In order to study the response law of vortex-induced vibration(VIV)of marine risers under the combined action of roughness and interference effects,and to reveal the coupling mechanism of roughness and interference effects on the riser,a VIV experiment of rough risers in tandem arrangement was conducted in a wave−current combined *** experiment characterized the risers’roughness by arranging different specifications of attachments on the surface of the *** rough risers with different roughness and smooth risers were arranged in tandem arrangement,with the rough risers arranged *** experimental results indicate that the suppression of the attachments on the downstream risers’vibration are more significant both in the CF and IL directions as the reduced velocity *** the downstream riser,the amplitude response of rough riser is more significantly weakened compared with the smooth one at high reduced *** the upstream risers,changes in the roughness and spacing ratio have an impact on their‘lock-in’*** the roughness of downstream risers is relatively large(0.1300)and the spacing between risers is small(S/D=4.0),the reduced velocity range of‘lock-in’region in the CF direction of upstream risers is obviously expanded,and the displacement in the‘lock-in’region is severer.

Fruitlet calyx shedding in pear plants is apparently regulated via numerous pathways that involve both environmental triggers and phytohormones cues such as auxin. In this study, we found at 10 days after full bloom (DAFB) higher levels of indoleacetic acid (IAA) and tryptophan (Trp) in calyx persistence fruitlet (CPF) than calyx shedding fruitlet (CSF) ofDanshan Suli’ pear (Pyrus bretschneideri Rhed.). Consisting with this, the activity of indolealdehyde oxidase (IAAIdO), which promotes IAA synthesis, was remarkably increased, and that of peroxidase(POD), which degrades IAA, dropped markedly in CPF but not in CSF. Further, qRT-PCR results revealed that most of 31 PbrARFs (encoding auxin response factors) in Pyrus bretschneideri were highly expressed in CPF, whereas PbrARF4, PbrARF24 and PbrARF26 were significantly downregulated in CPF vis-a-vis CSF. Phylogenetic analysis revealed that 6 PbrARFs clustered in the group III, where PbrARF4 showed the closest affinity with AtARF1 that promotes organ abscission, indicating a putative role of PbrARF4 in mediating the process of calyx shedding in pear. In fact, the ectopic overexpression of PbrARF4 in Solanum lycopersicum resulted in an earlier-formed and deeper abscission layer (AL) in the transgenic plants, whose calyxes were more prone to wilt at the mature red stage (MR) compared with the control plants (wild-type). More importantly, expression levels of the abscission genes SILS and Sl Cel2 in transgenic plants overexpressing PbrARF4 were significantly upregulated in comparation with the WT, whereas those of Sl BI and Sl TAPG2 were considerably inhibited. Further, PbrJOINTLESS and PbrIDA,the two genes related to calyx shedding in pear, were up-regulated more in CSF than CPF. The findings contribute to a better understanding of PbrARFs involved in fruitlet calyx shedding of pear, which could prove beneficial to improving the quality of pear fruit.

Glucose plays a vital part in milk protein synthesis through the mTOR signaling pathway in bovine mammary epithelial cells (BMEC). The objectives of this study were to determine how glucose affects hexokinase (HK) activity in BMEC and investigate the regulatory effect of HK in kappa casein (CSN3) synthesis via the mechanistic target of rapamycin complex 1 (mTORC1) signaling pathway in BMEC. For this,HK1 andHK2 were knocked out in BMEC using the CRISPR/Cas9 system. The gene and protein expression, glucose uptake, and cell proliferation were measured. We found that glucose uptake, cell proliferation,CSN3 gene expression levels, and expression of HK1 and HK2 increased with increasing glucose concentrations. Notably, glucose uptake was significantly reduced inHK2 knockout (HK2KO) BMEC treated with 17.5 mM glucose. Moreover, under the same glucose treatment conditions, the proliferative ability and abundance of CSN3 were significantly diminished in bothHK1 knockout (HK1KO) and HK2KO BMEC compared with that in wild-type BEMC. We further observed that the phosphorylation levels of ribosome protein subunit 6 kinase 1 (S6K1) were reduced in HK1KO and HK2KO BMEC following treatment with 17.5 mM glucose. As expected, the levels of glucose-6-phosphate and the mRNA expression levels of glycolysis-related genes were decreased in both HK1KO and HK2KO BMEC following glucose treatment. Microstructures, Fabrics, and Seismic Properties of Mylonitic Amphibolites: Implications for Strain Localization in a Thickening Anisotropic Middle Crust of the North China Craton

Strain localization processes in the continental crust generate faults and ductile shear zones over a broad range of scales affecting the long-term lithosphere deformation and the mechanical response of faults during the seismic *** anisotropy originated within the continental crust can be applied to deduce the kinematics and structures within orogens and is widely attributed to regionally aligned minerals,e.g.,***,naturally deformed rocks commonly show various structural layers(e.g.,strain localization layers).It is necessary to reveal how both varying amphibole contents and fabrics in the structural layers of strain localization impact seismic property and its interpretations in terms of *** present microstructures,petrofabrics,and calculate seismic properties of deformed amphibolite with the microstructures ranging from mylonite to *** transition from mylonite to ultramylonite is accompanied by a slight decrease of amphibole grain size,a disintegration of amphibole and plagioclase aggregates,and amphibole aspect ratio increase(from 1.68 to 2.23),concomitant with the precipitation of feldspar and/or quartz between amphibole *** intensities of amphibole crystallographic preferred orientations(CPOs)show a progressively increasing trend from mylonitic layers to homogeneous ultramylonitic layers,as indicated by the JAm index increasing from 1.9–4.0 for the mylonitic layers and 4.0–4.8 for the transition layer,to 5.1–6.9 for the ultramylonitic *** CPO patterns are nearly random for plagioclase and *** amphibole aggregates in the amphibolitic mylonite deform by diffusion,mechanical rotation,and weak dislocation creep,and develop CPOs *** polymineralic matrix(such as quartz and plagioclase)of the mylonite and the ultramylonite deform dominantly by dissolution-precipitation,combined with weak dislocation *** mean P and S wave velocities are estimated to be 6.3 and 3.5 km/
