spontaneous and Bleomycin-Induced γH2AX signals in CHO9 Metaphase Chromosomes

In eukaryotes, a cascade of events named DNA damage response (DDR) has evolved to handle DNA lesions. DDR engages the recruitment of signaling, checkpoint control, repair and chromatin remodeling protein complexes, allowing cell cycle delay, DNA repair or induction of apoptosis. An early DDR event involves the phosphorylation of the histone variant γH2AX on serine 139 (H2AX139 phosphorylation) originating the so-called γH2AX. DDR-related H2AX139 phosphorylation have been extensively studied in interphase nuclei. More recently, γH2AX signals on mitotic chromosomes of asynchronously growing cell cultures were observed. We performed a quantitative analysis of γH2AX signals on γH2AX immunolabeled cytocentrifuged metaphase spreads, analyzing the γH2AX signal distributions of CHO9 chromosomes harboring homologous regions both in control and bleomycin (BLM)-treated cultures. We detected γH2AX signals in CHO9 chromosomes of controls which significantly increase after BLM-exposure. γH2AX signals were uniformly distributed in chromosomes of controls. However, the γH2AX signal distribution in BLM exposed cells was significantly different between chromosomes and among chromosome regions, with few signals near the centromeres and a tendency to increase towards the telomeres. Interestingly, both basal and BLM-induced γH2AX signal distribution were statistically equal between CHO9 homologous chromosome regions. Our results suggest that BLM exerts an effect on H2AX139 phosphorylation, prevailing towards acetylated and gene-rich distal chromosome segments. The comparable H2AX139 phosphorylation of homologous regions puts forward its dependence on chromatin structure or function and its independence of the position in the karyotype.

The Mediterranean fruit fly (Medfly), Ceratitis capitata (Wiedemann) is one of the major pests of fruit crops in Argentina and it is a phytosanitary barrier to the export of fresh fruits. In the Province of san Juan, located in the central-eastern region of Argentina known as Cuyo, control strategies against Medfly in fruit-producing irrigated-valleys have been implemented by the National Fruit Fly Control and Eradication Program (ProCEM) jointly with the provincial government and the producers. This program uses an area-wide integrated pest management approach that includes the use of environment-friendly strategies to suppress or eradicate Medfly, such as the use of the sterile insect technique and the application of new-generation bait sprays, and more recently the release of the Indo-Pacific parasitoid Diachasmimorpha longicaudata (Ashmead). This exotic larval-prepupal endoparasitoid is being mass-reared on larvae of Vienna 8 TsL C. capitata strain at the BioPlanta san Juan facility. In this respect, the objective of this study was to assess the effectiveness of laboratory-reared D. longicaudata females in order to find and successfully parasitize Medfly larvae in different host fruit species once released under semi-arid environmental conditions in ecologically isolated fruit-growing valleys of san Juan. Ground releases were used to disperse parasitoids in 6 fruit-producing valleys. In total, 40,000 adult parasitoids were released at places with various Medfly host plants bearing fruits and in which no insecticides were regularly applied, such as backyards and small orchards. 119 D. longicaudata adults were recovered from 6 Medfly-infested fruit species that were collected in 5 release sites (Pocito, Zonda, santa Lucía, Caucete, and Rivadavia). The highest number of parasitoids was recovered from fig, followed by grape, rose, orange, tangerine, and persimmon. Data provided from this study open up the possibility of implementing a parasitoid mass-release program

Carpal Tunnel syndrome: A Marker for Amyloidosis

Introduction: Amyloidosis are systemic conditions and carpal tunnel syndrome (CTs) precedes the principal systemic complications and can be used as an early marker. Our objective was to determine the frequency of amyloid deposition in idiopathic CTs and its systemic impact. Methods: We retrospectively evaluated patients with CTs between september 2019 to January 2020. samples from the anterior carpal ligament were pathologically evaluated and amyloid deposition was confirmed by apple-green birefringence on polarized light using Congo red stain. When amyloid was detected we performed genetic testing for transthyretin variants (ATTRv), immunofixation electrophoresis in serum and urine for light chains and multidisciplinary evaluation. Results: Thirty consecutive patients were included, 19 women, 11 men, mean age 70 years old (range 42 - 89 years). We identified 3 patients (10%) with amyloid deposits (mean age: 78.6 years, 2 men, 1 woman). Genetic testing for ATTRv and light chains studies were negative. During follow-up: The first patient required aortic valve replacement. The second patient developed progressive cardiac failure with syncopal episodes, atrioventricular block and atrial fibrillation and required a pacemaker and anticoagulation. The third patient had unexplained chronic edemas. The cardiac evaluation in all 3 patients revealed left ventricular hypertrophy and myocardial uptake (Perugini score > 2) in their nuclear bone scintigraphies with technetium pyrophosphate. Two patients were treated with tafamidis and one patient died due to refractory cardiac insufficiency. Discussion: Our findings underline the importance of investigating amyloidosis in idiopathic CTs. The identification of deposits allows early diagnosis of cardiac amyloidosis leading to timely intervention and treatment.

Testing the effectiveness of conservation management within biosphere reserves:the case of the Mexican deer mouse(Peromyscus mexicanus)as a bioindicator

Biosphere reserves are protected areas whose purpose is to combine conservation and sustainable ***,their effectiveness has not been tested sufficiently,especially from an ecological and genetic ap*** this sense,the Peromyscus genus represents an excellent bioindicator to address these questions,due to its short life and high evolutionary rate and *** conservation managers,genetic structure can increase the rate of loss of genetic diversity because alleles exclusive of a subpopulation are more likely to disappear as a consequence of genetic drift in comparison with a panmictic *** we analyzed the abundance,movement distances,morphology/morphometry and genetic structure of 3 populations of Mexican deer mouse(Peromyscus mexicanus)located in different protected zones of La Tigra National Park(Honduras).Our results are consistent among the 3 approaches and showed the highest values of abundance,morphometry and genetic diversity in the population located at the core zone,whereas non-statistically significant differences were found between buffer and transition zone populations,suggesting suitable effectiveness of conservation management in the core zone but a lack of ecological buffering function of the other *** addition,the low movement distances and high genetic structure among the studied populations provide evidence of poor conservation management in the buffer and transition ***,we discuss the utility of the novel methodology used in this work,combining morphometry,abundance and genetics,in testing the effectiveness of conservation strategies in biosphere reserves,and the value of the Peromyscus genus as a bioindicator.

The principal factor to determine the economical value of the products manufactured in the electronics industry is due to the productive yielding. This is important for the cost of the articles fabricated in this type of industrial plants installed in Mexicali city, where around 80% of companies are, and which fabricate electronic devices and systems, or have industrial electronic systems and machines to their manufacturing process. Mexicalicity is located in theBaja Californiastateof the northwest ofMexico, which is a border city with Calexico in theCaliforniastateof the United states of America (UsA). The region located in Mexicali, is a desert area. Geothermal plant is located in this area, which is an important industry and supplies electricity to this city and its valleys and some cities on southwest of United states for daily activities. This company emits hydrogen sulfide (H2s) as a main air pollutant that reacts with oxygen in the atmosphere, generating sulfur oxides (sOX). This chemical is dispersed to the city of Mexicali in which industrial plants are located with electronic control systems, and penetrates to indoor rooms. Those cause the corrosion process. The presence of corrosion leads to the deterioration of electrical connectors, the connections of electronic systems and the decreasing of the lifetime of these control systems. Other air pollutants that are considered as chemical agents which cause damage to materials used in the electronics industry, are the sulfurs and nitrogen oxides (NOX), emitted from the traffic vehicle and some industries. This causes the low productive yielding of electrical and electronic devices and systems used in the companies of this city, and is a major concern to specialized people, managers and owners. Risk of colorectal polyps in patients with sporadic gastric polyps:A case-control study

AIM:To assess the risk of colonic polyps,adenomas and advanced neoplastic lesions(ANL) in patients with sporadic gastric polyps,especially those with fundic gland polyps(FGP).METHODs:Clinical records of patients who had performed an upper and a lower digestive endoscopy between september 2007 and August 2008 were retrospectively analyzed.A case-control study was carried out,calling patients with gastric polyps as 'cases' and patients without gastric polyps as 'controls'.The risk of colonic polyps,adenomas and ANL(villous component ≥ 25%,size ≥ 10 mm,or high grade dysplasia) was assessed [odds ratio(OR) and its corresponding 95%CI].REsULTs:Two hundred and forty seven patients were analyzed:78 with gastric polyps(cases) and 169 without gastric polyps(controls).Among the cases,the majority of gastric polyps were FGP(80%,CI:69-88) and hyperplastic(20%,CI:12-31);25% had colonic polyps(25% hyperplastic and 68% adenomas,from which 45% were ANL).Among the controls,20% had colonic polyps(31% hyperplastic and 63% adenomas,from which 41% were ANL).The patients with sporadic FGP had an OR of 1.56(CI:0.80-3.04) for colonic polyps,an OR of 1.78(CI:0.82-3.84) for colonic adenomas,and an OR of 0.80(CI:0.21-2.98) for *** results were found in patients with gastric polyps in ***:The results of this study did not show more risk of colorectal adenomas or ANL neither in patients with sporadic gastric polyps nor in those with FGP.

Endogenous endophthalmitis is a rare condition with a poor long-term visual prognosis and significant mortality, often associated with the hematogenous spread of intravitreal infections and subsequent disruption of the blood-ocular barrier. Its anatomical proximity to the central nervous system (CNs) poses a high risk of infection dissemination, although cases documented in the literature are rare, and endogenous endophthalmitis is typically described as secondary to neuroinfections. We report the case of an 82-year-old female patient with a history of hypertension who presented with fever, decreased visual acuity, severe headache, chemosis, and conjunctival injection. Endogenous endophthalmitis was diagnosed, and antimicrobial treatment was initiated alongside surgical intervention by the ophthalmology service. However, the patient’s condition worsened neurologically, and streptococcus pneumoniae was identified in cerebrospinal fluid cultures. Despite intensive medical treatment, the patient’s clinical course was poor, leading to death.
