The measurement of amino acid and monoamine neurotransmitters（NTs） in serum plays a central role in the identification and monitoring of clinical panic disorder（PD）. However, the strong polarity and chemical heterogeneity of NTs make direct extraction and rapid detection extremely challenging. Herein, we developed a strategy that integrated solid phase extraction based on magnetic Fe3O4coated with polydopamine（Fe3O4@PDA） and direct analysis of mass spectrometry in real time（DART-MS） for quantification of 10 NTs. We systematically investigated the optimal conditions of this strategy to achieve good accuracy and precision. Compared with existing magnetic solid phase extraction methods for NTs detection in biological samples, this strategy could adsorb more targets while having similar adsorption efficiencies, and the total process time was reduced by an average of 69.42%. We rapidly determined the metabolic abnormality of 6 types of NTs in the serum of PD patients and demonstrated their diagnostic accuracy. In addition, an outstanding feature of PD in which tryptophan was more metabolized by the KYN pathway and the 5-hydroxytryptamine pathway was inhibited could be elucidated by this strategy. Our results highlighted the potential ability of Fe3O4@PDA coupled with DART-MS to assist in the clinical diagnosis of PD.

Microglial activation that occurs rapidly after closed head injury may play important and complex roles in neuroinflammation-associated neuronal damage and *** previously reported that induced neural stem cells can modulate the behavior of activated microglia via CXCL12/CXCR4 signaling,influencing their activation such that they can promote neurological ***,the mechanism of CXCR4 upregulation in induced neural stem cells remains *** this study,we found that nuclear factor-κB activation induced by closed head injury mouse serum in microglia promoted CXCL12 and tumor necrosis factor-αexpression but suppressed insulin-like growth factor-1 ***,recombinant complement receptor 2-conjugated Crry(CR2-Crry)reduced the effects of closed head injury mouse serum-induced nuclear factor-κB activation in microglia and the levels of activated microglia,CXCL12,and tumor necrosis factor-α.Additionally,we observed that,in response to stimulation(including stimulation by CXCL12 secreted by activated microglia),CXCR4 and Crry levels can be upregulated in induced neural stem cells via the interplay among CXCL12/CXCR4,Crry,and Akt signaling to modulate microglial *** agreement with these in vitro experimental results,we found that Akt activation enhanced the immunoregulatory effects of induced neural stem cell grafts on microglial activation,leading to the promotion of neurological recovery via insulin-like growth factor-1 secretion and the neuroprotective effects of induced neural stem cell grafts through CXCR4 and Crry upregulation in the injured cortices of closed head injury ***,these beneficial effects of Akt activation in induced neural stem cells were positively correlated with the therapeutic effects of induced neural stem cells on neuronal injury,cerebral edema,and neurological disorders post–closed head *** conclusion,our findings reveal that Akt activation may enhance the immunoregulatory effects of induced neura

High-energy-density lithium(Li)–air cells have been considered a promising energy-storage system,but the liquid electrolyte-related safety and side-reaction problems seriously hinder their *** address these above issues,solid-state Li–air batteries have been widely ***,many commonly-used solid electrolytes generally face huge interface impedance inLi–air cells and also showpoor stability towards ambient air/Li ***,we fabricate a differentiating surface-regulated ceramic-based composite electrolyte(DSCCE)by constructing disparately LiI-containing polymethyl methacrylate(PMMA)coating and Poly(vinylidene fluoride-co-hexafluoropropylene)(PVDF-HFP)layer on both sides of Li_(1.5)Al_(0.5)Ge_(1.5)(PO_(4))_(3)(LAGP).The cathode-friendly LiI/PMMA layer displays excellent stability towards superoxide intermediates and also greatly reduces the decomposition voltage of discharge products in Li–air ***,the anode-friendly PVDF-HFP coating shows low-resistance properties towards ***,Li dendrite/passivation derived from liquid electrolyte-induced side reactions and air/I-attacking can be obviously suppressed by the uniformand compact composite *** a result,the DSCCE-based Li–air batteries possess high capacity/low voltage polarization(11,836mAh g^(-1)/1.45Vunder 500mAg^(-1)),good rate performance(capacity ratio under 1000mAg^(-1)/250mAg^(-1) is 68.2%)and longterm stable cell operation(~300 cycles at 750 mA g^(-1) with 750 mAh g^(-1))in ambient air.

Gradient magnetic heterointerfaces have injected infinite vitality in optimizing impedance matching,adjusting dielectric/magnetic resonance and promoting electromagnetic(EM)wave absorption,but still exist a significant challenging in regulating local phase ***,accordion-shaped Co/Co_(3)O_(4)@N-doped carbon nanosheets(Co/Co_(3)O_(4)@NC)with gradient magnetic heterointerfaces have been fabricated via the cooperative high-temperature carbonization and lowtemperature oxidation *** results indicate that the surface epitaxial growth of crystal Co_(3)O_(4) domains on local Co nanoparticles realizes the adjustment of magnetic-heteroatomic components,which are beneficial for optimizing impedance matching and interfacial ***,gradient magnetic heterointerfaces simultaneously realize magnetic coupling,and long-range magnetic ***,the synthesized Co/Co_(3)O_(4)@NC absorbents display the strong electromagnetic wave attenuation capability of−53.5 dB at a thickness of 3.0 mm with an effective absorption bandwidth of 5.36 GHz,both are superior to those of single magnetic domains embedded in carbon *** design concept provides us an inspiration in optimizing interfacial polarization,regulating magnetic coupling and promoting electromagnetic wave absorption.

Sr-Nd isotopes are widely utilized as reliable tracers in sediment provenance *** this study,we compiled 304Sr and 335 Nd isotope data of fine-grained surface sediments collected from major rivers in eastern China,including the Huanghe River,Changjiang River,southeastern coastal rivers,and rivers in Taiwan Island,as well as from marginal seas （Yellow Sea and East China Sea） and the Okinawa *** comprehensive dataset allowed us to examine the spatial distribution patterns of these isotopes and their implications for provenance *** variations of εNd values in the sediments in Changjiang and Huanghe Basins primarily reflect the diversity of sediment sources,whereas the87Sr/86Sr ratios are influenced by both grain size and source ***,the reduction in87Sr/86Sr ratios observed in the middle and lower Changjiang River is attributed to the inputs of sources characterized by distinct87Sr/86Sr signatures beyond mere grain size *** the southeastern coastal and Taiwan rivers,sediments in the tidal reaches of the Jiao River exhibit hybrid characteristics,a mix of Changjiang-derived sediment and those originating from local ***,the Min River and Jiulong River sediments predominantly reflect local sources,with minimal influence from the Changjiang *** significant isotopic variation of Sr-Nd isotopic characteristics observed between riverine sediments on the eastern and western sides of Taiwan Island underscores the diverse provenance heterogeneity of source rocks in the *** the marginal seas,our data indicates that the surface sediments in the central mud area of the South Yellow Sea and the southern Yellow Sea are predominantly sourced from the Changjiang River catchment,potentially augmented by inputs from Korean *** inner shelf sediments of the East China Sea are primarily influenced by Changjiang River inputs,whereas rivers in eastern Taiwan Island contribute to the sediments on the middle and

目的将已构建的具有抗慢性粒细胞性白血病(CML)融合基因bcr/abl mRNA的含核酶载体,进行细胞内外实验,了解抗bcr/abl mRNA核酶的切割效应。方法含有抗靶基因的真核表达载体pAVGS4经酶切后,其中抗CML核酶M1-GS RNA的DNA序列被构建到pUC19上得到pGS210,并经酶切和测序鉴定;合成bcr/abl融合位点前后的一段靶序列,克隆到pGEM-7zf(+)上得到p bcr-abl经测序鉴定,将pGS210和p bcr-abl进行体外转录,获得 M1-GS RNA及其作用底物,将两者混合、孵育,通过放射自显影术检测切割的效果;将pAVGS4转染到CML细胞株K652细胞内,通过RT-PCR方法了解核酶对于目标 RNA的作用效应。结果 p bcr-abl经酶切电泳及测序证实载体中含有目标序列,Ml-GS RNA作用于底物后,放射自显影术检测显示底物 RNA被有效切割。pAVGS4转染到K562细胞48和72h后,RT-PCR显示bcr/abl mRNA被有效切割。结论 pAVGS4可以有效地切割K562细胞内的bcr/abl mRNA,预期可作为CML分子靶向治疗的工具。

Bulging Performance and Quality Control of Aluminum Alloy Tailor-welded Overlapping Sheets Based on Interface Friction

In order to solve the problem of poor formability caused by different materials and properties in the process of tailor-welded sheets forming,a forming method was proposed to change the stress state of tailor-welded sheets by covering the tailor-welded sheets with better plastic properties overlapping *** the same time,the interface friction effect between the overlapping and tailor-welded sheets was utilized to control the stress magnitude and further improve the formability and quality of the tailor-welded *** this work,the bulging process of the tailor-welded overlapping sheets was taken as the research *** alloy tailor-welded overlapping sheets bulging specimens were studied by a combination of finite element analysis and experimental *** results show that the appropriate use of interface friction between tailor-welded and overlapping sheets can improve the formability of tailor-welded sheets and control the flow of weld seam to improve the forming *** increasing the interface friction coefficient on the side of tailor-welded sheets with higher strength and decreasing that on the side of tailor-welded sheets with lower strength,the deformation of the tailor-welded sheets are more uniform,the offset of the weld seam is minimal,the limit bulging height is maximal,and the forming quality is optimal.

In this work, we provide a comprehensive review on the formation, evolution, properties, and effects of supercritical geofluid. In Earth's interior, enhanced miscibility between H2O and silicate by the addition of special components or by the increase of pressure and temperature gives rise to supercritical geofluid with a significant amount of both H2O and silicate *** formation of supercritical geofluid in magmatic-hydrothermal systems, typified by pegmatite system, is governed by meltfluid critical curve. The formation of supercritical geofluid in metamorphic systems, typified by subducted slab, is governed by the second critical end point. Experimental results suggest that the presence of boron and fluorine in pegmatite system makes it possible to form supercritical geofluid at crustal depths, but the release of supercritical geofluid from subducted slab is withheld until almost 100 km depth. A major presence of both H2O and depolymerized structural units（monomers, dimers, etc.） endows supercritical geofluid with unique physical properties including low density, low elastic moduli, low viscosity, high diffusivity,and high electrical conductivity. Supercritical geofluid can effectively mobilize a variety of elements even including high field strength elements and heavy rare earth elements. The chemical signatures of supercritical geofluid can be inherited by metasomatized mantle and mantle-derived melts, and this could give an explanation of the oxidation of arc magmas. Phase separation of supercritical geofluid through the mechanism of spinodal decomposition leads to formation of a melt network. Multiphase fluid inclusions recovered from subduction zone rocks and pegmatites are possible relics of supercritical geofluid. Supercritical geofluid can cause electrical anomaly and low seismic velocity near the top of subducted slab, and can be linked with intermediate-focus earthquakes.
