the Upper Carboniferous to Middle Permian(C_(2)h-P_(2)q) from the eastern sichuan Basin are not significantly changed by TsR, show similar δ^(34)s values between the kerogens and some VOsCs, and may have been derived from the Lower silurian and Middle Permian source rocks. This study demonstrates a case for the first time showing the δ^(34)s values of VOsCs can be used as a tool for direct correlation between non-TsR altered gas and source rocks. Methane-dominated gas pools can be found using gas and source rock geochemistry combined with seismic data.

Accelerating lithium-sulfur battery reaction kinetics and inducing 3D deposition of Li_(2)s using interactions between Fe_(3)se_(4)and lithium polysulfides

Although lithium-sulfur batteries(LsBs)exhibit high theoretical energy density,their practical application is hindered by poor conductivity of the sulfur cathode,the shuttle effect,and the irreversible deposition of Li_(2)*** address these issues,a novel composite,using electrospinning technology,consisting of Fe_(3)se_(4)and porous nitrogen-doped carbon nanofibers was designed for the interlayer of *** porous carbon nanofiber structure facilitates the transport of ions and electrons,while the Fe_(3)se_(4)material adsorbs lithium polysulfides(LiPss)and accelerates its catalytic conversion ***,the Fe_(3)se_(4)material interacts with soluble LiPss to generate a new polysulfide intermediate,Li_(x)Fes_(y)complex,which changes the electrochemical reaction pathway and facilitates the three-dimensional deposition of Li_(2)s,enhancing the reversibility of *** designed LsB demonstrates a high specific capacity of1529.6 mA h g^(-1)in the first cycle at 0.2 *** rate performance is also excellent,maintaining an ultra-high specific capacity of 779.7 mA h g^(-1)at a high rate of 8 *** investigation explores the mechanism of the interaction between the interlayer and LiPss,and provides a new strategy to regulate the reaction kinetics and Li_(2)s deposition in LsBs.

BACKGROUND Type 2 diabetes is one of the most prevalent chronic diseases worldwide,significantly impacting patients'quality of *** treatment options like metformin(MET)effectively counteract hyperglycemia but fail to alleviate diabetes-associated complications such as retinopathy,neuropathy,nephropathy,hepatopathy,and cardiovascular *** To propose the supplementation of cholecalciferol(ChO)and taurine(TAU)to enhance MET efficacy in controlling diabetes while minimizing the risk of associated *** The study involved sixty rats,including ten non-diabetic control rats and fifty experimental rats with type 2 diabetes induced by *** experimental rats were further subdivided into positive control and treatment *** four treatment groups were randomly allocated to a single MET treatment or MET combined with supplements either ChO,TAU,or *** Diabetic rats exhibited elevated levels of glucose,insulin,homeostasis Model Assessment of Insulin Resistance(hOMA-IR),glycated hemoglobin percentage,lipid markers,aspartate aminotransferase,and malondialdehyde,along with reduced levels of antioxidant enzymes(catalase and superoxide dismutase).The administration of ChO and TAU supplements alongside MET in diabetic rats led to a noticeable recovery of islet *** antioxidative,anti-inflammatory,and anti-apoptotic properties of the proposed combination therapy significantly ameliorated the aforementioned *** The supplementation of ChO and TAU with MET showed the potential to significantly improve metabolic parameters and protect against diabetic complications through its antioxidative,anti-inflammatory,and antiapoptotic effects.

Chalcogen-dependent catalytic properties of RuX2 (X = s/se/Te) nanoparticles decorated carbon nanofibers for hydrogen evolution in acidic and alkaline media

Transition metal dichalcogenides(TMDs),with the general formula MX_(2)(M=Mo/W/Fe/Co/Ni,etc.;X=s/se/Te),have attracted extensive research interests for hydrogen evolution reaction(heR).Compared with numerous studies on noble-metal-free TMDs,the chalcogen-dependent heR catalytic properties of noble-metal-based TMDs are lack of sufficient research ***,a facile electrospinning-assisted synthetic strategy is proposed to synthesize ruthenium dichalcogenides(RuX_(2),X=s/se/Te)nanoparticles decorated carbon nanofibers(CNFs).Benefiting from the identical nanofibrous morphology and exposed crystal planes of RuX_(2)(111),the catalytic activities of RuX_(2)@CNFs samples were investigated and compared in a fair and direct *** electrochemical measurements coupled with density functional theory calculations were carried out to probe their intrinsic heR catalytic activities,resulting in the catalytic activity order of Rus_(2)@CNFs>Ruse_(2)@CNFs>RuTe_(2)@CNFs in acidic media and that of Rus_(2)@CNFs>RuTe_(2)@CNFs>Ruse_(2)@CNFs in alkaline *** superior catalytic performance of Rus_(2)@CNFs mainly stems from the relative lower heR energy barriers and thereby the higher intrinsic catalytic activity of Rus_(2)(111),leading to ultralow overpotentials of 44 and 9 mV at 10 mA·cm^(-2) in acidic and alkaline media,***_(2)(111)is endowed with the more optimized Gibbs free energy of hydrogen adsorption(ΔGh*)than RuTe_(2)(111),but RuTe_(2)(111)shows enhanced catalytic property for h_(2)O dissociation and Oh-desorption than Ruse_(2)(111),therefore,resulting in the altered catalytic activity sequences for Ruse_(2) and RuTe_(2) in acidic and alkaline media.
