Small Interfering RNA Targeting MDR1 Inhibits Ovarian Cancer Growth and Increases Efficacy of Chemotherapy in vivo

Objective： To further validate a knockdown approach for circumventing the multidrug resistance gene （MDR1）, we used small interfering RNA（siRNA） targeting MDR1 gene to inhibit the expression of MDR1 gene and P-glycoprotein（P-gp） in vivo. Methods： Ascite tumor xenografts were established by implanting human ovarian carcinoma cells SKOV3/AR intraperitoneally into the nude mice. The mice were randomized into the following three treatment groups with each group six mice respectively： Taxol, Taxol with lipofectamine and Taxol with siRNA/MDR1- lipofectamine intraperitoneal injection. The tumor growth rate and the ascite growth rate of mice were investigated. The expressions of MDR1 gene and P-gp in mice were determined by reverse transcription-polymerase chain reaction（RT-PCR） and immunohistochemistry respctively. Results： The growth of tumors and ascites in mice treated with Taxol and siRNA/MDR1- lipofectamine was significantly inhibited compared with those in mice of other groups. After 28 days＇ treatment, the average tumor weight and ascite volume decreased by 43.6% and 29.7% in the group treated with Taxol and siRNA/MDRl-lipofectamine compared with these treated with Taxol alone （P〈0.001）. The expressions of MDR1 gene and P-gp in the group treated with Taxol and siRNA/MDRl-lipofectamine were also decreased compared with those in the group treated with Taxol alone （P〈0.001）. Conclusion： Small interfering RNA targeting-MDR1 can effectively and specifically suppress the expression of MDRl（P-glycoprotein） and inhibit ovarian cancer growth in vivo.

Dys-psychological Stress Effect on Expressions of P53 and NFκBp65 in Human Ovarian Carcinoma In Vivo

Objective： To investigate the dys-psychological stress effect on the growth of subcutaneous xenotransplanted tumor in nude mice bearing human epithelium ovarian carcinoma, and the influence on P53 and NFκBp65 expressions. Methods： The subcutaneous tumor xenografts were established by implanting human epithelium ovarian carcinoma tissues into nude mice and the dys-psychological stress model was established with restraint. The mice were randomized into the following four treatment groups with each group six mice respectively： tumor group （group A）, normal saline intraperitoneal injection; tumor with stress group （group B）, normal saline intraperitoneal injection; tumor therapy group （group C）, cisplatin intraperitoneal injection; and tumor therapy with stress group （group D）, cisplatin intraperitoneal injection. The expressions of P53 and NFκBp65 in tumor tissues were determined by Western blotting. Results： The expressions of P53 and NFκBp65 in each restraint group were enhanced compared with the control groups （P0.05）. Conclusion： The dys-psychological stress may induce the high expressions of P53 and NFκBp65 proteins and further promote tumor growth.

Effect of WFDC 2 silencing on the proliferation,motility and invasion of human serous ovarian cancer cells in vitro

Objective:To investigate effect and possible mechanisms of silencing human WFDC2（HE4） gene on biological behavior changes as cell proliferation,apoplosis,movement and invasion of human serous ovarian cancer cell line ***:Lentiviral WFDC2 gene sequence of small interfering siRNA was stablely transfected into SKOV3 identified by Q-PCR and western-blot. Obtained SKOV3 stable strains with silenced HE4 were measured by proliferation,apoplosis, migration,and ***:Gene sequencing showed that the oligonucleotides were successfully inserted into the expected *** silencing HE4 in the SKOV3,proliferation was significandy inhibited（P0/G1 phase was arrested by the cell cycle（P<0.01） and capacity of the migration and invasion decreased significandy（P<0.01）.Slight early apoptosis ratio and no change of late apoplosis were found without change of Caspase-3 or Bcl-2 *** involed in ERK pathway as phosphorylated protein as p-EGFR,p- ERK decreased and protease protein involved in tissue remoding as matrix metalloproteinases MMP-9,MMP-2 and cathepsin B decreased compared with control ***:HE4 gene plays an important role in regulating proliferation,apoptosis,migration,invasion of serous ovarian cancer cells by ERK pathway and protease *** role in apoptosis needs to be further explored,and it may be a potential target for serous ovarian cancer.

Background： Ambient aerosol fine particulate matter （PM2.5） is associated with male reproductive toxicity in experiments and may have adverse effects in the female. However, studies evaluating the protective effects and precise mechanisms of aspirin, Vitamin C, Vitamin E, or ozone against toxic effects of PM2.5 are sparse. This study was conducted to investigate the possible protective effects and mechanisms of aspirin, Vitamin C, Vitamin E, or ozone on fertility in female mice treated with PM2.5, Methods： Eighty-four ICR mice were divided into six groups： control group, PM2.5 group, PM2.5 ＋ aspirin group, PM2.5 ＋ Vitamin C group, PM2.5 ＋ Vitamin E group, and PM2.5 ＋ ozone group. PM2.5 was given by intratracheal instillation every 2 days for 3 weeks. Aspirin, Vitamin C, and Vitamin E were given once a day by oral gavage for 3 weeks, and ozone was administered by intraperitoneal injection once a day for 3 weeks. The levels ofanti-Mullerian honnone （AMH）, interleukin-6 （IL-6）, tumor necrosis factor-alpha （TNF-α）, and 8-hydroxy-2＇-deoxyguanosine （8-OHdG） were measured using enzyme-linked immunosorbent assay. Western blotting analysis was used to analyze the expressions ofBcl-2, Bax, and caspase-3 in ovaries. Changes in histological structure were examined by light microscope and electron microscopy was used to detect ultramicrostructure. Results： The results demonstrated that PM2.5 decreased AMH levels （P 〈 0.001）; however, aspirin （P 〈 0.001）, Vitamin C （P 〈 0.001）, Vitamin E （P = 0.001）, and ozone （P = 0.002） alleviated the decrease. Changes of IL-6, TNF-α, 8-OHdG, Bax/Bcl-2, and caspase-3 in PM2.5 group were increased compared to control group （P 〈 0.001）, while in PM2.5 ＋ aspirin, PM2.5 ＋ Vitamin C, PM2.5 ＋ Vitamin E, and PM2.5 ＋ ozone groups, they were statistically decreased compared to PM2.5 group （P 〈 0.001 or P 〈 0.05）. Induced neural stem cells regulate microglial activation through Akt-mediated upregulation of CXCR4 and Crry in a mouse model of closed head injury

Microglial activation that occurs rapidly after closed head injury may play important and complex roles in neuroinflammation-associated neuronal damage and *** previously reported that induced neural stem cells can modulate the behavior of activated microglia via CXCL12/CXCR4 signaling,influencing their activation such that they can promote neurological ***,the mechanism of CXCR4 upregulation in induced neural stem cells remains *** this study,we found that nuclear factor-κB activation induced by closed head injury mouse serum in microglia promoted CXCL12 and tumor necrosis factor-αexpression but suppressed insulin-like growth factor-1 ***,recombinant complement receptor 2-conjugated Crry(CR2-Crry)reduced the effects of closed head injury mouse serum-induced nuclear factor-κB activation in microglia and the levels of activated microglia,CXCL12,and tumor necrosis factor-α.Additionally,we observed that,in response to stimulation(including stimulation by CXCL12 secreted by activated microglia),CXCR4 and Crry levels can be upregulated in induced neural stem cells via the interplay among CXCL12/CXCR4,Crry,and Akt signaling to modulate microglial *** agreement with these in vitro experimental results,we found that Akt activation enhanced the immunoregulatory effects of induced neural stem cell grafts on microglial activation,leading to the promotion of neurological recovery via insulin-like growth factor-1 secretion and the neuroprotective effects of induced neural stem cell grafts through CXCR4 and Crry upregulation in the injured cortices of closed head injury ***,these beneficial effects of Akt activation in induced neural stem cells were positively correlated with the therapeutic effects of induced neural stem cells on neuronal injury,cerebral edema,and neurological disorders post–closed head *** conclusion,our findings reveal that Akt activation may enhance the immunoregulatory effects of induced neura

Small extracellular vesicles derived from human induced pluripotent stem cell-differentiated neural progenitor cells mitigate retinal ganglion cell degeneration in a mouse model of optic nerve injury

Several studies have found that transplantation of neural progenitor cells(NPCs)promotes the survival of injured ***,a poor integration rate and high risk of tumorigenicity after cell transplantation limits their clinical *** extracellular vesicles(sEVs)contain bioactive molecules for neuronal protection and *** studies have shown that stem/progenitor cell-derived sEVs can promote neuronal survival and recovery of neurological function in neurodegenerative eye diseases and other eye *** this study,we intravitreally transplanted sEVs derived from human induced pluripotent stem cells(hiPSCs)and hiPSCs-differentiated NPCs(hiPSC-NPC)in a mouse model of optic nerve *** results show that these intravitreally injected sEVs were ingested by retinal cells,especially those localized in the ganglion cell *** with hiPSC-NPC-derived sEVs mitigated optic nerve crush-induced retinal ganglion cell degeneration,and regulated the retinal microenvironment by inhibiting excessive activation of *** analysis further revealed that hiPSC-NPC derived sEVs transported neuroprotective and anti-inflammatory miRNA cargos to target cells,which had protective effects on RGCs after optic nerve *** findings suggest that sEVs derived from hiPSC-NPC are a promising cell-free therapeutic strategy for optic neuropathy.

High-energy-density lithium(Li)–air cells have been considered a promising energy-storage system,but the liquid electrolyte-related safety and side-reaction problems seriously hinder their *** address these above issues,solid-state Li–air batteries have been widely ***,many commonly-used solid electrolytes generally face huge interface impedance inLi–air cells and also showpoor stability towards ambient air/Li ***,we fabricate a differentiating surface-regulated ceramic-based composite electrolyte(DSCCE)by constructing disparately LiI-containing polymethyl methacrylate(PMMA)coating and Poly(vinylidene fluoride-co-hexafluoropropylene)(PVDF-HFP)layer on both sides of Li_(1.5)Al_(0.5)Ge_(1.5)(PO_(4))_(3)(LAGP).The cathode-friendly LiI/PMMA layer displays excellent stability towards superoxide intermediates and also greatly reduces the decomposition voltage of discharge products in Li–air ***,the anode-friendly PVDF-HFP coating shows low-resistance properties towards ***,Li dendrite/passivation derived from liquid electrolyte-induced side reactions and air/I-attacking can be obviously suppressed by the uniformand compact composite *** a result,the DSCCE-based Li–air batteries possess high capacity/low voltage polarization(11,836mAh g^(-1)/1.45Vunder 500mAg^(-1)),good rate performance(capacity ratio under 1000mAg^(-1)/250mAg^(-1) is 68.2%)and longterm stable cell operation(~300 cycles at 750 mA g^(-1) with 750 mAh g^(-1))in ambient air.

Low‑Temperature Oxidation Induced Phase Evolution with Gradient Magnetic Heterointerfaces for Superior Electromagnetic Wave Absorption

Gradient magnetic heterointerfaces have injected infinite vitality in optimizing impedance matching,adjusting dielectric/magnetic resonance and promoting electromagnetic(EM)wave absorption,but still exist a significant challenging in regulating local phase ***,accordion-shaped Co/Co_(3)O_(4)@N-doped carbon nanosheets(Co/Co_(3)O_(4)@NC)with gradient magnetic heterointerfaces have been fabricated via the cooperative high-temperature carbonization and lowtemperature oxidation *** results indicate that the surface epitaxial growth of crystal Co_(3)O_(4) domains on local Co nanoparticles realizes the adjustment of magnetic-heteroatomic components,which are beneficial for optimizing impedance matching and interfacial ***,gradient magnetic heterointerfaces simultaneously realize magnetic coupling,and long-range magnetic ***,the synthesized Co/Co_(3)O_(4)@NC absorbents display the strong electromagnetic wave attenuation capability of−53.5 dB at a thickness of 3.0 mm with an effective absorption bandwidth of 5.36 GHz,both are superior to those of single magnetic domains embedded in carbon *** design concept provides us an inspiration in optimizing interfacial polarization,regulating magnetic coupling and promoting electromagnetic wave absorption.

Sr-Nd isotopic characteristics and their significance in provenance tracing of detrital sediment in major rivers and marginal seas of eastern China

Sr-Nd isotopes are widely utilized as reliable tracers in sediment provenance *** this study,we compiled 304Sr and 335 Nd isotope data of fine-grained surface sediments collected from major rivers in eastern China,including the Huanghe River,Changjiang River,southeastern coastal rivers,and rivers in Taiwan Island,as well as from marginal seas （Yellow Sea and East China Sea） and the Okinawa *** comprehensive dataset allowed us to examine the spatial distribution patterns of these isotopes and their implications for provenance *** variations of εNd values in the sediments in Changjiang and Huanghe Basins primarily reflect the diversity of sediment sources,whereas the87Sr/86Sr ratios are influenced by both grain size and source ***,the reduction in87Sr/86Sr ratios observed in the middle and lower Changjiang River is attributed to the inputs of sources characterized by distinct87Sr/86Sr signatures beyond mere grain size *** the southeastern coastal and Taiwan rivers,sediments in the tidal reaches of the Jiao River exhibit hybrid characteristics,a mix of Changjiang-derived sediment and those originating from local ***,the Min River and Jiulong River sediments predominantly reflect local sources,with minimal influence from the Changjiang *** significant isotopic variation of Sr-Nd isotopic characteristics observed between riverine sediments on the eastern and western sides of Taiwan Island underscores the diverse provenance heterogeneity of source rocks in the *** the marginal seas,our data indicates that the surface sediments in the central mud area of the South Yellow Sea and the southern Yellow Sea are predominantly sourced from the Changjiang River catchment,potentially augmented by inputs from Korean *** inner shelf sediments of the East China Sea are primarily influenced by Changjiang River inputs,whereas rivers in eastern Taiwan Island contribute to the sediments on the middle and
