Objective: To investigate the effect of FTY720 on lymphocytes proliferation,differentiation and apoptosis of in vitro mixed lymphocyte reaction (MLR). Methods: Different concentrations of FTY720 were added to the primary MLR containing the splenocytes from BALB/c and C57BL/6J *** proliferative status,cell phenotype and apoptosis of the lymphocytes of MLR were measured by MTT assay and flow ***: FTY720 at concentration of 400 ng/mL could significantly suppress lymphocytes proliferation (P<0.05) with the inhibition rate of47.6% ,and that was 51.6% at concentration of 3200 ng/*** a dose-dependent *** concentration of l600 ng/mL,the percentage of CD4+CD25+T cell in the MLR increased significantly (P<0.05).The percentage of apoptosis also rose significantly in the presence of FTY720 at concentration of 200 ng/mL (P <0.05). Conclusion: FTY720 at high concentration significantly suppresses the lymphocytes proliferation in MLR;increases the percentage of regulatory T lymphocytes;and facilitates lymphocytes apoptosis.
