Cluster analysis reveals a homogeneous subgroup of PCOS women with metabolic disturbance associated with adverse reproductive outcomes

Background:Polycystic ovarian syndrome(PCOS)is a heterogeneous and complex reproductive endocrinological disease that could lead to *** were many attempts to classify PCOS but it remains unclear whether there is a specific subgroup of PCOS that is associated with the best or worst reproductive outcomes of assisted reproductive techniques(ARt).methods:Infertile PCOS patients who underwent their first cycle of in vitro fertilization(IVF)in west China Second University hospital,Sichuan University from January 2019 to december 2021 were *** clinical and laboratory information of each individual were *** cluster analysis was *** ovarian stimulation parameters and reproductive outcomes were collected and compared between the different clusters of ***:Our analysis clustered women with PCOS into"reproductive","metabolic",and"balanced"clusters based on nine *** group was characterized by high levels of testosterone(t),sex hormone-binding globulin(ShBG),follicular stimulation hormone(FSh),luteinizing hormone(Lh),and anti-müllerian hormone(Amh).metabolic group was characterized by high levels of body mass index(BmI),fasting insulin,and fasting *** group was characterized by low levels of the aforementioned reproductive and metabolic parameters,except for *** with PCOS patients in reproductive and balanced clusters,those in metabolic cluster had lower rates of good quality day 3 embryo and blastocyst ***,PCOS patients in the reproductive cluster had greater fresh embryo transfer(Et)cancelation rate and clinical pregnancy rate after fresh Et than metabolic cluster(odds ratio[OR]=3.37,95%confidence interval[CI]:1.77-6.44,and OR=6.19,95%CI:1.58-24.24,respectively).And compared with PCOS of metabolic cluster,PCOS of balanced cluster also had higher chance for fresh Et cancelation(OR=2.83,95%CI:1.26-6.35).Conclusion:Our study suggested that PCOS patients in met

Crystalline carbon nitride with in-plane built-in electric field accelerates carrier separation for excellent photocatalytic hydrogen evolution

Achieving a high carrier migration efficiency by constructing built-in electric field is one of the promising approaches for promoting photocatalytic activity. herein, we have designed a donor-acceptor(d-A) crystalline carbon nitride(APmCN) with 4-amino-2,6-dihydroxypyrimidine(AP) as electron donor, in which the pyrimidine ring was well embedded in the heptazine ring via hydrogen-bonding effect during hydrothermal process. the APmCN shows superior charge-transfer due to giant built-in electric field(5.94times higher than pristine carbon nitride), thereby exhibiting excellent photocatalytic h_(2) evolution rate(1350 μmol/h) with a high AQY(62.8%) at 400 nm. mechanistic analysis based on detailed experimental investigation together with theoretical analysis reveals that the excellent photocatalytic activity is attributed to the promoted charge separation by the giant internal electric field originated from the d–A structure.

Zeolitic Imidazolate Framework-8(ZIF-8)material was prepared by chemical precipitation *** microstructure and physical properties of the as-prepared samples were characterized by xRd,BEt,FESEm and UV *** self-made four-channel measurement device was used to test the gas sensitivity of ZIF-8 material toward ethanol gas under photo-thermal synergistic *** results showed that the sample was typical ZIF-8(E_(g)=4.96 eV)with a regular dodecahedron shape and the specific surface is up to 1793 m^(2)/*** as-prepared ZIF-8 has a gas response value of 55.04 to 100 ppm ethanol at 75℃ and it shows good gas sensing selectivity and repeated *** excellent gas sensitivity can be attributed to the increase of free electron concentration in the ZIF-8 conduction band by photo-thermal synergistic excitation,and the large specific surface area of ZIF-8 material provides more active sites for gas-solid surface *** reaction mechanism of ZIF-8 material under multi-field excitation was also discussed.

Autosomal recessive spinocerebellar ataxias（SCARs） are one of the most common neurodegenerative diseases characterized by progressive ataxia. Although SCARs are known to be caused by mutations in multiple genes, there are still many cases that go undiagnosed or are misdiagnosed. In this study, we presented a SCAR patient, and identified a probable novel pathogenic mutation（c.1A>G, p.m1V） in the AFG3L2 start codon. the proband's genotype included heterozygous mutations of the compound AFG3L2(p.[m1V]; [R632x]（c.[1A>G];[1894.C>t]）), which were inherited from the father（c.1A>G, p.m1V） and mother（c.1894C>t, p.R632x）. Functional studies performed on hiPSCs（human induced pluripotent stem cells） generated from the patients and hEK293t cells showed that the mutations impair mitochondrial function and the unbalanced expression of AFG3L2 mRNA and protein levels. Furthermore, this novel mutation resulted in the degradation of the protein and the reduction of the stability of the AFG3L2 protein, and mCU（mitochondrial calcium uniporter） complex mediated Ca2+overload.

Au-Fe_(3)O_(4) dumbbell-like nanoparticles based lateral flow immunoassay for colorimetric and photothermal dual-mode detection of SARS-CoV-2 spike protein

Lateral flow immunoassay(LFIA)has become popular in laboratories,at-home testing,and medical diagnostics due to its minimal cost and ***,conventional test strips based on colloidal gold can only obtain qualitative or semi-quantitative results with low *** this work,AuFe_(3)O_(4) dumbbell-like nanoparticles were synthesized and used as the LFIA labelling marker for highly sensitive colorimetric-photothermal dual-mode detection of SARS-CoV-2 spike(S)*** unique dumbbell structure of Au-Fe_(3)O_(4) NPs makes it possible to combine the best features of both Au NPs and Fe_(3)O_(4) *** increased surface area of these NPs enhances their LSPR effect and photothermal effect,which achieves signal amplification to increase *** Au-Fe_(3)O_(4) NPs modified with S protein antibody could identify S protein in samples,which were recognized and accumulated on t-line by another antibody,generating color band for qualitative colorimetric *** t-line was irradiated by laser to obtain temperature change for quantitative detection of *** optimized conditions,the detection limit was 1.22 pg/m L,three orders of magnitude more sensitive than colorimetric ***,the approach was performed on SARS-CoV-2 pseudovirus samples and outperformed traditional colloidal gold *** LFIA platform exhibits significant promise for practical implementation,as it can satisfy the need for low-cost,high-sensitivity,and home-based quantitative detection for respiratory infectious diseases.

Chronic obstructive pulmonary disease(COPd)is characterised by persistent airway inflammation even after cigarette smoking *** extracellular traps(NEts)have been implicated in COPd severity and acute airway inflammation induced by short-term cigarette smoke(CS).however,whether and how NEts contribute to sustained airway inflammation in COPd remain *** study aimed to elucidate the immunoregulatory mechanism of NEts in COPd,employing human neutrophils,airway epithelial cells(AECs),dendritic cells(dCs),and a long-term CS-induced COPd mouse model,alongside cyclic guanosine monophosphate-adenosine monophosphate synthase and toll-like receptor 9 knockout mice(cGAS-^(−/−)tLR9^(−/−));Additionally,bronchoalveolar lavage fluid(BALF)of COPd patients was *** from COPd patients released greater cigarette smoke extract(CSE)-induced NEts(CSE-NEts)due to mitochondrial respiratory chain *** CSE-NEts,containing oxidatively-damaged dNA(NEts-dNA),promoted AECs proliferation,nuclear factor kappa B(NF-κB)activation,NF-κB-dependent cytokines and type-I interferons production,and dC maturation,which were ameliorated/reversed by silencing/inhibition of cGAS/*** the COPd mouse model,blocking NEts-dNA-sensing via cGAS^(−/−)and tLR9^(−/−)mice,inhibiting NEtosis using mitotEmPO,and degrading NEts-dNA with dNase-I,respectively,reduced NEts infiltrations,airway inflammation,NF-κB activation and NF-κBdependent cytokines,but not type-I interferons due to IFN-α/βreceptor *** NEts components(myeloperoxidase and neutrophil elastase activity)in BALF of COPd smokers correlated with disease severity and NF-κB-dependent cytokine levels,but not type-I interferon *** conclusion,NEts-dNA promotes NF-κB-dependent autoimmunity via cGAS/tLR9 in long-term CS exposure-induced ***,targeting NEts-dNA and cGAS/tLR9 emerges as a potential strategy to alleviate persistent airway inflammation in COPd.
