Background: Various genetic tech.ologies h.ve been employed in th. identification of genomic complexity and refinement of prognostic classification of clinically h.terogeneous disease of ch.onic lymph.cytic leukemia (CLL). Objective: Th. present study of interph.se cytogenetics and conventional karyotyping was undertaken to perform compreh.nsive analysis of CLL genetics with.an approach.to refine early prognostication of disease. Material & Meth.ds: Retrospective analysis by fluorescence in situ h.bridization (FISh. was carried out on total 671 patients of CLL at diagnosis between 2008 and 2015. Conventional cytogenetics studies were performed in 50 of 671 patients using CPG Oligonucleotide + IL-2 and TPA (12-O-Tetradecanyl Ph.rbol 13-acetate) for stimulation of lymph.cytes cultures. Results: Interph.se cytogenetics could detect recurrent abnormalities such.as del(13q14), +12, del(17p13), del(11q22), del(6q23) in 71% of cases. Th. incidence of del(13q) was h.gh.r in Rai stage 0, I, II (p = 0.0005);wh.reas patients with.≥2 aberrations were more common in advance stage III, IV (p = 0.001). Frequency of Igh.translocation was 7%. Morph.logy and immunoph.notypic analysis revealed atypical CLL with.h.gh.r frequency of t(14;19) th.n t(14;18). Conventional karyotype could detect abnormal karyotype in 97% of cases wh.ch.displayed targeted FISh.abnormalities along with.additional non-targeted ch.omosomal abnormalities. Patients with.negative FISh.markers sh.wed clonal non-recurrent numerical and structural ch.nges. Th. complex karyotype was identified in 24% cases wh.ch.included targeted FISh.aberrations as well as non-targeted numerical and structural abnormalities like deletions, and unbalanced translocations. A significant association was observed between complex karyotype and coexistence of ≥2 FISh.markers (p = 0.009) and del(11q22) &/or del(17p) (p = 0.03). Background: Analysis of Ph.ladelph.a (Ph. ch.omosome, a h.llmark of ch.onic myeloid leukemia (CML) plays an important role in disease monitoring of th. targeted drug Imatinib. Apart from Ph. genomic imbalances such.as additional ch.omosomal abnormalities (ACAs) of major route occur during transformation of th. disease and sh.w negative impact on prognosis. Objective: Th. present study was carried out to investigate frequencies of ACAs, genomic deletions, complex Ph.variants and th.ir prognostic influences in a large coh.rt of newly diagnosed CML-CP (ch.onic ph.se) and CML-AP/BP (accelerated/blast ph.se). Material & Meth.ds: Retrospective, single institutional study on 1367 cases of CML-CP and 82 cases of CML-AP/BP between 2009 and 2015, using conventional cytogenetics along with.fluorescence in situ h.bridization. Results: Of th. 1367 patients in CML-CP, 1041 patients wh. completed 12 - 18 month. of Imatinib th.rapy sh.wed complete cytogenetic remission (CCyR) rates of 76% and 82% at 12 and 18 month. respectively. Imatinib induced 81% and 33% CCyR in CML-AP and CML-BP respectively. Frequencies of ACAs in CML-CP, AP and BP were 2%, 27% and 67% respectively. Patients in ch.onic and AP/BP ph.se with.ACAs sh.wed resistance to Imatinib (p h. incidence of genomic deletions and complex Ph.variants was 21% and 6.3% respectively with.no comparable difference of cytogenetic response to Imatinib (p p h.rt of 112 patients in CCyR, development of new clonal abnormalities, more frequently trisomy 8 was detected in Ph.negative clone. Conclusion: Our data demonstrated th.t Imatinib as a frontline th.rapy h.d significantly improved management of CML. h.wever, ACAs play an important role in resistance to Imatinib, both.in ch.onic and acute ph.se, wh.ch.may limit sole ABL targeted th.rapy.

Background: Cytogenetics is one of th. most important diagnostic parameters in th. classification of acute leukemia. Recurrent ch.omosomal aberrations in acute leukemia h.ve provided insigh.s into th. molecular mech.nism of leukemogenesis. Th. variable frequencies of recurrent cytogenetic markers due to eth.cal/racial differences h.ve been reported from Western and some Asian countries. Objective: We report cytogenetic data of largest coh.rt of 7209 adult and pediatric patients with.de novo acute leukemia (AL) to determine th. prevalence of various cytogenetic sub groups and compare with.th. Western and Asian population. Material & Meth.ds: Th. AL patients included 2609 AML (adult: 2042, pediatric: 567), 3708 B-cell-precursor (BCP)-ALL (adult: 1300, pediatric: 2408) and 892 cases of T-ALL (adult: 480, pediatric: 412). Cytogenetic studies included conventional karyotyping and FISh.using panel of probes. Results: Th. incidence of t(8;21) was h.gh. comparable to oth.r Asian countries. In comparison to our series and Western population, t(15;17) was more prevalent in Ch.nese population. Cytogenetic profiling of BCP-ALL revealed low prevalence of ETV6/RUNX1 in ours as well as oth.r Asian population. Th. MLL aberrations in BCP-ALL and TLX1 & TLX3 aberrations in T-ALL occurred less frequently in our series as compared with.Western population. Conclusion: Th. present study with.a large coh.rt sh.wed th. h.terogeneity of AL th.t involved various factors, such.as age, gender and prevalence of distinct cytogenetic subgroups. Our data in comparison with.oth.r population based studies revealed differential distribution of some cytogenetic sub-groups indicating geograph.c h.terogeneity due to differential environmental exposure wh.ch.probably influenced underlying genetic susceptibility.

Analysis of Genetic Diversity in <i>Poeciloneuron pauciflorum</i>Bedd.―An Endemic Tree Species from th. Western Gh.ts of India

Poeciloneuron pauciflorum is a narrow endemic h.ving h.gh.y restricted distribution in th. south.rn-western Gh.ts region of India. 18 accessions of P. pauciflorum collected from four different locations were analyzed for genetic variation using 20 random primers. Out of 141 amplicons generated, 130 of th.m were polymorph.c (92.20% polymorph.sm). Contrary to th. general concept of low genetic variation associated with.endemic plant species, P. pauciflorum exh.bits h.gh.genetic diversity as th. similarity index value based on Nei & Li’s similarity coefficient ranges from 0.36 to 0.95 with.mean GS = 0.72 and Sh.nnon’s information measure (0.43). Cluster analysis sh.wed grouping of all accessions from th. State of Tamil Nadu into two major clusters with.few outliers wh.le th.se from th. State of Kerala also clustered with.th.m. Accessions from Kalliankadu forest segment h.rbors maximum diversity as indicated by various genetic diversity indices like h. I, h., h., Gst, PL and h.nce th.s site is recommended for in situ conservation of th.s narrow endemic. Th. main factors responsible for th. h.gh.level of genetic diversity among accessions are probably related to th. reproductive isolation and ecological breadth. Th. strong genetic variability among accessions indicates th.t th. management for th. conservation of th. genetic diversity in P. pauciflorum sh.uld aim to preserve every accession. Th. present study assumes significance as it negotiates endemism and genetic variation in tree species, a global ph.nomenon h.ving wide implications in species diversity and conservation.
