In situ measurement of ECm rheology and microheterogeneity in embedded and overlaid 3D pancreatic tumor stroma co-cultures via passive particle tracking

Tumor growth is regulated by a diverse set of extraellular infuences,including paracrine crosstalk with stromal partners,and biophysical interactions with surrounding cells and *** elucidating the role of physical force and the mechanical properties of the extracellular matrix(ECm)itself as regulators of tumor growth and invasion have been greatly catalyzed by the use of in ritro three-dimensional(3D)tumor *** systems provide the ability to systematically isolate,manipulate,and evaluate impact of stromal components and extracellular mechanics in a platform that is both conducive to imaging and biologically ***,recognizing that mechanoregulatory crosstalk is bi-directional and fully utilizing these models requires complementary methods for in situ measurements of the local mechanical ***,in 3D tumor/fbroblast co-culture models of pancreatic canocer,a discase characterized by its prominent stromal involvement,we evaluate the use of particle-tracking microrhoology to probe dynamic mechanical *** videos of fuorescently labeled polystyrene micro-spheres embedded in ollagen I ECm,we measure spatiotemporal changes in the Brownian motion of probes to report local ECm shear modulus and *** approach reveals st ffening of collagen in fibroblast co-cultures relative to cultures with cancer cells only,which exhibit degraded ECm with heterogeneous *** further show that these effects are dependent on culture geometry with contrast ing behavior for embedded and overlay *** addition to potential application to screening stroma targeted therapeutics,this work also provides insight into how the compoition and plating geometry impact the mechanical propertios of 3D cell cultures that are increasingly widey used in cancer biology.

Electromyographic and mechanomyographic Responses During Isokinetic Leg Extensions in Children Versus Adolescents

Purpose The purpose of this study was to examine peak torque(PT),mean power(mP),electromyographic(EmG)amplitude,mechanomyographic(mmG)amplitude,and neuromuscular efficiency from EmG and mmG amplitude(NmE_(EmG)and NmE_(mmG),respectively)across the velocity spectrum in children versus *** Seventeen children(n=8 males,n=9 females)and 22 adolescents(n=11 males,n=11 females)*** completed maximal concentric isokinetic leg extensions at 60,120,180,240 and 300°/***,mP,EmG amplitude,and mmG amplitude were quantified during all muscle ***_(EmG)and NmE_(mmG)were quantified by expressing PT relative to EmG and mmG amplitude,respectively,for each muscle *** PT,mP,NmE_(EmG),and NmE_(mmG)were greater for the adolescents than the children collapsed across sex and veloc-ity(P≤0.021).There were no significant differences for EmG amplitude(P≥0.051),while mmG amplitude increased significantly from 60 to 240°/s(Pm 240 to 300°/s(PmG and mmG amplitude(NmE_(EmG)and NmE_(mmG),respectively),in conjunction with measurements of torque and power production,may provide practitioners a unique method of examining growth and development-related augmentations in neuromuscular function during dynamic movements.

Effects of Eccentric Pre-loading on Concentric Vertical Jump Performance in Young Female Athletes

Purpose This study examined changes in vertical jump performance with progressively greater eccentric pre-loading in relation to growth and development in young female *** Twenty young female athletes ranging from 9 to 17 years old performed the following vertical jumps in random order:static jumps(SJs),counter-movement jumps(CmJs),and drop jumps(DJs)from drop heights of 20,30,and 40 cm(DJ20,DJ30,and DJ40,respectively).measurements included peak force(PF),peak rate of force development(RFD),peak power(PP),eccentric impulse(ECC),and concentric impulse(CON).measurements of growth included age,maturity offset,height,body mass,fat-free mass,and thigh muscle cross-sectional area(CSA).Results PF increased from the SJ-DJ20(Pm DJ20-DJ40(P=1.000).RFD remained the same from SJ-CmJ(P=1.000),increased from CmJ-DJ20(Pm DJ20-DJ40(P=0.874).PP increased from the SJ-CmJ(Pm the CmJ-DJ40(P≥0.486).CON remained the same across all verti-cal jumps(P=1.000),while ECC increased from the SJ-DJ40(P≤0.038).Jump height(JH)increased from the SJ-CmJ(Pm CmJ-DJ20(Pm DJ20-DJ40(P=1.000).The change in PP from the SJ-CmJ(ΔCmJ-SJ)was related to all measurements of growth except CSA(r=0.558-0.815).Conclusion Young females produced greater power during the CmJ than SJ,but equivalent power from the CmJ-DJ40,despite increases in ***,ΔCmJ-SJ was not related to CSA,which suggests other underlying mechanisms affect stretch-shortening cycle utilization in young female athletes.

Preliminary Estimation of Paleoproductivity via TOC and Habitat Types:Which method Is more Reliable?—A Case Study on the Ordovician-Silurian Transitional Black Shales of the Upper Yangtze Platform, South China

New total organic carbon （TOC） data from the two Ordovician-Silurian transitional graptolite-bearing black shale intervals, the Wufeng （五峰） Formation and the Longmaxi （龙马溪） Formation in Central Guizhou （贵州） and West Hubei （湖北）, respectively, as well as previously reported TOC data from the same intervals in other places on the Yangtze platform of South China, have been used to produce an initial estimate of the primary paleoproductivity via a conventional inverse method （i.e., ***）. The values of the *** are estimated to be 32 （43-21） gC/（m^2·a）（Wufeng Formation） and 21 （27-16） gC/（m^2·a）（Longmaxi Formation）. Also, simultaneously, the habitat types （i.e., HT; cf., BA： benthic assemblage） and their temporal and spatial changes have been documented from the same succession, and an initial estimate of the primary paleoproductivity has been produced using a forward method （i.e., Rpp-forward） Beingbounded mainly by the peritidal to inner-shelf environment shelly-facies or mixed-facies successions with BA1 to BA3 faunas both at the top and the base, which indicates the habitat types from HT Ⅱ to HT Ⅲ2, the biohabitat type of the two graptolite-bearing black shale intervals can be limited to HT III to HT IV, corresponding to the inner shelf to the outer shelf, with depths from roughly 60 m to 200-300 m. Based on the current data from the South China Sea and the southern part of the East China Sea, values of Rpp-forward should be about 100 to 400 gC/（m^2·a）. The difference in the results via the two methods suggests that paleoproductivity estimates from the geological strata need to be made cautiously, with particular attention paid to the paleogeographic setting, oxic-anoxic conditions, as also the preservation factor of organic carbon.

Leg Extension Strength, Explosive Strength, muscle Activation, and Growth as Predictors of Vertical Jump Performance in Youth Athletes

Purpose The purpose of this study was to examine the relationships among leg extension strength,explosive strength,muscle activation,and measurements of growth to predictions of vertical jump performance in youth *** Height,body mass,skinfolds,and quadriceps femoris muscle cross-sectional area were measured in 39 sports active children(20 females,19 males,age=12.52±0.62 years old).Peak torque(PT),rate of torque development(RTD),rate of velocity development(RVD),and rate of electromyographic amplitude rise(RER)were measured during isometric and isokinetic leg ***,RTD,and RVD were expressed in absolute terms and normalized to body *** jump height(JH)and peak power(PP)were assessed during static(SJ),counter-movement(CmJ),and drop(DJ)*** JH exhibited greater correlations with PT normalized to body mass(r=0.387-0.758)than absolute PT(r=0.338-0.417),whereas PP exhibited greater correlations with absolute PT(r=0.368-0.837).Only negligible to moderate relation-ships existed between JH and PP across all jumps(r=0.053-0.605).Over 50%of the variability in PP was predicted in 24 of 30 regression models with absolute muscle strength,muscle activation,and measurements of growth,while only 6 of 30 models predicted more than 50%of the variability in *** Overall,absolute static and dynamic muscle strength,muscle activation,and growth better explained PP meas-ured during vertical jumps than estimated JH.

Viticulture,like other fields of agriculture,is currently facing important challenges that will be addressed only through sustained,dedicated and coordinated *** the methods used in biology have evolved tremendously in recent years and now involve the routine production of large data sets of varied nature,in many domains of study,including grapevine research,there is a need to improve the findability,accessibility,interoperability and reusability(FAIR-ness)of these *** the heterogeneous nature of the data produced,the transnational nature of the scientific community and the experience gained elsewhere,we have formed an open working group,in the framework of the International Grapevine Genome Program(***),to construct a coordinated federation of information systems holding grapevine data distributed around the world,providing an integrated set of interfaces supporting advanced data modeling,rich semantic integration and the next generation of data mining *** achieve this goal,it will be critical to develop,implement and adopt appropriate standards for data annotation and *** development of this system,the GrapeIS,linking genotypes to phenotypes,and scientific research to agronomical and oeneological data,should provide new insights into grape biology,and allow the development of new varieties to meet the challenges of biotic and abiotic stress,environmental change,and consumer demand.

Cholangiocarcinoma, gone without the Wnt?

Cholangiocarcinoma(CCA) is a relatively rare malignancy of the intra- or extra-hepatic bile ducts that is classified according to its anatomical localization as intrahepatic, perihilar or distal. Overall, CCA has a dismal prognosis due to typical presentation at an advanced irresectable stage, lack of effective non-surgical treatments, and a high rate of disease recurrence. CCA frequently arises on a background of chronic liver inflammation and cholestasis. Chronic inflammation is accompanied by enhanced cell turnover with generation of additional inflammatory stimuli, and a microenvironment rich in pro-inflammatory mediators and proliferative factors that enable accumulation of mutations, transformation and expansion of mutated cells. A recent study by Boulter et al implicates the Wnt signaling cascade in cholangiocarcinogenesis. Wnt ligands Wnt7 B and Wnt10 A were found to be highly overexpressed in human CCA tissue. Wnt7 B protein was present throughout the tumor stroma, and often co-localized with a subset of CD68+ macrophages. To address in a direct manner whether Wnt signaling is engaged in development of CCA, Boulter et al explored the Wnt signaling pathway in an experimental model that recapitulates the multi-stage progression of human *** ligands found to be elevated in human CCA were also upregulated during the course of CCA development following thioacetamide treatment. Wnt10 a increased during the(pre-cancerous) regenerative phase, while Wnt7 b induction paralleled tumor growth. Along with upregulation of target genes, the findings demonstrate that the canonical Wnt pathway is progressively activated during cholangio-carcinogenesis. macrophage depletion,eliminating a major source of Wnt7 b, prevented activation of the canonical Wnt cascade, and resulted in reduced number and volume of tumors in this model. moreover,specific inhibitors of the canonical Wnt pathway(ICG-001 and C-59) caused reduction of tumor area and number,in xenograft and thioacetami
