Krüppel-like factor 10 (KLF10), also known as TGFβ-inducible early gene-1 (TIEG1), was first found in human osteoblasts. Early studies show that KLF10 plays an important role in osteogenic differentiation. Through decades of research, KLF10 has been found to have complex functions in many different cell types, and its expression and function is regulated in multiple ways. As a downstream factor of transforming growth factor β (TGFβ)/SMAD signaling, KLF10 is involved in various biological functions, including glucose and lipid metabolism in liver and adipose tissue, the maintenance of mitochondrial structure and function of the skeletal muscle, cell proliferation and apoptosis, and plays roles in multiple disease processes, such as nonalcoholic steatohepatitis (NASH) and tumor. Besides, KLF10 shows gender-dependent difference of regulation and function in many aspects. In this review, the biological functions of KLF10 and its roles in disease states is updated and discussed, which would provide new insights into the functional roles of KLF10 and a clearer view of potential therapeutic strategies by targeting KLF10.

Higd1a is a conserved gene in evolution which is widely expressed in many tissues in mammals.Accumulating evidence has revealed multiple functions of Higd1a,as a mitochon-drial inner membrane protein,in the regulation of metabolic homeostasis.It plays an impor-tant role in anti-apoptosis and promotes cellular survival in several cell types under hypoxic condition.And the survival of porcine Sertoli cells facilitated by Higd1a helps to support repro-duction.In some cases,Higd1a can serve as a sign of metabolic stress.Over the past several years,a considerable amount of studies about how tumor fate is determined and how cancerous proliferation is regulated by Higd1a have been performed.In this review,we summarize the physiological functions of Higd1a in metabolic homeostasis and its pathophysiological roles in distinct diseases including cancer,nonalcoholic fatty liver disease(NAFLD),type Il diabetes and mitochondrial diseases.The prospect of Higd1a with potential to preserve mammal health is also discussed.This review might pave the way for Higd1a-based research andapplicationinclinicalpractice.

Ortho-sulfur substituted arylphosphine oxides and arylphosphonates are the precursors of orthosulfurated arylphosphanes,which are widely utilized as ligands in transition metal-catalyzed reactions.However,efficient synthetic methods to access such compounds are sparse.Herein,highly valuable ortho-sulfur substituted arylphosphine oxides and arylphosphonates were constructed via difunctionalization of aryne precursors with P(O)H and elemental sulfur.This method avoids the use of transition metal catalysts and corrosive phosphorus chloride.The synthetic utility of this reaction is further demonstrated by the reduction of the synthesized products to access monophosphine ligands and their applications in cross-coupling reactions.Moreover,by introducing an alkyne moiety into the P(O)H substrate,cyclic sulfur-containing phosphines could be obtained in a one-pot reaction,which represents a new P,S structure that is inaccessible by known strategies.

Clinicopathological practice in the differential diagnosis of mucoepidermoid carcinoma from neoplasms with mucinous component

Background:The differential diagnosis of mucoepidermoid carcinoma(MEC)from neoplasm undergoing mucinous features brings more pitfalls to pathologists.Combining specific MAML2 gene rearrangement and histological characteristics may be the solution.Methods:Twenty-five tumors with mucinous components were selected for differential diagnosis of MEC.All the cases were detected for MAML2 gene rearrangement.The cases diagnosed as MEC were classified into four variants:classic,oncocytic,Warthin-like,and nonclassified,and they were graded using the Brandwein system.The histological characteristics of non-MECs were summarized for differential diagnosis.Univariate survival analysis was performed on MECs.Results:There were 16 MECs;62.5%were MAML2 rearranged.For the low-,intermediate-,and high-grade MECs,the rate of rearrangement was 83.3%,100%,and 28.6%,respectively.Both the oncocytic and Warthinlike MECs were MAML2 rearranged.For the classic and nonclassified MECs without MAML2 rearrangement,non-keratinized squamoid cells and distinctive mucinous cells were essential diagnostic criteria.On survival analysis,all the disease progression occurred in high-grade MECs(p=0.038).Nine cases were diagnosed as non-MECs:pleomorphic adenoma with mucinous metaplasia showed no ex-capsular involvement;metaplastic Warthin tumor appeared with overt keratinization and residual oncocytic bilayered epithelium;mix squamous cell and glandular papilloma showed an endobronchial papillary growing pattern;adenosquamous carcinoma was accompanied by squamous carcinoma in situ of the overlying mucosa.All the non-MECs were negative for MAML2 rearrangement.Conclusion:The application of combining MAML2 rearrangement and histological characteristics is helpful in the differential diagnosis between MEC and other tumors with mucinous components.

Dear Sir,UVA/riboflavin cross-linking （CXL） has been used clinically applied for the treatment of keratoconus and corneal edema via enhancement of corneal stiffness The safety of the corneal endothelium is of prime importance during CXL treatment. In clinical practice, a corneal thickness （CT） of 400um has traditionally been regarded as the minimum treatable thickness, thereby avoiding damage to the corneal endothelium Although CXL has been applied to thinner corneas, using a hypoosmotic solution onto cornea and inducing edema . CXL safety still needs further evaluation because of lower relative concentration of collagen in the hydrated stroma . This study aims to evaluate the changes of corneal endothelial density （ECD） in cases where the CT is 〈400 um before iatrogenic corneal swelling and CXL treatment.

Accumulating evidence in recent years indicates that DNA methylation(5-methyl-2-deoxycytidine,5-mdC) and hydroxymethylation(5-hydroxymethyl-2-deoxycytidine, 5-hmd C) have been implicated in various biological processes, and the aberrations of these DNA cytosine modifications is tightly associated with cancer. N6-methyl-2-deoxyadenosine(m~6dA), as a newly discovered epigenetic modification in genome of mammals, has been demonstrated to play vital regulatory roles in tumorigenesis. However, the content information of m~6dA in human tumor tissues is still limited and pan-cancer analysis of these DNA epigenetic modifications is lacked. Herein, we developed a sensitive and robust stable isotopediluted hydrophilic interaction liquid chromatography-tandem mass spectrometry(HILIC-MS/MS) method for accurate quantification of m~6dA, 5-mdC and 5-hmdC in genomic DNA from 82 pairs of human tumor tissues and matched tumor-adjacent normal tissues. The types of tumors included esophagus cancer, lung cancer, breast cancer, liver cancer, pancreatic cancer, gastric cancer, stromal tumor and colorectal cancer.Compared to the normal tissues, we revealed the level of m6dA was increased in tumor tissues of esophagus cancer, lung cancer and liver cancer, whereas the level of m~6dA was diminished in tumor tissues of pancreatic cancer and gastric cancer;while the contents of 5-mdC and 5-hmdC exhibited significant decrease in tumor tissues of most types of cancer. It is worth noting that we revealed, for the first time,the content of genomic m~6dA in pancreatic cancer, stromal tumor and colorectal cancer. The significant changes of these DNA epigenetic modifications indicate they may serve as indicators of cancers. In addition, this study will benefit for better understanding of the regulatory roles of these DNA epigenetic modifications in cancers.

A novel approach of tight oil reservoirs stimulation based on fracture controlling optimization and design

To deal with the stress interference caused by simultaneous propagation of multiple fractures and the wettability reversal and physical property changes of the reservoir caused by fracturing fluid getting in during large-volume fracturing of tight oil reservoirs through a horizontal well, a non-planar 3D fracture growth model was built, wettability reversal characterizing parameters and change of relative permeability curve were introduced to correct the production prediction model of fractured horizontal well, a fracturing design optimization software(Fr Smart) by integrating geological and engineering data was developed, and a fracturing design optimization approach for tight oil reservoirs based on fracture control was worked out. The adaptability of the method was analyzed and the fracture parameters of horizontal wells in tight oil reservoirs were optimized. The simulation results show that fracturing technology based on fracture control is suitable for tight oil reservoirs, and by optimizing fracture parameters, this technology makes it possible to produce the maximum amount of reserves in the well-controlled unit of unconventional reservoirs. The key points of fracturing design optimization based on fracture control include increasing lateral length of and reducing the row spacing between horizontal wells, increasing perforation clusters in one stage to decrease the spacing of neighboring fractures, and also avoiding interference of old and new fracturing wells. Field tests show that this technology can increase single well production and ultimate recovery. Using this technology in developing unconventional resources such as tight oil reservoirs in China will enhance the economics significantly.

The corneal crosslinking （CXL） with riboflavin and ultraviolet-A （UVA） is a new therapy method to successfully treat infectious keratitis in clinical practice. However, there are rare reports on the complications of CXL such as the secondary keratitis. The diverse clinical outcomes on keratitis have highlighted the necessity to further evaluate the efficacy and complications of CXL. We reviewed the positive and negative reports on UVA/riboflavin related with keratitis and provided our opinion on the therapeutic and side effect of UVA/riboflavin crosslinking on keratitis.
