AIM To explore the effect of miR-382 on esophageal squamous cell carcinoma (ESCC) in vitro and its possible molecular mechanism. METHODS Eca 109 cells derived from human ESCC and Het-1A cells derived from human normal esophageal epithelium were used. Lentivirus-mediated miR-382 was overexpressed in Eca109 cells. The effect of miR-382 on cell proliferation was evaluated by MTT and colony formation assay. For cell cycle analysis, cells were fixed and stained for 30 min with propidium iodide (PI) staining buffer containing 10 mg/mL PI and 100 mg/mL RNase A, and analyzed by BD FACSCalibur (TM) flow cytometer. For cell apoptosis assay, cells were stained with an Annexin V-FITC/PI Apoptosis Detection Kit according to the manufacturer's instructions and analyzed by a dual-laser flow cytometer. Cell invasion and migration abilities were determined through use of transwell chambers, non-coated or pre-coated with matrigel. Levels of proteins related to cell growth and migration were examined by western blotting. RESULTS Endogenous miR-382 was down-regulated in Eca109 cells compared with Het-1A. Introduction of miR-382 not only significantly inhibited proliferation and colony formation, but also arrested cell cycle at the G2/M phase, as well as promoted apoptosis and autophagy in Eca109 cells. Migration, invasion and epithelial-mesenchymal transition of Eca109 cells were suppressed by overexpressing miR-382. Western blotting results showed that miR-382 inhibited the phosphorylation of mTOR and 4E-BP1. CONCLUSION miR-382 functions as a tumor suppressor against ESCC development and metastasis, and could be considered as a potential drug source for the treatment of ESCC patients.

High chalkiness is a major problem in many rice-producing areas of the world, especially in hybrid rice （Oryza sativa L.） in China. We previously showed a major quantitative trait locus for the percentage of grains with white chalkiness （QTLqPGWC-8） in the interval G1149-R727 on chromosome 8 using a chromosome segment substitution line （CSSL）. Here, we selected the line-CSSL50 harboring the QTLqPGWC-8 allele from the CSSLs derived from a cross between Asominori （as a recurrent parent） and IR24 （as a donor parent）, which had higher percentage chalkiness, markedly different from that of Asominori. There were also significant differences in starch granules, appearance of amylose content （AAC） and milling qualities between Asominori and CSSL50, but not in grain size or thousand grain weight （TGW）. The BC4F2 and BC4F3 populations from a cross between CSSL50 and Asominori were used for fine mapping of qPGWC-8. We narrowed down the location of this QTL to a 142 kb region between Indel markers 8G-7 and 8G-9. QTLqPGWC-8 accounted for 50.9% of the difference in PGWC between the parents. The markers tightly linked to qPGWC-8 should facilitate cloning of the gene underlying this QTL and will be of value for marker-assisted selection in breeding rice varieties with better grain quality.

Flow velocity plays an important role in recirculating aquaculture systems(RAS)and the growing practice of culturing juvenile largemouth bass(Micropterus salmoides).In this study,the effects of flow velocity on the water quality as well as the ammonia excretion were discussed from the perspective of actual production,and a polynomial model of ammonia nitrogen excretion was established,using the juvenile largemouth *** showed that the range of ammonia nitrogen and nitrite nitrogen decreased with flow velocity increasing,while the number and volume share of large particles *** to the polynomial model,compared with the medium flow velocity(11 cm/s,2.45 body length(bl)/s),the ammonia excretion of juvenile largemouth bass at high(18 cm/s,4.00 bl/s),and low(4 cm/s,0.90 bl/s)flow velocity changed faster with time,and the excretion rate peaked at the 6th hour after feeding,earlier than that under medium flow ***,it is suggested to increase the flow velocity at the 5th hour after feeding and then decreased it at the 10th hour,to ensure better water quality in RAS culturing juvenile largemouth bass.
