Transcriptome and metabolic Profiling Provides Insigh into Betalain Biosynthesis and Evolution in mirabilis jalapa

Betalains are tyrosine-derived pigments that occur solely in one plant order, the Caryophyllales, where they largely replace the anthocyanins in a mutually exclusive manner. In this study, we conducted multi-species transcriptome and metabolic profiling in mirabilisjalapa and additional betalain-producing species to identify candidate genes possibly involved in betalain biosynthesis. Among the candidates identified, betalainrelated ytochrome P450 and glucosyltransferase-type genes, which catalyze tyrosine hydroxylation or （hydroxy）cinnamoyl-glucose formation, respectively, were further functionally characterized. We detected the expression of genes in the flavonoid/anthocyanin biosynthetic pathways as well as their metabolite intermediates in betalain-accumulating m. jalapa flowers, and found that the anthocyanin-related gene ANTHOCYANIDIN SYNTHASE （mjANS） is highly expressed in the betalain-accumulating petals. However, it appears that mjANS contains a significant deletion in a region spanning the corresponding enzyme active site. These findings provide novel insights into betalain biosynthesis and a possible explanation for how anthocyanins have been lost in this plant species. Our study also implies a complex, non-uniform history for the loss of anthocyanin production across betalain producers, previously assumed to be strictly due to diminished expression of anthocyanin-related genes.

Temperature and water flow through a culvert beneath the Alaska Highway near Beaver Creek,Yukon,were measured at hourly intervals between June and October *** data were used to simulate the effect of the culvert on the thermal regime of the road embankment and subjacent permafrost.A 2-D thermal model of the embankment and permafrost was developed with TEmP/W and calibrated using field *** relations were obtained between water temperatures at the entrance to the culvert,flow into the culvert,and water temperatures inside the *** temperatures at the entrance and inside the culvert had a linear relation,while water temperatures inside the culvert and water flow were associated by a logarithmic relation.A multiple linear regression was used to summarize these *** this relationship,changes in the flow rate and water temperatures at the entrance of the culvert were simulated to obtain predicted water temperatures in the *** temperatures in the culvert were used in the thermal model to determine their effects on the ground thermal regime near the *** of ±10% in water flow rate had no impact on the thermal regime underneath the *** of water temperature at the entrance of the culvert had a noticeable influence on the thermal regime.A final simulation was conducted without insulation beneath the *** thaw depth was 30 cm with insulation,and 120 cm without insulation,illustrating the importance of insulation to the ground thermal regime.

The realization of a mixed-phase microstructure in strained BiFeO_(3)(BFO)thin films has led to numerous novel effects derived from the coexistence of the tetragonal-like monoclinic phase(T phase)and rhombohedral-like monoclinic phase(R phase).Strong strain and polarization diiferences between the phases should result in a high level of transformation plasticity,which enables the continuous alteration of the relative proportion of R and T states in response to external *** the potential for utilizing such plasticity to control mixed-phase populations under external stimuli is evident,direct experi・mental evidence backed by equilibrium predictions has not yet been fully *** we demonstrate deterministic control of mixed-phase populations in an epitaxially strained BFO thin film through the application of localized stresses and electric fields in a reversible manne匚The results illustrate and rationalize deterministic control of mixed phases in strained BFO films,which could be crucial in tuning their functional *** findings also highlight a new multiparametric technique in the scanning probe lithography toolbox based on tip-assisted electric and strain field manipulation of functional properties that might find application beyond the ferroelectric domain and structural phase lithography.

Pain sensation may appear under long-lasting mechanical stimulation. Although people have the experience that pain sensation generally decreases with time while the stimulation remains, the underlying mechanism remains elusive. We experimentally studied the thermal and strain rate- dependent viscoelastic behavior of skin in uniaxial stretch and numerically investigated the effects of temperature and strain rate on pain sensation. The results indicate that the viscosity of skin tissue decreases with increasing temperature and reducing strain rate, which subsequently decreases the discharge frequency of skin nociceptor and thus relieves the pain sensation. The results would contribute to the understanding of pain relief mechanism and optimizing for mechanical treatment.

most olefins （e.g., ethylene and propylene） will continue to be produced through steam cracking （SC） ofhydrocarbons in the coming decade. In an uncertain commodity market, the chemical industry is investingvery little in alternative technologies and feedstocks because of their current lack of economic viability,despite decreasing crude oil reserves and the recognition of global warming. In this perspective, some of themost promising alternatives are compared with the conventional SC process, and the major bottlenecks ofeach of the competing processes are highlighted. These technologies emerge especially from the abundanceof cheap propane, ethane, and methane from shale gas and stranded gas. From an economic point of view,methane is an interesting starting material, if chemicals can be produced from it. The huge availability ofcrude oil and the expected substantial decline in the demand for fuels imply that the future for proventechnologies such as Fischer-Tropsch synthesis （FFS） or methanol to gasoline is not bright. The abundance ofcheap ethane and the large availability of crude oil, on the other hand, have caused the SC industry to shiftto these two extremes, making room for the on-purpose production of light olefins, such as by the catalyticdehydrogenation of orooane.

AIm:To evaluate the association between CYP1A1 and GSTs genetic polymorphisms and susceptibility to esophageal squamous cell carcinoma(SCC)and esophageal adenocarcinoma(ADC)in a high risk area of northwest of France. mETHODS:A case-control study was conducted to investigate the genetic polymorphisms of these enzymes (CYPIAI*2C and GSTP1 exon 7 Val alleles,GSTmI*2/*2 and GSTTl *2/*2 null genotypes).A total of 79 esophageal cancer cases and 130 controls were recruited. RESULTS:GSTmI*2/*2 and CYPIAI*IA/*2C genotype frequencies were higher among squamous cell carcinomas at a level dose to statistical significance(OR =1.83,95% CI 0.88-3.83,P=0.11;OR=3.03,95% CI 0.93-9.90,P=0.07, respectively).For GSTP1 polymorphism,no difference was found between controls and cases,whatever their histological *** frequency of GSTT1 deletion was observed in ADC group compared to controls with a statistically significant difference(OR=13.31,95% CI 1.66-106.92,Pmour with tobacco *** ADC,our results suggest 3 distinct hypotheses: (1)activation of exogenous procarcinogens,such as small halogenated compounds by GSTT1;(2)contribution of GSTT1 to the inflammatory response of esophageal mucosa,which is known to be a strong risk factor for ADC, possibly through leukotriene synthesis;(3)higher sensitivity to the inflammatory process associated with intracellular depletion of glutathione.

This paper reviews and discusses recent developments in passively mode-locked vertical external cavity surface emitting lasers （mL-VECSELs） for short pulse generation at 1.55 gin. After comparing mL- VECSELs to other options for short pulse generation, we reviewed the results of mL-VECSELs operating at telecommunication wavelength and point out the chal- lenges in achieving sub-picosecond operation from a mL- VECSEL at 1.55 gm. We described our recent work in the VECSELs and semiconductor saturable absorber mirrors （SESAms）, their structure design, optimization and characterization, with the goal of moving the pulse width from picosecond to sub-picosecond.
