The idea of using ultrashort X-ray pulses to obtain images of single proteins frozen in time has fasc.nated and inspired *** was one of the arguments for building X-ray free-elec.ron *** to theory,the extremely intense pulses provide suffic.ent signal to dispense with using c.ystals as an amplifier,and the ultrashort pulse duration permits c.pturing the diffrac.ion data before the sample inevitably *** was first demonstrated on biologic.l samples a dec.de ago on the giant *** then,a large c.llaboration has been pushing the limit of the smallest sample that c.n be *** ability to c.pture snapshots on the timesc.le of atomic.vibrations,while keeping the sample at room temperature,may allow probing the entire c.nformational phase spac. of *** we show the first observation of an X-ray diffrac.ion pattern from a single protein,that of Esc.eric.ia c.li GroEL whic. at 14 nm in diameter is the smallest biologic.l sample ever imaged by X-rays,and demonstrate that the c.nc.pt of diffrac.ion before destruc.ion extends to single *** the pattern,it is possible to determine the approximate orientation of the *** experiment demonstrates the feasibility of ultrafast imaging of single proteins,opening the way to single-molec.le time-resolved studies on the femtosec.nd timesc.le.

c.itosan oligosac.harides(c.S)are the hydrolyzed produc. of c.itosan and have multifunc.ional health *** objec.ive of this study was to eluc.date the effec. of c.S as a dietary supplement to gilts on their produc.ivity and health and that of their *** were randomly assigned to either a treatment(n=30)or c.ntrol group(n=30).The treatment gilts were fed a standard dry sow ration supplemented with c.S at 0.12 and 0.24 g/gilt per d during gestation and lac.ation,respec.ively,and the c.ntrol group was fed the standard dry sow ration *** body weight,reproduc.ive performanc.,milk produc.ion and litter size for eac. gilt and body weight of c.rresponding litters were *** serum immunoglobulins(IgA,IgG,IgM)and sec.etory immunoglobulin A(sIgA)c.nc.ntrations of gilts and piglets and fec.l sIgA c.nc.rtation of gilts were measured by Enzyme-linked immunosorbent assay(ELISA).Our study showed that maternal c.S supplementation 1)signific.ntly inc.eased gilt body weight in late pregnanc.(Pc.ntly inc.eased milk produc.ion of gilts at different stages(d 1,3,7 and19)of lac.ation(Pc.ntly inc.eased body weight gain of piglets at weaning(Pc.ntly inc.eased the serum c.nc.ntrations of IgM and sIgA in piglets,and slgA in fec.l sample of gilts(Pc.ease the pregnanc. suc.ess rate(P>0.05)in the treatment group c.mpared to the c.ntrol *** results suggest that maternal c.S intervention in gilts c.n improve gilt milk produc.ion,piglet pre-weaning growth and immunity parameters in both gilts and piglets.

目的研究选择性头部降温对缺血性脑损伤胎羊纹状体神经元凋亡和星形胶质细胞增殖的影响。方法胎羊于妊娠117~124d时通过双侧颈动脉阻塞30min造成双侧脑缺血损伤,损伤后将胎羊随机分为：损伤组（n=10）、2h低温组（损伤后2h开始亚低温治疗,n=7）和6h低温组（损伤后6h开始亚低温治疗,n=8）,另设正常对照组（n=5）。通过冷循环水进行选择性头部降温,取脑组织用免疫组化法检测胎羊纹状体c.spase-3（半胱天冬氨酸酶-3）,GFAP（胶质纤维酸性蛋白）和Pc.A（增殖细胞核抗原）的表达。结果①纹状体神经元凋亡：正常对照组中,c.spase-3表达极少（11.00±13.77）,损伤组c.spase-3免疫阳性细胞为177.70±48.69,明显增加（P=0.000）,损伤后2h治疗组（54.14±39.44,P=0.000）和损伤后6h治疗组（122.43±52.36,P=0.017）均能减少c.spase-3免疫阳性细胞。②纹状体星形胶质细胞增殖：与正常对照组（163.40±21.98）相比,缺血性脑损伤组的GFAP免疫阳性细胞明显增多（433.25±66.69,P=0.000）,损伤后2h开始亚低温治疗（219.50±35.31,P=0.000）和损伤后6h开始亚低温治疗（272.50±86.20,P=0.000）均能减少GFAP免疫阳性细胞。③纹状体Pc.A阳性细胞的表达：在正常对照组中,Pc.A免疫阳性细胞较少,为153.40±12.46,缺血性脑损伤组的Pc.A免疫阳性细胞明显增多（353.70±45.60,P=0.000）,损伤后2h开始亚低温治疗（187.14±26.26,P=0.000）和损伤后6h开始亚低温治疗（230.25±67.46,P=0.000）均能减少Pc.A免疫阳性细胞。结论亚低温可以抑制纹状体神经元的凋亡和星形胶质细胞的增殖,该作用可能为选择性头部降温的脑保护作用机制之一。

背景：儿童高血压的发病率随着儿童肥胖的盛行而呈上升趋势。在儿童中，正常和异常血压值会随年龄、性别和身高的变化而变化，难以记忆，因此儿童高血压的诊断较为复杂。目的：明确高血压和高血压前期的漏诊率以及与漏诊相关的患者因素。设计、地点及参试者：于俄亥俄州东北部某大型医疗学术系统进行队列研究，对1999年6月至2006年9月于该系统至少接受3次健康保健检查的14187例3—18岁儿童青少年进行观察。对于3次以上健康检查结果符合高血压和高血压前期诊断标准的儿童和青少年，按照国际疾病分类第9版诊断列表、问题列表和既往病史列表，确定其相关比例。主要观测指标：电子病历诊断为高血压或高血压前期患儿的比例以及≥3次检查结果显示血压升高患儿（校正年龄和身高）的比例。应用多变量logistic.归确定与正确诊断相关的患者因素。结果：在507例（3．6％）高血压儿童青少年中，131例（26％）电子病历中有高血压或血压增高诊断。可增加正确诊断率的患者因素包括3岁以上年龄增加1岁（优势比[oddsratio，OR]，1．09；95％c.，1．03—1．16），血压升高超过3次的次数（OR，1．77；95％c.，1．21～2．57），年龄别身高百分位数增长1％（OR，1．02；95％c.，1．01—1．03），有肥胖相关诊断（OR，2．61；95％c.，1．49—4．55）以及处于高血压Ⅱ期的次数（OR，1．68；95％c.，1．29—2．19）。在485例（3．4％）高血压前期儿童青少年中，55例（11％）电子病历记录有适宜的诊断。增加高血压前期诊断率的患者因素包括3岁以上年龄增加1岁（OR，1．21；95％c.，1．09—1．34）以及血压升高超过3次的次数。结论：在本儿童人群中，高血压和高血压前期常被漏诊。患儿的年龄、身高、肥胖相关诊断以及异常血压读数大小和频率均可增加诊断的几率。
