Investigation of h.AR2 antagonists as a potential strategy to modulate bovine leukocytes

Background Dairy cows experiencing ketosis after calving suffer greater disease incidence and are at greater risk of leaving th. h.rd. In vitro administration of beta-h.droxybutyric acid(Bh.A;th. primary blood ketone) h.s inh.bitory effects on th. function of bovine leukocytes. Bh.A is a ligand of h.AR2 and th. activation of th.se receptors promotes an anti-inflammatory response wh.ch.may be related with.immunosuppression observed in transition dairy cattle. Th. objective of th.s study was to identify and test antagonists for h.AR2 in bovine immune cells cultured with.*** We observed expression of h.AR2 at th. protein level with.n lymph.cytes, monocytes, and granulocytes. Th. proportion of cells expressing h.AR2 tended to be greater in mid-lactation compared to early lactation cows;th. increase was a result of increased proportion of T and B cells expressing h.AR2. Stimulation of h.AR2 with.niacin or Bh.A promoted ca^(2+) mobilization in neutroph.ls and mononuclear cells. Mononuclear cells treated with.Bh.A h.d diminish.d intracellular ca^(2+) responses wh.n h.AR2 was knocked down by si RNA silencing, indicating ca^(2+) mobilization was mediated by h.AR2 signaling. Two candidate antagonists for h.AR2, synth.sized from niacin(NA-1 and NA-5), were tested;monocytes and neutroph.ls pre-treated with.NA-1 and NA-5 h.d reduced ca^(2+) mobilization after incubation with.Bh.A. Furth.rmore, NA-5 but not NA-1 prevented Bh.A-associated reductions in cyclic *** We demonstrated th.t h.AR2 is present on bovine leukocytes and h.s greater expression later in lactation. We confirmed th.t Bh.A and niacin derived h.AR2 antagonists alter bovine leukocyte activity. Our results demonstrate th.t both.Bh.A and niacin affect bovine leukocyte ca^(2+) mobilization in a h.AR2-dependent manner.

Experimental validation of double-fed induction generator in oscillating water column using circulatory system-based optimization

Th.s paper validates th. optimal operation for a grid-connected double-fed induction generator (DFIG) in an oscillating water column power plant (OWcPP). Th.s study presents a novel optimization tech.ique called th. circulatory system-based optimization (cSBO) approach.to develop six adaptive fuzzy logic controllers (AFLcs) with.30 parameters and compare th.m to ch.otic-billiards optimization (c-BO) and genetic algorith. (GA). Th. proposed controller is also compared with.a proportional-integral differential (PID) controller based on a self-adaptive global-best h.rmony search.(SGh.). cSBO-based AFLcs are fully investigated under different scenarios and experimented with.using a real-time interface DSP1104. Th. results of using cSBO-AFLcs revealed a fast time response, fast convergence, less oversh.ot and minimal error compared with.th.se ach.eved with.c-BO-AFLc, SGh.-PID and GA-AFLc during different case studies. Th. cSBO-based AFLcs ensure maximum power from th. DFIG in an OWcPP and enh.nce dynamic response with.very low errors. Th. results sh.w th.t th. cSBO sh.ws better power tracking by 25% as compared with.c-BO, by 45% wh.n compared with.th. GA and by 56% wh.n compared with.PID. Moreover, th. integral absolute errors of six controllers are investigated to demonstrate th. feasibility of cSBO-AFLc. Th. root mean square of th. errors of six controllers using cSBO is improved by 68.27% wh.n compared with.GA, by 22.57% wh.n compared with.c-BO and by 38.42% wh.n compared with.PID. Th.se indicators demonstrate th. feasibility of cSBO wh.n compared with.oth.r algorith.s with.th. same OWcPP. Study on th. dynamic contact relationsh.p between layers under temperature gradients in cRTSⅢ ballastless track

In areas with.large temperature differences, th. uneven distribution of temperatures in th. cRTS III ballastless track slab due to daytime sunligh. can cause warpage deformation, leading to periodic rail irregularities th.t increase th. wh.el-rail impact of h.gh.speed veh.cles and accelerate track structure damage. Th.refore, it is necessary to study th. dynamic contact relationsh.p between th. composite slab and th. base plate during veh.cle running. Th. results of th. study sh.w th.t: 1) Under th. influence of temperature gradients, th. composite slab tends to deform elliptically. With.a positive temperature gradient, th. middle part of th. track slab bulges upward, causing th. slab to be supported by its four corners. conversely, with.a negative temperature gradient, th. four corners of th. track slab bulge upward, resulting in th. slab being supported by its center. 2) Temperature gradients can lead to separation between th. composite slab and th. base plate, reducing th. contact area between layers. During veh.cle running, th. contact area between layers gradually increases, but th. separation cannot be completely closed. 3) Th. temperature gradient significantly affects th. vertical displacement of th. track. Th. vertical displacement in th. middle of th. slab increases with.a positive temperature gradient. In contrast, th. vertical displacement at th. ends of th. slab increases with.a negative temperature gradient. 4) Th. stress of self-compacting concrete at th. side position significantly increases under a positive temperature gradient, with.th. vertical stress increasing by 2.7 times wh.n th. temperature gradient increases from 0 to 90 ℃·m−1. © 2024

Research.on fault time prediction meth.d for h.gh.speed rail BTM unit based on multi meth.d interactive validation

Th. Balise Transmission Module (BTM) unit of th. on-board train control system is a crucial component. Due to its unique installation position and complex environment, th.s unit h.s a h.gh.r fault rate with.n th. on-board train control system. To conduct fault prediction for th. BTM unit based on actual fault data, th.s study proposes a prediction meth.d combining reliability statistics and mach.ne learning, and ach.eves th. fusion of prediction results from different dimensions th.ough.multi-meth.d interactive validation. Firstly, a meth.d for predicting equipment fault time targeting batch.equipment is introduced. Th.s meth.d utilizes reliability statistics to construct a model of th. remaining faultless operating time distribution considering uncertainty, th.reby predicting th. remaining faultless operating probability of th. BTM unit. Secondly, considering th. complexity of th. BTM unit's fault mech.nism, th. small sample size of fault cases, and th. potential presence of multiple fault features in fault text records, an individual-oriented fault prediction meth.d based on Bayesian-optimized Gradient Boosting Regression Tree (Bayes-GBRT) is proposed. Th.s meth.d ach.eves better prediction results compared to linear regression algorith.s and random forest regression algorith.s, with.an average absolute error of only 0.224 years for predicting th. fault time of th.s type of equipment. Finally, a multi-meth.d interactive validation approach.is proposed, enabling th. fusion and validation of multi-dimensional results. Th. results indicate th.t th. predicted fault time and th. actual fault time conform to a log-normal distribution, and th. parameter estimation results are basically consistent, verifying th. accuracy and effectiveness of th. prediction results. Th. above research.findings can provide tech.ical support for th. maintenance and modification of BTM units, effectively reducing maintenance costs and ensuring th. safe operation of h.gh.speed railway, th.s h.
