The effect of the application of ethanolic extract of “Haden” mango seeds to increase the antioxidant and antimicrobial capacity of fresh-cut mango flesh was evaluated. Phenolic HPLC analysis revealed that gallic acid was the major identified compound in the ethanolic extract (586.68 mg/g). Fresh-cut mango was treated for immersion within the ethanolic extract, packed and stored at 5℃ during 15 days. The content of phenolic compounds, antioxidant capacity and microbial growth of the treated mango were evaluated during the storage. Compared with the controls the treated fruit presented the highest content of the total phenolic (7.4 times), flavonoids (3.1 times), and antioxidant capacity DPPH·, TEAC and ORAC (2.9, 2.3 and 2.8 times, respectively), and showed a microbial reduction of 80% of mesophilic plate count and 97% of total molds. This study demonstrates the potential of phenolic compounds derived from mango seed as antimicrobials and antioxidants.

Determinants of percutaneous coronary intervention success in repeat chronic total occlusion procedures following an initial failed attempt

AIM To investigate the rates and determinants of success of repeat percutaneous coronary intervention(PCI) following an initial failed attempt at recanalising the chronic total occlusions(CTO) *** In 445 consecutive first attempt CTO-PCI procedures in our institution,procedural failure occurred in 149(33.5%). Sixty-four re-PCI procedures were performed in 58 patients(39%) all had a single CTO. Procedural and outcome data in the re-PCI population was entered into the institutional database. A retrospective analysis of clinical,angiographic and procedural data was performed. RESULTS Procedural success was achieved in 41(64%) procedures. Univariate analysis of clinical and angiographic characteristics showed that re-PCI success was associated with intravascular ultrasound(IVUS) guidance(19.5% vs 0%,P = 0.042),while failure was associated with severecalcification(30.4% vs 9.7%,P = 0.047) and a JCTO score > 3(56.5% vs 17.1% P = 0.003). Following multiple regression analysis the degree of lesion complexity(J-CTO score > 3),IVUS use,involvement of an experienced CTO operator and LAD CTO location were significant predictors of successful re-PCI. Overall the complication rate was low,with the only MACCE two periprocedural MI's neither of which required intervention. CONCLUSION Re-PCI substantially increases the overall success rate of CTO revascularization. Predictors of re-PCI success included the use of IVUS,the involvement of an experienced CTO operator in the repeat attempt and the location of the CTO.

Ethnicity has been associated with the incidence of diseases and consequently it is a cornerstone in medical genetic studies. It is mainly important in admixture populations, where the population stratification can produce spurious results that lead to erroneous conclusions. Consequently, population stratification has become one of the most important confounding factors in population-based genetic association studies, especially in Latino populations. Cartagena de Indias is a cosmopolitan city with dissimilar ancestry proportions due to recent miscegenation. This population mainly exhibits African and Amerindian matrilineal ancestries. Nevertheless, important asymmetries in the paternal genetic history related to the complex patterns of migration in the colonial period increase the male genetic diversity in this population. As a result of this recent admixture, population stratification has arisen, where each subpopulation is not equally represented. Consequently, the allele differences between cases and controls could be related with different frequencies among different population strata rather than the association of the genes with the disease. In order to define the patrilineal substructure of the Cartagena’s population, a total of 130 unrelated men were ancestrally studied using 15 Y-STR loci routinely employed in anthropological, forensic and population genetics. Our results show that Cartagena is an admixture population consisting of European (80%), Amerindian (10%) and African ancestries (10%), which are represented by haplogroups R1b and I2a (xI2a1), Q-M242/Q-M3, and E1b1a/E1b1b, respectively. Complex genetic patterns found in Cartagena’s population emphasize the importance to know the genetic variation in order to diminish the inconsistence for future genetic association studies. This study evaluated the nutritional characteristics of Brachiaria brizantha cultivars subjected to three cutting intensities in the course of one year. The experiment was conducted at the University of Rio Verde. The experiment was performed as a randomized 3 × 3 factorial in complete block design with three replications and repeated measures in the time. Three Brachiaria brizantha cultivars (Marandu palisadegrass, Xaraes palisadegrass, and Piata palisadegrass), were tested at three cutting intensities (10, 20, and 30 cm sward height). The evaluations were conducted on the same plots throughout one year and during all four seasons (autumn, winter, spring, and summer). The results showed that the Piata palisadegrass had the best chemical composition compared to the Marandu palisadegrass and Xaraes palisadegrass. The management of Brachiaria brizantha cultivars at the lowest residual height (10 cm) affected the nutritional value of these feed grasses. Seasonality also influenced the nutritional value of these forages.

At present, Brazil imports approximately 11 billion liters/year of diesel. With the interruption of the works in the new Petrobras refineries, the projection is that by 2025 this volume will increase to 24.2 billion liters of diesel/year. In this sense, the biodiesel factory Grand Valle Bio Energy Ltda., located in the state of Rio de Janeiro, in conjunction with the FAPERJ makes some investments in technology development for the cultivation and use of microalgae as an alternative raw material in the production of biodiesel. Based on arguments previously said, this work presents the results of the microalgae cultivation Monoraphidium sp. in photobioreactors the pilot plant of the company. The installation with an area of 120 m2 is included with 2 open photobioreactors of type falling film (20 m × 1 m), with a cascade of 18mm and capacity of 4000 L. The lineage cultivated is selected from previous ecophysiological studies that are identified as promising for biodiesel production by having a high potential for the production of lipids. This lineage is maintained at collection of the stock of cultures Laboratory of Green Technologies of the School of Chemistry/ UFRJ. The cultivation was performed in means ASM-1 (Gorham et al., 1964), initial pH 8.0, with aeration and circulation average of 8 hours a day during 19 days. The culture was started with an inoculum of 1 × 107 cel/ml. The lipid production was determined in two phases of growth: on day 4 (exponential phase) and 15 day (stationary phase). For the determination and quantification of lipid content, two different methods were assessed for a sample of biomass, submitted to the same processes the separation and drying. The results showed the methodology of Bligh & Dyer with modifications as the most efficient in extracting lipids. The total lipid content of the biomass Monoraphidium sp. was 30.58%. The growth rate varied between 0.74 ± 0.01 and 0.68 ± 0.02.

Extraction of phenolic compounds from bean seed coats by acetic acid was evaluated and compared to conventional extraction with 80% methanol. Total phenols, flavonoids and free radical scavenging capacity were determined by colorimetric methods. Furthermore, qualitative flavonoid characterization was performed via HPLC diode array. The acetic acid extract contained 73.4 ± 7 mg/g of sample expressed as gallic acid equivalents (GAE), and 41.3 ± 4 mg/g as catechin equivalents (CE). The acetic acid extract (at 1 mg/mL) showed over 89% efficiency to scavenge the 1,1 diphenyl-2-picrylhydrazil (DPPH) radical (EC50 = 6.13 mg extract/mg standard). Gallic acid, delphinidin-3-glycoside, petunidinglycoside, petunidin, malvidin-3-glycoside, myricetin-glycoside, quercitin-glycoside, saponin (β-g), and 3-hydroxy-5, 7-megastigmadien-9-one were identified in both extracts. Additionally, kaempferol-3-glycoside was found in the acetic acid extract. Sequential acetic acid extractions indicated that the antiradical activity was about 90% in DPPH radical scavenging within the first 8 h of extraction, when compared to a 24 h extraction. Introduction: Breast cancer is the leading cause of cancer mortality among women. Some biomarkers and clinical features are used for the diagnosis and prognosis of this tumor, but no prognostic or predictive marker is routinely available specifically for hormone receptor positive tumors. Homocysteine is well known as a risk factor in atherosclerotic vascular diseases, but its participation in cancer biology is still unclear. The aim of this study was to evaluate serum Homocysteine and Cysteine as biomarkers of disease progression in breast tumor. As a secondary objective, the effect of a short course (one month) of hormonal treatment on Homocysteine, Cysteine and DNA methylation levels was also evaluated. Methods: Blood samples, tumor samples and normal adjacent tissue were collected during the initial biopsy (pre-treatment) and after one month of hormonal therapy (post-treatment). Serum Homocysteine and Cysteine were analyzed by HPLC and tissue global DNA methylation was determined by the Methylation-Sensitive Restriction Enzyme (MSRE) technique. Results: Variations in Homocysteine levels were significantly correlated with Disease-Free Survival. Cox proportional risk model demonstrated that nodal status and Homocysteine levels were independent prognostic factors for disease-free survival (DFS). A significant difference was observed between pre-and post-treatment levels of Homocysteine and Cysteine in advanced tumors, suggesting a prognostic role in patients with poor clinical characteristics. Conclusion: Although more studies are needed to confirm these results, our research suggests that Hcy might be used as a prognostic biomarker for breast cancer.

Evaluation of the Eta Simulations Nested in Three Global Climate Models

To provide long-term simulations of climate change at higher resolution, Regional Climate Models (RCMs) are nested in global climate models (GCMs). The objective of this work is to evaluate the Eta RCM simulations driven by three global models, the HadGEM2-ES, BESM, and MIROC5, for the present period, 1961-1990. The RCM domain covers South America, Central America, and Caribbean. These simulations will be used for assessment of climate change projections in the region. Maximum temperatures are generally underestimated in the domain, in particular by MIROC5 driven simulations, in summer and winter seasons. Larger spread among the simulations was found in the minimum temperatures, which showed mixed signs of errors. The spatial correlations of temperature simulations against the CRU observations show better agreement for the MIROC5 driven simulations. The nested simulations underestimate precipitation in large areas over the continent in austral summer, whereas in winter overestimate occurs in southern Amazonia, and underestimate in southern Brazil and eastern coast of Northeast Brazil. The annual cycle of the near-surface temperature is underestimated in all model simulations, in all regions in Brazil, and in most of the year. The temperature and precipitation frequency distributions reveal that the RCM and GCM simulations contain more extreme values than the CRU observations. Evaluations of the climatic extreme indicators show that in general hot days, warm nights, and heat waves are increasing in the period, in agreement with observations. The Eta simulations driven by HadGEM2-ES show wet trends in the period, whereas the Eta driven by BESM and by MIROC5 show trends for drier conditions.
