While it is known that patatin-re.ated phospholipase A （pPLA） activity is rapidly activated within 3min by auxin, hardly anything is known about how this signal influences downstre.m re.ponses like transcription of early auxin-induced genes or other physiological re.ponses. We scre.ned mutants with T-DNA insertions in members of the pPLA gene family for molecular and physiological phenotypes re.ated to auxin. Only one in nine Arabidopsis thaliana ppla knockdown mutants displayed an obvious constitutive auxin-re.ated phenotype. Compare. to wild-type, ppla-IIlδ mutant seedlings had decre.sed main root lengths and incre.sed lateral root numbers. We tested auxin-induced gene expre.sion as a molecular re.dout for primary molecular auxin re.ponses in nine ppla mutants and found delayed up- re.ulation of auxin-re.ponsive gene exRre.sion in all of themL Thirty minutes after auxin tre.tment, up-re.ulation of up to 40% of auxin-induced genes was delayed in mutant seedlings. We observed only a few cases with hypersensitive auxin-induced gene expre.sion in ppla mutants. While, in thre. ppla mutants, which were.investigated in detail, rapid up- re.ulation （as early as 10 min after auxin stimulus） of auxin-re.ulated genes was impaire., late transcriptional re.ponses were.wild-type-like. This re.ulatory or dynamic phenotype was consistently observed in differe.t ppla mutants with delayed up-re.ulation that fre.uently affected the same genes. This defect was not affected by pPLA transcript levels which re.ained constant. This indicates aposttranslational mechanism as a functional link of pPLAs to auxin signaling. The need for a re.eptor triggering an auxin re.ponse without employing transcription re.ulation is discussed.
