Distinct immune escape and microenvironment between RG-like and pri-OPC-like glioma revealed by single-cell RNA-seq analysis

The association of neurogenesis and gliogenesis with glioma remains *** conducting single-cell RNA-seq analyses on 26 gliomas,we reported their classification into primitive oligodendrocyte precursor cell(pri-OPC)-like and radial glia(RG)-like tumors and validated it in a public cohort and TCGA *** RG-like tumors exhibited wild-type isocitrate dehydrogenase and tended to carry EGFR mutations,and the pri-OPC-like ones were prone to carrying TP53 *** subclones only in pri-OPC-like tumors showed substantially down-regulated MHC-I genes,suggesting their distinct immune evasion ***,the two subgroups appeared to extensively modulate glioma-infiltrating lymphocytes in distinct *** specific genes not expressed in normal immune cells were found in glioma-infiltrating *** example,glial/glioma stem cell markers OLIG1/PTPRZ1 and B cell-specific receptors IGLC2/IGKC were expressed in pri-OPC-like and RG-like glioma-infiltrating lymphocytes,*** expression was positively correlated with those of immune checkpoint genes(e.g.,LGALS33)and poor survivals as validated by the increased expression of LGALS3 upon IGKC overexpression in Jurkat *** finding indicated a potential inhibitory role in tumor-infiltrating lymphocytes and could provide a new way of cancer immune evasion.

[目的/意义]作物农艺性状与形态结构表型智能识别是作物智慧育种的主要内容,是研究“基因型—环境型—表型”相互作用关系的基础,对现代作物育种具有重要意义。[进展]大规模、高通量作物表型获取设备是作物表型获取、分析、测量、识别等的基础和重要手段。本文介绍了高通量作物表型主流平台和感知成像设备的功能、性能以及应用场景。分析了作物株高获取、作物器官检测与技术等农艺性状智能识别和作物株型识别、作物形态信息测量以及作物三维重建等形态结构智能识别技术的研究进展及挑战。[结论/展望]从研制新型低成本田间智能作物表型获取与分析装备、提升作物表型获取田间环境的标准化与一致性水平、强化田间作物表型智能识别模型的通用性,研究多视角、多模态、多点连续分析与时空特征融合的作物表型识别方法,以及提高模型解释性等方面,展望了作物表型技术主要发展方向。

目的:探究大鼠视网膜中对N-甲基-D天冬氨酸(NMDA)敏感的无长突细胞(AC)亚型。方法:H-E染色观察NMDA损伤后视网膜组织病理变化,免疫荧光染色观察大鼠视网膜中神经节细胞标志物(Brn3a)、AC标志物(syntaxin)阳性细胞数量变化;在NMDA致AC损伤的剂量-效应变化关系和时间-效应变化关系的研究中用免疫荧光染色检测大鼠视网膜中GABA能AC标志物(GAD65/67)、甘氨酸能AC标志物(GlyT1)、胆碱能AC标志物(ChAT)、多巴胺能AC标志物(TH)、AⅡAC标志物(PV)阳性细胞的形态和数目变化;利用碘化丙啶(PI)和cleaved casapse-3染色确定死亡的AC亚型。结果:NMDA可损伤视网膜节细胞层(GCL)中的神经节细胞和移位的AC以及内核层(INL)中的AC。在NMDA致AC损伤的剂量-效应关系变化中,所有AC均随着NMDA剂量的增加而显著减少,且在100 nmol NMDA中未见胞体位于INL中的胆碱能AC和多巴胺能AC(DAC)的胞体;在NMDA致AC损伤的时间-效应关系变化中,INL中的GAD65/67^(+)、GlyT1^(+)、ChAT^(+)、TH^(+)细胞的数量均在损伤后6 h开始减少,GCL中的GAD65/67^(+)细胞、视网膜中GAD65/67^(+)细胞总数以及PV+细胞均在损伤后12 h开始减少,且均随着NMDA损伤时间的推移而减少。结论:视网膜GABA能AC和甘氨酸能AC均在NMDA作用下出现损伤,其中以INL中的胆碱能AC和DAC对NMDA损伤敏感。

Fenofibrate,a peroxisome proliferator-activated receptor α(PPARα)agonist,is widely prescribed for hyperlipidemia *** studies also showed that it has therapeutic potential in various liver ***,its effects on hepatomegaly and liver regeneration and the involved mechanisms remain ***,the study showed that fenofibrate significantly promoted liver enlargement and regeneration post-partial hepatectomy in mice,which was dependent on hepatocyteexpressed PPARα.Yes-associated protein(YAP)is pivotal in manipulating liver growth and *** further identified that fenofibrate activated YAP signaling by suppressing its K48-linked ubiquitination,promoting its K63-linked ubiquitination,and enhancing the interaction and transcriptional activity of the YAP-TEAD *** inhibition of YAP-TEAD interaction using verteporfin or suppression of YAP using AAV Yap shRNA in mice significantly attenuated fenofibrate-induced *** factors,such as MYC,KRT23,RAS,and RHOA,might also participate in fenofibrate-promoted hepatomegaly and liver *** studies demonstrate that fenofibratepromoted liver enlargement and regeneration are PPARα-dependent and partially through activating the YAP signaling,with clinical implications of fenofibrate as a novel therapeutic agent for promoting liver regeneration.

Anthocyanins,ubiquitous in the Compositae family,are regulated by MYB(v-myb avian myeloblastosis viral oncogene homolog),playing an important role in anthocyanin *** this study,we analyzed the regulation pathway in which the MYB protein of subgroup 6 promotes dihydroflavonol reductase(DFR)expression in Compositae,and validated this law in Saussurea medusa through yeast one-hybrid *** results showed that MYB and DFR underwent purification selection,DFR promoter analysis revealed the presence of MYB binding site(GAGTTGAATGG)and bHLH binding site(CANNTG)at the sense strand of 84–116 nucleotide residues from the start *** two motifs were separated by 9–10 nucleotide residues,as existed in the DFR promoters of many Compositae ***,the yeast one-hybrid experiment demonstrated that SmMYB1 can activate the promoter of *** results provide a reference for further functional study of DFR in Compositae.

ShCTR1 Interacts with ShRBOH1 to Positively Regulate Aerenchyma Formation in Saussurea inversa through ROS Mediation

Saussurea inversa is one of the Tibetan medicine with developed *** this study,we investigated the function of the ShCTR1 gene related to the formation of *** study of the occurrence and formation of aerenchyma in *** has certain theoretical significance in revealing the special mechanism of alpine plants adapting to the *** results of yeast two-hybrid experiments showed that *** ShCTR1 interacted with the ShRBOH1,which is a key producer of reactive oxygen species(ROS),and it was hypothesized that ShCTR1 regulation of aerenchyma formation in *** mainly mediated by *** induction and inhibition experiments were carried out on *** seedlings,it was shown that ROS inducer(AT)and Ethene(ETH)induced the formation of aerenchyma in *** well in roots,stems,and leaves,and Ethylene inhibitor(1-MCP)and ROS inhibitor DPI(diphenylene iodonium)inhibited the formation of aerenchyma of *** in different degrees,which proved that ROS and ethylene(ET)were positively correlated with the formation of ***,AT and ETH well induced the expression of the ShCTR1 gene,and 1-MCP and DPI inhibited the expression of the ShCTR1 gene to different degrees,which proved that ROS and ET were also positively correlated with the expression of the ShCTR1 ***1 was transferred into tobacco(Nicotiana tabacum L.),then repeat the above *** was shown that positive rate and porosity were lowest and highest after DPI and AT treatments,respectively,it showed a positive correlation,which proved that the ShCTR1 gene was positively regulating the formation of *** of the ShCTR1 gene increased the content of ROS,consequently,the regulation of aerenchyma formation by the ShCTR1 gene is mainly mediated by *** suggested that the formation of aerenchyma in *** was related to hypoxia in the alpine environment,and ShCTR1 interacts with ShRBOH1 to positively regulate the formation of a

Nitrate(NO_(3)^(−))electroreduction reaction(NO_(3)^(−)RR)provides an attractive and sustainable route for NO_(3)^(−)pollution mitigation or energy-saved ammonia(NH3)*** this work,high-quality B and Fe co-doped Co_(2) P hollow nanocubes(B/Fe-Co_(2) P HNCs)are successfully synthesized though simultaneous boronation-phosphorization treatment,which reveal outstanding selectivity,activity,stability for the NO_(3)^(−)to NH_(3) conversion in neutral electrolyte because of big surface area,fast mass transport,superhydrophilic surface,and optimized electronic structure.B/Fe-Co_(2) P HNCs can achieve the high NH3 yield rate(22.67 mg h^(−1) mg_(cat)^(−1))as well as Faradaic efficiency(97.54%)for NO_(3)^(−)RR,greatly outperforming most of non-precious metal based NO_(3)^(−)RR electrocatalysts.

A Scoping Review of Real-World Study in Vaccine Evaluation

Real-world study(RWS)gained prominence starting with a significant investigation on ramipril’s impact on hypertension in 1993(1).A key advancement for the Food and Drug Administration(FDA)occurred with the enactment of the 21st Century Cures Act in 2016,emphasizing RWS(2).The FDA’s Real-world Evidence Program establishes real-world data(RWD)as“data relating to patient health status and/or the delivery of health care routinely collected from a variety of sources”,while real-world evidence(RWE)is“clinical evidence about the usage and potential benefits or risks of a medical product derived from analysis of RWD”(3).The proliferation of big data,electronic medical records(EMR),electronic health records(EHR),and medical claims data offers vast information resources facilitating *** access to RWD allows for worldwide monitoring of the impacts of various public health initiatives like vaccination *** randomized clinical trials(RCTs)traditionally gauge vaccine efficacy and shortterm safety,RWS can assess vaccine performance and safety across larger,more diverse populations and are adept at identifying rare events not easily discernible in RCTs due to their infrequency.

A synergetic strategy of NIR-Ⅱ squaraine dyes with ultrahigh photothermal conversion efficiency for photothermal therapy

Recent research on photothermal therapy(PTT) has sparked significant interest in the development of new organic photothermal agents(PTAs),ranging from single-molecule to aggregated ***,controlling aggregation pathways for PTAs with ultrahigh photothermal conversion efficiency(PCE) remains a major ***,a two-pronged approach utilizing “Haggregation” and “intramolecular motion” was employed to enhance the PCE of an acceptor-substituted squaraine dye(NSQs).The C2vmolecular symmetry of the NSQs,which possess a ground state dipole moment(μg),promotes H-dimeric aggregates through dipole-dipole *** triphenylethylene or diphenylamine groups were added to this H-dimeric *** was done to enhance intramolecular motions for heat generation and also to extend conjugation,which redshifted the optical absorption and balanced the blue-shift induced by *** this technique,an organic PTA with NIR-II absorption was developed,and its nanoparticle achieved a remarkable PCE of 86.3% under 1,064 nm laser *** transient absorption spectroscopy and quantum mechanical calculations demonstrated the accelerated internal conversion process in NIR-II PTAs for rapid heat *** NSQs nanoparticles exhibit superior photothermal therapeutic properties for in vivo photoacoustic imaging-guided PTT,demonstrating the potential of bottom-up design to enable synergistic engineering strategies towards efficient phototheranostic agents.
