Stochastic weather generators create time series that reproduce key weather dynamics present in long-term *** dataset detailed herein is a large-scale gridded parameterization for CLImate GeNerator(CLIGeN)that fills spatial gaps in the coverage of existing regional CLIGeN parameterizations,thereby obtaining near-global availability of combined *** dataset primarily covers countries north of 40°latitude with 0.25°spatial *** CLIGeN parameters were estimated based on 20-year records from four popular global climate *** parameters were statistically downscaled to estimate point-scale values,while point-scale temperature and solar radiation parameters were approximated by direct calculation from high-resolution *** parameter values were used in some cases,such as with wind ***-validation was done to assess the downscaling approach for six precipitation parameters using known point-scale values from ground-based CLIGeN *** parameter values were derived from daily accumulation records at 7,281 stations and high temporal resolution records at 609 *** sensitive parameters,monthly average storm accumulation and maximum 30-minute intensity,were shown have RMSe values of 1.48 mm and 4.67 mm hr^(−1),*** precipitation and the annual number of days with precipitation occurrence were both within 5%of ground-based parameterizations,effectively improving climate data availability.

CLIGeN is a stochastic weather generator that creates statistically representative timeseries of daily and sub-daily point-scale weather variables from observed monthly statistics and other parameters. CLIGeN precipitation timeseries are used as climate input for various risk-assessment modelling applications as an alternative to observe long-term, high temporal resolution records. Here, we queried gridded global climate datasets (TerraClimate, eRA5, GPM-IMeRG, and GLDAS) to estimate various 20-year climate statistics and obtain complete CLIGeN input parameter sets with coverage of the African and South American continents at 0.25 arc degree resolution. The estimation of CLIGeN precipitation parameters was informed by a ground-based dataset of >10,000 locations worldwide. The ground observations provided target values to fit regression models that downscale CLIGeN precipitation input parameters. Aside from precipitation parameters, CLIGeN’s parameters for temperature, solar radiation, etc. were in most cases directly calculated according to the original global datasets. Cross-validation for estimated precipitation parameters quantified errors that resulted from applying the estimation approach in a predictive fashion. Based on all training data, the RMSe was 2.23 mm for the estimated monthly average single-event accumulation and 4.70 mm/hr for monthly maximum 30-min intensity. This dataset facilitates exploration of hydrological and soil erosional hypotheses across Africa and South America.

UAV and Satellite-Based Sensing to Map ecological States at the Landscape Scale

Mapping ecological states in semi-arid rangelands is crucial for effective land management and conservation efforts because it identifies difference in the ecological conditions across a landscape. This study presents an innovative approach for mapping two ecological states, Large Shrub Grass (LSG) and Large Shrub eroded (LSe), within the Sandy Loam Upland and Deep (SLUD) ecological sites using a combination of drone and satellite data. The methodology leverages the Largest Patch Index (LPI) as a proxy metric to estimate eroded areas and classify ecological states. The integration of unmanned aerial vehicle (UAV) data with satellite-based remote sensing provides a scalable approach that can benefit various stakeholders involved in rangeland management. The study demonstrates the potential of this methodology by generating spatial layers at the landscape scale to inform on the state of rangeland ecosystems. The workflow showcases the power of remote sensing technology to map ecological states and addresses limitations in spatial coverage by integrating UAV and satellite data. By utilizing the bare ground LPI metric, which indicates the connectedness of bare ground, the methodology enables the classification of ecological states at a regional scale. This cost-effective approach potentially offers a standardized and reproducible method applicable across different sites and regions. The accuracy of the classification process is evaluated by comparing the results to ground-based polygons, dirt roads, and water locations. While the model performs well in identifying eroded areas, misclassifications occur in regions with mixed vegetation cover or low biomass. Future research should focus on incorporating temporal information from historical remote sensing archives to improve understanding of ecological state dynamics. Food-derived peptides with inhibitory capacity for HMG-CoA reductase activity: a potential nutraceutical for hypercholesterolemia

Cardiovascular diseases (CVDs) are the leading global cause of mortality and disease burden. Statins are the most prescribed lipid-lowering drugs to treat hypercholesterolemia and prevent CVDs. The biochemical mechanism of statins consists of competitive inhibition of the 3-hydroxy-3-methylglutaryl-CoA reductase enzyme (HMG-CoAR)， the limiting enzyme in cholesterol biosynthesis. Due to statin intolerance in some patient groups， the search for new inhibitors is a field of great interest. This review focusses on the studies reporting the inhibitory effect of protein hydrolysates and biopeptides on the HMG-CoAR enzyme activity. The analysis of the action mechanism and physicochemical characteristics of the HMG-CoAR inhibitory peptides revealed that the molecular weight， amino acid composition， charge， and polarity are key aspects of the interaction with the HMG-CoAR enzyme. In conclusion， this review reveals the potential of using food peptides as new cholesterol-lowering agents and opens a new interesting field of research. However， clinical approaches are mandatory to confirm their therapeutic hypercholesterolemic effect.

Identification of the autotransporter Pet toxin in Proteus mirabilis strain isolated from patients with urinary tract infections

Proteus mirabilis, a motile Gram-negative bacterium, represents a common cause of complicated urinary tract infections. Autotransporters are a family of secreted proteins from Gram-negative bacteria that direct their own secretion across the outer membrane (type V autotransporter secretion mechanism). Serine protease autotransporters of enterobacteriaceae (SPATes) include adhesins, toxins, and proteases that can contribute to the virulence. Plasmid-encoded toxin (Pet) is the predominant protein in culture supernatants of enteroaggregative e. coli prototype strain 042 and has been extensively studied. Pet toxin is encoded on the 65-MDa adherence-related plasmid of eAeC 042 strain. In this work, Pet protein was found in the supernatant obtained from Proteus mirabilis RTX339 strain isolated from a psychiatric patient suffering complicated urinary tract infections (UTIs). The nucleotide sequence of pet gene was obtained using primers designed from e. coli 042 pet gene reported. The alignment of the sequence showed 100% identity with the pet gene reported. Is important to note that Proteus mirabilis RTX339 pet gene has chromosomal location. The chromosomal location of the gene was established since no plasmids were harbored by this strain.
