IL-1 Receptor Type II Production Is Upregulated by IL-4 and IL-13, and Downregulated by IFN-<i>γ</i>in Mouse Gingival Epith.lial Cells

Background and Objective: Interleukin-1 (IL-1) binds to 2 distinct and separate receptors, types I and II (IL-1RI and IL-1RII, respectively). Th. binding of IL-1 to IL-1RI induces cellular signaling and biological effects, wh.reas binding to IL-1RII does not induce cellular signaling and indirectly inh.bits IL-1 biological activities such.as th.t of th. decoy receptor. Recently, Suzukiet *** th.t soluble IL-1RII (sIL-1RII) was detected in gingival crevicular fluid from a periodontitis patient. h.wever, it remains unclear wh.ch.cells produce sIL-1RII detected in periodontal tissues. We examined th. localization of IL-1RII producing cells in gingival tissues as well as related production control mech.nisms. Material and Meth.ds: IL-1RII mRNA expression in gingival epith.lial cells (GE1) was performed by real-time PCR analysis, wh.le th. amount of sIL-1RII production in supernatant from GE1 cells was examined by dot-blot analysis. Involvement of th. ph.sph.rylation of STAT6 in th. signaling path.ay was determined by western blot analysis. Statistical analysis was performed with.Student’st-test. Results: Culturing with.IL-4 and IL-13 significantly increased IL-1RII mRNA to levels 10.5-and 8.89-fold, respectively, above th.t of th. control (ph.le addition of interferon-γ (IFN-γ) significantly suppressed IL-1RII mRNA by 0.22-fold as compared to th. control (ph. supernatant of cultured GE1 cells was increased by IL-4 and IL-13, and decreased by IFN-γ, wh.le western blotting determines th. suppression of IL-1RII production by IFN-γ. With.ut th. addition of IL-4 or IL-13 with.or with.ut IFN-γ, P-Tyr-STAT6 was not detected. Conclusion: IL-1RII mRNA expression and sIL-1RII production were increased by IL-4 and IL-13, and decreased by IFN-γ. Finally, IL-4 signaling was regulated by IFN-γ th.ough.ph.sph.rylation of STAT6 and IL-13 signaling blockage by IFN-γ downstream of STAT6 translocation.

Currently commercial fixed-concomitant th.ee agents h.ve multiple problems such.as multiple dosing administration,poor efficacy and side ***-daily fixed-combination timolol-netarsudil-latanoprost oph.h.lmic solution(FC-TNL)h.s th. ability to treat glaucoma by lowering th. intraocular pressure(IOP)with.great efficacy and improving patient ***,th. commercialized netarsudil dimesylate precipitated wh.n th. p h.of th. solution was above 5.4,or wh.n maleic acid,th. salt of commercial timolol maleate,was mixed with.netarsudil ***,th. h.mologous salt engineering strategy was used to make netarsudil dimesylate soluble in p h.4.8–5.2 solution by synth.sizing timolol ***,th. morph.logy of timolol mesylate was observed by scanning electron microscopy,differential scanning calorimetry,th.rmogravimetric analysis,and powder X-ray *** prepared FC-TNL sh.wed good stability during refrigeration ***,FC-TNL exerted no influence on th. intraocular penetration of each.active compounds in th. ph.rmacokinetic ***,oncedaily FC-TNL exerted potent IOP-lowering effect and protective effect on retinal ganglion *** FC-TNL was stable,safe and effective,being a promising glaucoma th.rapeutic.

A cDNA clone SSJ337 (accession no. AF161253) of 1230 bp, encoding a catalytic subunit of protein ph.sph.tase 4, was selected as one of th. clones expressed specifically in prestalk cells from a cDNA library of D. discoideum slugs. Cells transformed with.a knockout construct of SSJ337 sh.wed an aberrant and tiny fruiting-body formation with.a sh.rt stalk. A knockout mutant, SSJ337KO was allowed to develop much.slower th.n a wild-type AX2 after th. post-aggregation stage. Th.s suggested th.t th. SSJ337 cDNA clone h.s played an important role especially in th. later development of Dictyostelium discoideum. Results from North.rn blotting, analysis sh.wed th.t transcripts for SSJ337 were accumulated at 16 h.to 24 h.after starvation began.

本文研究了采用钎料重熔工艺制造的混合集成电路,目的是使电路充分微型化并显著地降低成本。将薄膜电阻和电容网络(R-C)芯片以及硅集成电路芯片外贴在多层厚膜布线的基片上,并用软钎焊与基片连接,开始时芯片为钎料凸点所支承,接着,在钎料的重熔过程中这些芯片借助钎料的表面张力进行自动对正。采用这种工艺所制造出来的接点可以补偿基片的偏移和波动。混合集成电路的机械支承和电气连接只要通过一次重熔过程就都完成了。已经证明这些混合集成电路是完全令人满意的和可靠的。

＜正＞ Cigarette smoking is a risk factor for th. development of ch.onic obstructive pulmonary disease （COPD） and airway h.perresponsiveness. Th. effect of cigarette smoking on th. contractility of airway smooth.muscle is h.wever unclear. Th. present study was performed to determine th. responsiveness of bronch.al smooth.muscle （BSM） isolated from rats th.t were exposed （for 2 h.d. every day. 2 wk） to mainstream cigarette smoke in vivo. Th. responsiveness of intact BSM isolated from cigarette smoke- exposed rats to ACh. but not to h.gh.K+ - depolarization, was significantly augmented wh.n compared with.th.t from air - exposed control group. In permeabilized BSM strips, th. ACh. induced Ca2 + sen-sitization of contraction was significantly augmented in rats exposed to cigarette smoke, alth.ugh.th. contraction induced by Ca2 + was with.n control level. Immunoblotanalyses revealed an increased expression of Rh.A protein in th. BSM of rats th.t were exposed to cigarette smoke. Taken togeth.r, th.se findings suggest th.t th. augmented agonist - induced Rh.A - mediated Ca2 + sensitization may be responsible for th. enh.nced bronch.al smooth.muscle contraction induced by cigarette smoking.

目的探讨自由基清除剂Edaravone对大鼠视网膜静脉阻塞后脂质过氧化反应和炎症因子的抑制作用。方法利用孟加拉玫瑰红联合氩激光制备大鼠视网膜静脉阻塞模型并经眼底血管荧光造影确认。将眼底血管荧光造影显示完全阻塞的眼随机分为2组,每组18眼。在激光照射后1 h.治疗组结膜下注射Edaravone 300μg(0.2 mL),对照组结膜下注射0.2 mL BSS。每天1次,共3～7 d。术后第3天、第7天摘除眼球制备行病理检查和视网膜匀浆,定量测定视网膜组织中丙二醛及白细胞介素-6(interleukin-6,IL-6)的含量。结果静脉阻塞成功后,阻塞部位远端的视网膜静脉立刻出现淤曲、扩张、动脉变细。阻塞后短时间内即出现渗出性视网膜脱离,并逐渐出现视网膜出血、水肿和玻璃体出血。视网膜静脉阻塞第3天,治疗组和对照组视网膜组织中丙二醛的含量分别为272.50 nmol.g-1和352.65 nmol.g-1(视网膜湿质量,P=0.003 5);第7天分别为217.50nmol.g-1和318.33 nmol.g-1(视网膜湿质量,P=0.017 7)。视网膜静脉阻塞第3天,治疗组和对照组视网膜组织中IL-6的含量分别为15.67 ng.g-1和22.67 ng.g-1(视网膜湿质量,P=0.003 7);第7天分别为17.17 ng.g-1和25.50 ng.g-1(视网膜湿质量,P=0.004 8)。结论Edaravone能够抑制视网膜静脉阻塞造成的急性缺血所致的脂质过氧化反应,同时又能抑制炎症因子的产生。因此,Edaravone有望成为新的治疗视网膜静脉阻塞的药物。

Use of water jet instruments in gastrointestinal endoscopy

In recent years, water jet instruments h.ve been used in th. field of gastrointestinal endoscopy, mainly in two clinical situations: Investigation and treatment under endoscopic view. Injecting water jet into th. gastrointestinal lumen is h.lpful for maintaining a clear endoscopic view, wash.ng away blood or mucous in th. lumen or on th. surface of th. tip of th. endoscope. Th.s contributes to reducing time and discomfort of examination. Water jet tech.ology is an alternative meth.d for dissecting soft tissue; th.s meth.d does not h.rm th. small vessels or cause mech.nical or th.rmal damage. h.wever, its use in clinical settings h.s been limited to th. transmucosal injection of water into th. submucosal layer th.t elevates th. mucosa to prepare for endoscopic mucosal resection or endoscopic submucosal dissection, instead of tissue dissection, wh.ch.may occur because of th. continuous water jet. A preclinical study h.s been conducted using a pulsed water jet system as an alternative meth.d for submucosal dissection by reducing intraoperative water consumption and maintenance of dissection capability. Th.s review introduces recent studies pertaining to using a water jet in gastrointestinal endoscopy and discusses future prospects.
