目的探究布鲁氏菌104M疫苗株A抗原的主要成分,从A抗原中筛选潜在候选蛋白,为布鲁氏菌新型疫苗的研制提供基础。方法本研究利用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳考马斯亮蓝染色、鲎试剂检测脂多糖、脂多糖银染对布鲁氏菌104M疫苗株A抗原的主要成分进行解析,通过与免疫绵羊血清的蛋白免疫印记实验,选取有印迹的部分切胶酶解,利用液相色谱–串联质谱对这部分蛋白进一步解析,通过抗原分析工具VaxiJen和毒力蛋白分析工具VirulentPred,筛选布鲁氏菌A抗原评分0.6以上的毒力相关蛋白作为潜在的候选疫苗。结果布鲁氏菌A抗原是一种淡黄色棉花糖状、质量较轻的物质,主要由蛋白和脂多糖构成,对A抗原的蛋白质组鉴定显示,104M A抗原共有1142种蛋白,能够与绵羊血清反应的蛋白有172种,VaxiJen抗原性评分在0.6以上的有87种蛋白,其中毒力相关蛋白有38种。结论布鲁氏菌104M疫苗株A抗原的主要成分是蛋白质和脂多糖,对筛选到的A抗原中的候选蛋白需要进一步的实验来测试这些蛋白质的免疫原性和保护水平以设计出针对布鲁氏菌病更为有效和安全的疫苗。

目的本研究旨在建立一种基于微滴式数字聚合酶链式反应(ddPCR)的检测方法,能够实现对布鲁氏菌病(布病)患者血液中布鲁氏菌核酸的定量与精准检测。方法以布鲁氏菌Bcsp31基因为靶基因,设计引物与探针并构建pUC57-Bcsp31重组阳性质粒,评价ddPCR方法检测布鲁氏菌的灵敏度、重复性与特异度。此外,收集疑似布病患者血液120份,采用ddPCR方法对样本中布鲁氏菌DNA载量进行定量检测。结果本研究建立的ddPCR检测布鲁氏菌的方法具有很高的灵敏性,最低检出限为100拷贝/μL,并且对布鲁氏菌的检测呈现出良好的特异性与准确性。120份血液样本中,ddPCR方法检出阳性103份,实时荧光定量聚合酶链式反应(qPCR)方法检出阳性52份。105份抗体阳性样本中,ddPCR方法与qPCR方法分别检出阳性93份和51份,阳性率分别为88.57%和49.52%。15份抗体阴性样本中,两种方法分别检测出阳性12份和1份。结论本研究建立了一种基于ddPCR方法的高灵敏度、高特异度的检测布鲁氏菌的方法,能够实现布病患者血液样品中布鲁氏菌核酸的定量与精准检测。尤其对于抗体阴性的疑似患者早期布病筛查具有重要意义。

Research on flexible strain sensors has advanced rapidly in recent years, with particular attention being devoted to two-dimensional(2D) semiconductor materials owing to their exceptional mechanical and electrical properties that are conducive to sophisticated sensing performance. However, resistive strain sensors based on 2D semiconductor materials typically exhibit positive gauge factors(GF), while materials for strain sensors with a negative GF remain elusive. We have identified a trend of reduction in the band gap of the emerging 2D semiconductor material tellurium(Te) under strain in simulations reported in past research,and have observed a negative GF in the Te-based strain sensor. In this study, we combined Te with a flexible polyethylene terephthalate(PET) substrate to manufacture a flexible strain sensor with a significantly negative GF. The results of tests revealed that the Te-based strain sensor achieved an impressive maximum sensitivity of-139.7 within a small range of bending-induced strain(< 1%). Furthermore, it exhibited excellent linearity and good cyclic stability, and was successfully applied to monitor limb movements. The work here verifies the significant potential for the use of Te-based strain sensors in next-generation flexible electronics.

[背景]细菌是地铁室内空气中种类最多、来源最广泛的微生物,其中的病原菌可通过空气在人群中传播,导致健康风险增加。[目的]了解中南某城市地铁站内及车厢内空气细菌污染状况及其菌群分布特征,为制定改善地铁室内空气细菌污染干预措施提供科学依据。[方法]根据客流量差异,选择中南某市三个地铁站及停靠列车车厢进行同步空气采样监测。采用直读法测定温度、湿度、风速、二氧化碳(CO_(2))、细颗粒物(PM_(2.5))和可吸入颗粒物(PM_(10))。按照GB/T 18204.3—2013《公共场所卫生检验方法第3部分:空气微生物》要求,以28.3 L·min^(−1)流量采集空气样本,培养检测细菌总数,对细菌微生物种类进行质谱鉴定,根据国家卫生健康委员会制定的《人间传染的病原微生物目录》区分病原菌和非病原菌。采用Kruskal-Wallis H检验进行不同区域和时段地铁卫生指标的比较分析,采用Bonferroni检验进行两两比较,采用Spearman相关性检验对CO_(2)浓度与细菌总数进行关联性分析。[结果]地铁站内和车厢内空气细菌总数、PM_(2.5)和PM_(10)合格率均为100.0%,温度、风速合格率均为94.4%,CO_(2)合格率为98.6%,湿度合格率为0%。地铁站内和车厢内空气细菌总数的中位数及第25、75百分位数为177(138,262)CFU·m^(−3);不同区域、不同时段的比较分析显示,空气细菌总数站厅高于站台,早高峰高于晚高峰,差异均具有统计学意义(P9分;空气细菌优势菌为藤黄微球菌(52.2%)和人葡萄球菌(9.8%)。在3个地铁站内和车厢内检出表皮葡萄球菌(2.2%)、鲍氏不动杆菌(0.3%)和纹带棒杆菌(0.1%)这3种病原菌共23例(2.6%),检出位置主要分布于晚高峰时的车厢内。[结论]地铁室内不同区域、不同时段空气细菌总数存在差异,且存在一定的条件致病菌感染风险,应加强客流高峰时段及各区域的清洁与消毒。

BACKGROUND： Changes in the cardiac autonomic nerve are considered to be important factors in the mechanisms of heart failure. It is possible to reduce or slow down nerve degeneration and necrosis, provided that patients take effective neuroprotectants during the early stages of heart failure. Moreover, it is possible to relieve the pathological process and reduce the risk of death. OBJECTIVE： To study the effect of growth hormone releasing peptide （GHRP） on cardiac cholinergic nerve fiber density distribution in a rat model of heart failure, and verify whether GHRP can ameliorate denervation. DESIGN, TIME AND SETTING： A randomized controlled study was performed at the Key Laboratory of Anatomy, Harbin Medical University, between June and October 2009. MATERIALS： Fifty adult, healthy, female, Wistar rats, weighing （200± 20） g, were randomly divided into GHRP （n = 30）, model （n = 10）, and sham operation （n = 10） groups. GHRP-2 was made in Shanghai, China （batch No. z071212-03）. METHODS： Acute myocardial infarction was established by ligating the left anterior descending coronary artery in the GHRP and model groups. Five weeks later, myocardial function was detected using color ultrasound electrocardiograph a successful marker of chronic heart failure models Ejection fraction 〈 60% was considered to be However, the left anterior descending coronary artery was not ligated in the sham operation group. The GHRP group was injected with 100 μ g/kg GHRP-2, and the other two groups were injected with the same volume of physiological saline, once per day. Most current studies focus on statistics of the incidence, etiology, and rehabilitative treatment of cerebral palsy. However, very few studies focus on the environmental risk factors for this syndrome In the present study, a total of 202 children with cerebral palsy in the Jiamusi district of China were selected as the case group, and 404 children were randomly selected for the control group. The survey focused on environmental factors during pregnancy, which included indoor pollution as the most dangerous factor （odds ratio = 2.432, 95% confidence interval = 1.172-5.047; P = 0.017）, followed by use of high-radiation mobile phones, more than 5 hours of watching television per day, and cooking with a marble kitchen platform during pregnancy. Results demonstrated that environmental risk factors play a role in the development of cerebral palsy in children.

目的建立实时荧光定量PCR方法检测布鲁氏菌基因组内IS711转座酶基因(orfA基因)的拷贝数,并应用于布鲁氏菌种与生物型的鉴定。方法构建基于Taqman探针法实时荧光定量PCR技术的orfA基因拷贝数的检测体系。设计bcsp31基因和orfA基因的引物和探针,应用实时荧光定量PCR方法同时检测同一菌株相同DNA浓度下的bcsp31基因和orfA基因含量,获得2个基因的循环数(CT值),再根据bcsp31基因和orfA基因CT值的差异,换算出待检测布鲁氏菌菌株基因组内orfA基因的拷贝数。同时,将布鲁氏菌16M菌株DNA进行2倍递减稀释,验证检测体系的稳定性。结果当16M菌株DNA浓度相差2倍时,实时荧光定量PCR方法同时检测bcsp31基因和orfA基因,测得的CT值差值均值均为1.00,95%置信区间为0.95~1.05,标准差为0.17,变异系数为0.17。应用该检测体系检测30株布鲁氏菌菌株DNA中orfA基因的拷贝数,发现羊种生物1~3型菌株分别有6、9和7个拷贝数;猪种生物2型菌株有10个拷贝数,与其他4个猪种生物型(1、3~5)菌株拷贝数不同;绵羊附睾种菌株有37个拷贝数;8个牛种生物型(1~7、9)菌株拷贝数稳定在5~6个。结论成功建立了一种检测布鲁氏菌orfA基因拷贝数的实时荧光定量PCR方法,该方法能够鉴定部分布鲁氏菌种与生物型菌株。
