Optimizing ABA-based chemically induced proximity for enhanced intracellular transcriptional activation and modification response to ABA

Abscisic acid(ABA)-based chemically induced proximity(CIP) is primarily mediated by the interaction of the ABA receptor pyrabactin resistance 1-like 1(PYL1) and the 2C-type protein phosphatase ABI1, which confers ABA-induced proximity to their fusion proteins, and offers precise temporal control of a wide array of biological processes. however, broad application of ABA-based CIP has been limited by ABA response intensity. In this study, we demonstrated that ABA-induced interaction between another ABA receptor pyrabactin resistance 1(PYR1) and ABI1 exhibited higher ABA response intensity than that between PYL1 and ABI1 in hEK293t cells. we engineered PYR1-ABI1and PYL1-ABI1 into ABA-induced transcriptional activation tools in mammalian cells by integration with CRISPR/dCas9 and found that the tool based on PYR1-ABI1 demonstrated better ABA response intensity than that based on PYL1-ABI1 for both exogenous and endogenous genes in mammalian cells. we further achieved ABA-induced RNA m6A modification installation and erasure by combining ABA-induced PYR1-ABI1 interaction with CRISPR/dCas13, successfully inhibiting tumor cell proliferation. we subsequently improved the interaction of PYR1-ABI1 through phage-assisted continuous evolution(PACE), successfully generating a PYR1 mutant(PYR1m) whose interaction with ABI1 exhibited a higher ABA response intensity than that of the wild-type. In addition, we tested the transcriptional activation tool based on PYRm-ABI1 and found that it also showed a higher ABA response intensity than that of the wild type. these results demonstrate that we have developed a novel ABA-based CIP and further improved upon it using PACE, providing a new approach for the modification of other CIP systems.

the ground-state mass excess of the t_(z)=−2 drip-line nucleus ^(22)Al is measured for the first time as 18103(10)keV using the newly-developed Bρ-defined isochronous mass spectrometry method at the cooler storage ring in *** new mass excess value allowed us to determine the excitation energies of the two low-lying 1+states in ^(22)Al with significantly reduced uncertainties of 51 *** compared to the analogue states in its mirror nucleus ^(22)F,the mirror energy differences of the two 1^(+)states in the ^(22)Al-^(22)F mirror pair are determined to be−625(51)keV and−330(51)*** excitation energies and mirror energy differences are used to test the state-of-the-art ab initio valence-space in-medium similarity renormalization group calculations with four sets of interactions derived from the chiral effective field *** mechanism leading to the large mirror energy differences is investigated and attributed to the occupation of theπs_(1/2) orbital.

目的探讨不同剂量艾司氯胺酮复合罗哌卡因臂丛上干阻滞对肩关节镜术后反弹性疼痛的影响。方法选择行肩关节镜下治疗的患者90例,男35例,女55例,年龄18~60岁,BmI 18.5~28.0 kg/m^(2),ASAⅠ或Ⅱ级,按照随机数字表法将患者分为三组:E1组、E2组和R组,每组30例。三组患者神经阻滞局麻药物分别按照以下方案配置,E1组予0.2%罗哌卡因、艾司氯胺酮0.25 mg/kg和生理盐水共10 ml;E2组予0.2%罗哌卡因、艾司氯胺酮0.5 mg/kg和生理盐水共10 ml;R组予0.2%罗哌卡因和生理盐水共10 ml。所有患者在神经阻滞麻醉起效后行全身麻醉。记录神经阻滞前(t_(1))、阻滞后30 min(t_(2))、置入喉罩(t_(3))、术中30 min(t_(4))时hR、mAP、手术体积描计指数(SPI)。记录术后6、8、12、24、48 h时静息和活动时NRS评分、反弹性疼痛的发生情况。记录神经阻滞麻醉后感觉和运动阻滞起效时间以及持续时间、阻滞前后镇静评分、术后24 h内睡眠情况、术后48 h内不良反应的发生情况。结果三组不同时点hR、mAP差异均无统计学意义。与R组比较,E1、E2组舒芬太尼用量明显减少,术后12~48 h静息和活动时NRS评分明显降低(Pd睡眠时间明显延长,睡眠效率明显升高(Ph内恶心呕吐、头晕头痛等术后不良反应发生率明显降低(Pmg/kg复合罗哌卡因能降低臂丛上干神经阻滞肩关节镜术后反弹性疼痛的发生率,改善患者术后睡眠质量,但不能缩短臂丛上干神经阻滞的感觉阻滞起效时间和增加阻滞持续时间。

稠油井内衬保温油管热损失分析及开采参数优化

针对塔河油田稠油开采过程中原油热量损失大、资源浪费的问题,开展内衬保温油管开采技术研究。基于井筒稳态热传递与地层非稳态热传递理论,建立了稠油井井筒热传递数学模型,评价了不同内衬材料油管的保温性能,揭示了含水率、日产液量及内衬保温油管下深对井筒温度的影响规律。研究表明:内衬聚酮防腐层与气凝胶保温层的保温油管保温性能最佳,日产液量和保温油管下深对井口温度影响较大,含水率对井口温度的影响较小;日产液量大于72 t/d且保温油管下深大于3 500 m时,可满足稠油开采的温度要求。该研究明确了内衬保温油管保温的可行性,可为油田保温油管内衬的选材、稠油开采工艺参数的制订提供技术依据。

流星进入地球大气层时会发生空中解体和爆炸,威胁地球局部区域乃至全球安全针对流星进入大气演化的超高速动力学仿真研究具有重要意义.为提高预报的精确性,应关注流星体内部结构和非均匀烧蚀等对演化过程和解体后碎片运动的影响。本文提出了一种可变形不规则离散元方法,用于仿真分析流星进入大气层时轨迹和姿态变化、烧蚀、解体和爆轰等现象。利用多面体离散元模型可模拟不同结构的异质流星;通过拉伸多面体模型顶点,仿真流星的粗糙外形和烧蚀形变;利用Jones-wilkins-Lee状态方程计算烧蚀产生的集中爆轰产物的压力.以Chelyabinsk流星事件中发生轨迹偏离的碎片为例,数值仿真集中爆轰产物的膨胀驱动作用对碎片运动方向和轨迹演化的影响.仿真结果表明,碎片弹出轨迹特征和烧蚀剩余质量均在观测值范围内,验证了新方法的有效性.

Search for production of dark fermion candidates in association with heavy neutral gauge boson decaying to dimuon in proton-proton collisions at√s=8 teV using CmS open data

In this study,we conducted a search for dark matter using a part of the data recorded by the CmS experiment during run-I of the LhC in 2012 with a center of mass energy of 8 teV and an integrated luminosity of 11.6 fb−*** data were gathered from the CmS open *** matter,in the framework of the simplified model(mono-Z′),can be produced from proton-proton collisions in association with a new hypothetical gauge boson,Z′.thus,the search was conducted in the dimuon plus large missing transverse momentum *** benchmark scenario of mono-Z′,which is known as light vector,was used for interpreting the CmS open *** evidence of dark matter was observed,and exclusion limits were set on the masses of dark matter and Z′at 95%confidence level.

近20年季风爆发前后珠江三角洲前汛期短时强降水的时空演变特征与成因

相较于暴雨这种日尺度强降水,短时强降水(≥20 mm h^(−1))是造成山洪滑坡与城市内涝等灾害更为直接的因素。本文利用地面气象观测站和ERA5再分析数据,重点研究南海季风爆发前后珠江三角洲地区(简称珠三角)短时强降水的时空演变特征,并探索了短时强降水在季风爆发前后特征差异的可能成因。研究表明:(1)相较于季风爆发前,珠三角地区季风爆发后的降水明显增多,其中短时强降水贡献的比例显著增加。对短时强降水本身而言,区域平均强度以及极端性在季风爆发前后差异总体较小,但短时强降水频率在季风爆发后增加70%。(2)短时强降水高发区主要集中在珠三角东北部和珠江口西侧沿海,季风爆发后上述两个地区的频次增多最明显。短时强降水频率由季风爆发前的单峰型(下午)转为季风爆发后的双峰型(早晨与下午)。(3)短时强降水具有明显的区域性变化特征,短时强降水在季风爆发后的平均雨强和极端性在珠江口西侧沿海较内陆地区明显增强,其频次峰值时间在沿海地区从季风爆发前的午后转为季风爆发后的早晨,内陆地区在季风爆发前后均集中在下午。(4)季风爆发后,短时强降水期间的低层环境水汽超过同期气候态水平的16%。充沛的水汽在夜间在季风加速作用下被输送至沿海,并与陆风作用增强了辐合,这解释了沿海短时强降水的在季风爆发前后频次峰值时间转换现象。(5)相较于季风爆发前,季风爆发后珠三角短时强降水频率与低层水汽通量的相关性明显升高。珠三角沿海地区夜间—早晨短时强降水的增多与中低层风场结构改变造成的动力强迫有关。内陆地区季风爆发前后短时强降水与环境热力和不稳定条件关系更大。这些结果有助于我们更好地了解珠三角地区在季风爆发前后短时强降水的时空分布特征和理解其产生机制。

we report the detection of aγ-ray bubble spanning at least 100deg2 in ultra-high energy(UhE)up to a few PeV in the direction of the star-forming region Cygnus x,implying the presence super PeVatron(s)accelerating protons to at least 10 PeV.A log-parabola form with the photon indexΓ(E)=(2.71±0.02)+(0.11±0.02)×log10(E/10 teV)is found fitting the gamma-ray energy spectrum of the bubble *** sources,“hot spots”correlated with very massive molecular clouds,and a quasi-spherical amorphousγ-ray emitter with a sharp central brightening are observed in the *** the core of~0.5°,spatially associating with a region containing massive OB association(Cygnus OB2)and a microquasar(Cygnus x-3),as well as previously reported multi-teV sources,an enhanced concentration of UhEγ-rays is observed with 2 photons at energies above 1 *** general feature of the bubble,the morphology,and the energy spectrum,are reasonably reproduced by the assumption of a particle accelerator in the core,continuously injecting protons into the ambient medium.

目的以moS_(2)/ti_(3)C_(2)t_(x)为固体润滑剂,在25(室温)~400℃下制备具有优异摩擦学性能的moS_(2)/ti_(3)C_(2)t_(x)磷酸盐涂层,并研究它在不同温度下的减摩抗磨机制。方法以氢氟酸为mAx相(ti_(3)AlC_(2))粉体的蚀刻剂,制备具有“手风琴”形貌的ti3C2txmxene。以硫脲、钼酸铵、mxene为原料,制备moS_(2)/ti_(3)C_(2)t_(x)复合材料。以Al(h2PO4)3为黏结剂,以Cu O为固化剂,分别以ti_(3)C_(2)t_(x)和moS_(2)/ti_(3)C_(2)t_(x)为固体润滑剂,制备ti3C2tx磷酸盐涂层和moS_(2)/ti_(3)C_(2)t_(x)磷酸盐涂层。通过高温摩擦磨损试验机和光学数码显微镜测试涂层在25~400℃时的摩擦因数和磨损率,采用扫描电子显微镜和显微共焦激光拉曼光谱仪分析磨痕表面形貌、物相,进而探讨磨损机理。结果当ti_(3)C_(2)t_(x)与Al(h_(2)PO_(4))_(3)的质量比为2∶1时,ti3C2tx磷酸盐涂层在室温(25℃)下的摩擦因数和磨损率均最低,分别为0.38和2.75×10-4mm3/(N·m)。在ti_(3)C_(2)t_(x)表面负载moS2,将moS_(2)/ti_(3)C_(2)t_(x)作为固体润滑剂,能够显著降低磷酸盐涂层在25~400℃下的摩擦因数,同时磨损率也有所降低。moS_(2)/ti_(3)C_(2)t_(x)磷酸盐涂层在室温下的摩擦因数低至0.11,相较于ti3C2tx磷酸盐涂层降低了71.1%,其磨损率相较于ti3C2tx磷酸盐涂层降低了1个数量级。在25~400℃范围内,moS_(2)/ti_(3)C_(2)t_(x)磷酸盐涂层的摩擦因数均低于0.21。随着温度的升高,摩擦因数呈先减小后增大的趋势,磨损率整体上呈增大趋势。结论相较于ti_(3)C_(2)t_(x)磷酸盐涂层,moS_(2)/ti_(3)C_(2)t_(x)磷酸盐涂层在25~400℃下的摩擦学性能均得到显著提升。
