Ultrastructural and enzymatic alterations in the ovary of Rhodnius prolixus infected with Trypanosoma rangeli

Objective:To investigate the morphological structure of ovarian follicular cells and biochemical parameters of both ovaries and fat bodies(sites of vitellogenesis)from Rhodnius(R.)prolixus infected with Trypanosoma(T.)rangeli.Methods:Adult virgin females of R.prolixus were fed upon a membrane apparatus containing heat-inactivated citrated rabbit blood and a suspension of T.rangeli epimastigotes(Macias strain).Females from the control group and all the males received parasite-free blood.Transmission electron microscopy was used to reveal the morphological aspects of ovarian follicle cells in both control and parasite-infected groups.Protein profile,proteolytic activities and Western blotting analyses were performed in either ovary or fat body samples of control and parasite-infected groups.Results:According to the ultrastructural data,T.rangeli infection elicited a degeneration process in the ovarian follicular cells of R.prolixus.Proteolytic assays indicated a reduction in the activity of aspartic peptidases in the ovary and fat body from parasite-infected group,while a significant increase in the cysteine peptidase activity was measured in both insect organs.Additionally,immunoblotting revealed that vitellogenin was overexpressed in the ovary of parasite-infected insects.Conclusions:T.rangeli infection seems to elicit an early programmed cell death in the ovarian follicle cells as well as induces the modulation on the activities of different peptidase classes in either ovaries or fat bodies and the overexpression of the vitellogenin in the ovary of R.prolixus.

Chitin powder and chitin-aerogel were prepared from shrimp wastes and used to uptake Y^(3+)from aqueous solutions and concentrate this rare earth element from phosphogypsum(PG).Chitin aerogel displays a specific surface area of 945 m^(2)/g,while chitin powder is 3.6 m^(2)/g,which largely influences its adsorption ability.Regarding the adsorption in synthetic solutions,the effect of pH on Y^(3+)removal is strong for chitin powder adsorbent.In contrast,no big pH influence was detected for chitin aerogel.Electrostatic interactions and chelation can highlight the proposed mechanism of Y^(3+)on chitin adsorbents for the powder and aerogel.Furthermore,in addition to these interactions,pore filling/pore diffusion is the main mechanism of Y^(3+)removal in the chitin aerogel.Chitin aerogel is efficient in concentrating 8 times the Y^(3+)from PG,a very complex matrix.The complex chitin aerogel-Y^(3+)can be a secondary source of rare earth elements for other applications.

Carbon-based solid lubricants are excellent options to reduce friction and wear,especially with the carbon capability to adopt different allotropes forms.On the macroscale,these materials are sheared on the contact along with debris and contaminants to form tribolayers that govern the tribosystem performance.Using a recently developed advanced Raman analysis on the tribolayers,it was possible to quantify the contactinduced defects in the crystalline structure of a wide range of allotropes of carbon-based solid lubricants,from graphite and carbide-derived carbon particles to multi-layer graphene and carbon nanotubes.In addition,these materials were tested under various dry sliding conditions,with different geometries,topographies,and solid-lubricant application strategies.Regardless of the initial tribosystem conditions and allotrope level of atomic ordering,there is a remarkable trend of increasing the point and line defects density until a specific saturation limit in the same order of magnitude for all the materials tested.

Anomalies in the short-range local environment and atomic diffusion in single crystalline equiatomic CrMnFeCoNi highentropy alloy

Multi-edge extended X-ray absorption fine structure(EXAFs)spectroscopy combined with reverse Monte Carlo(RMC)simulations was used to probe the details of element-specific local coordinations and component-dependent structure relaxations in single crystalline equiatomic CrMnFeCoNi high-entropy alloy as a function of the annealing temperature.Two representative states,namely a high-temperature state,created by annealing at 1373 K,and a low-temperature state,produced by long-term annealing at 993 K,were compared in detail.specific features identified in atomic configurations of particular principal components indicate variations in the local environment distortions connected to different degrees of compositional disorder at the chosen representative temperatures.The detected changes provide new atomistic insights and correlate with the existence of kinks previously observed in the Arrhenius dependencies of component diffusion rates in the CrMnFeCoNi high-entropy alloy.

BACKGROUND Helicobacter pylori(H.pylori)infection has been well-established as a significant risk factor for several gastrointestinal disorders.The urea breath test(UBT)has emerged as a leading non-invasive method for detecting H.pylori.Despite numerous studies confirming its substantial accuracy,the reliability of UBT results is often compromised by inherent limitations.These findings underscore the need for a rigorous statistical synthesis to clarify and reconcile the diagnostic accuracy of the UBT for the diagnosis of H.pylori infection.AIM To determine and compare the diagnostic accuracy of 13C-UBT and 14C-UBT for H.pylori infection in adult patients with dyspepsia.METHODs We conducted an independent search of the PubMed/MEDLINE,EMBAsE,and Cochrane Central databases until April 2022.Our search included diagnostic accuracy studies that evaluated at least one of the index tests(^(13)C-UBT or ^(14)C-UBT)against a reference standard.We used the QUADAs-2 tool to assess the methodo-logical quality of the studies.We utilized the bivariate random-effects model to calculate sensitivity,specificity,positive and negative test likelihood ratios(LR+and LR-),as well as the diagnostic odds ratio(DOR),and their 95%confidence intervals.We conducted subgroup analyses based on urea dosing,time after urea administration,and assessment technique.To investigate a possible threshold effect,we conducted spearman correlation analysis,and we generated summary receiver operating characteristic(sROC)curves to assess heterogeneity.Finally,we visually inspected a funnel plot and used Egger’s test to evaluate publication bias.endorsing both as reliable diagnostic tools in clinical practice.CONCLUsION In summary,our study has demonstrated that ^(13)C-UBT has been found to outperform the ^(14)C-UBT,making it the preferred diagnostic approach.Additionally,our results emphasize the significance of carefully considering urea dosage,assessment timing,and measurement techniques for both tests to enhance diagnostic precision.Nevertheless,it is cru

The Use of secondary Grape Biomass in Beef Cattle Nutrition on Carcass Characteristics, Quality and shelf Life of Meat

We determined whether the inclusion of 100 g/kg dry matter of grape pomace silage (GPs) and grape pomace bran (GPB) as substitutes for other traditional fiber sources in the diet of steers (Charolais x Nellore) would improve carcass characteristics, meat quality and composition, and shelf life. Twenty-four animals (248 ± 19.32 kg of initial body weight) were fed a high concentrate diet for 121 days. Carcass characteristics were measured, and the longissimus dorsi muscle was analyzed for fatty acid (FA) profile and composition. The meat was sliced and stored in air-permeable packages for 10 days. On each sampling day (d 1, 3, 7, and 10), oxidative stability, bacterial load, lipid and protein oxidation, and staining were analyzed. The experimental diets influenced the pH of cold carcasses only. The GPs group had a higher pH than the control. The GPs and GPB groups showed improved oxidant status (i.e., lower lipid peroxidation and concentrations of reactive oxygen species were in the meat of both groups than in control). On the first day of storage, the antioxidant enzyme glutathione s-transferase activity was more significant in the meat of the GPs and GPB groups than in the control. The bacterial loads in the meat were attenuated by GPs inclusion;there were lower total coliform counts and a trend toward lower counts for enterobacteria in the control group. The diets altered the FA profile of the meat;i.e., the GPB diet allowed for a more significant amount of the n-6 omegas in the meat, while the GPs diet showed a tendency for a more significant amount of n-6 and 9 omegas. Both diets (GPs and GPB) increased the amounts of long-chain FAs. The GPs diet decreased saturated FA levels. We conclude that the dietary treatments GPs and GPB are a promising alternative to maintain meat quality standards throughout in real-world retail conditions. These treatments gave rise to an improvement in the nutritional value of the meat due to the more significant amounts of FAs that improve human health.

Current and future perspectives on acute-on-chronic liver failure: Challenges of transplantation, machine perfusion, and beyond

Acute-on-chronic liver failure(ACLF)is a syndrome that occurs in patients with chronic liver disease and is characterized by acute decompensation,organ failure and high short-term mortality.Partially due to the lack of universal diagnostic criteria,the actual ACLF prevalence remains unclear;nevertheless,it is expected to be a highly prevalent condition worldwide.Earlier transplantation is an effective protective measure for selected ACLF patients.Besides liver transplantation,diagnosing and treating precipitant events and providing supportive treatment for organ failures are currently the cornerstone of ACLF therapy.Although new clinical specific therapies have been researched,more studies are necessary to assess safety and efficacy.Therefore,future ACLF management strategies must consider measures to improve access to liver transplantation because the time window for this life-saving therapy is frequently narrow.Thus,an urgent and global discussion about allocation and prioritization for transplantation in critically ill ACLF patients is needed because there is evidence suggesting that the current model may not portray their waitlist mortality.In addition,while donor organ quality is meant to be a prognostic factor in the ACLF setting,recent evidence suggests that machine perfusion of the liver may be a safe tool to improve the donor organ pool and expedite liver transplantation in this scenario.

Greenstone belts contain several clues about the evolutionary history of primitive Earth.Here,we describe the volcano-sedimentary rock association exposed along the eastern margin of the Gavião Block,named the Northern Mundo Novo Greenstone Belt(N-MNGB),and present data collected with different techniques,including U–Pb–Hf–O isotopes of zircon and multiple sulfur isotopes(^(32)s,^(33)s,^(34)s,and ^(36)s)of pyrite from this supracrustal sequence.A pillowed metabasalt situated in the upper section of the N-MNGB is 3337±25 Ma old and has zircon with ε_(Hf)(t)=2.47 to1.40,Hf model ages between 3.75 Ga and 3.82 Ga,and δ^(18)O=+3.6‰to+7.3‰.These isotopic data,together with compiled whole-rock trace element data,suggest that the mafic metavolcanic rocks formed in a subduction-related setting,likely a back-arc basin juxtaposed to a continental arc.In this context,the magma interacted with older Eoarchean crustal components from the Gavião Block.Detrital zircons from the overlying quartzites of the Jacobina Group are sourced from Paleoarchean rocks,in accordance with previous studies,yielding a maximum depositional age of 3353±22 Ma.These detrital zircons have ε_(Hf)(t)=5.40 to0.84,Hf model ages between 3.66 Ga and 4.30 Ga,and δ^(18)O=+4.8‰to+6.4‰.The pyrite multiple sulfur isotope investigation of the 3.3 Ga supracrustal rocks from the N-MNGB enabled a further understanding of Paleoarchean sulfur cycling.The samples have diverse isotopic compositions that indicate sulfur sourced from distinct reservoirs.significantly,they preserve the signal of the anoxic Archean atmosphere,expressed by MIF-s signatures(Δ^(33)s between1.3‰to+1.4‰)and a Δ^(36)s/Δ^(33)s slope of0.81 that is indistinguishable from the so-called Archean array.A BIF sample has a magmatic origin of sulfur,as indicated by the limited Δ^(34)s range(0 to+2‰),Δ^(33)s~0‰,and Δ^(36)s~0‰.A carbonaceous schist shows positive Δ^(34)s(2.1‰–3.5‰)and elevated Δ^(33)s(1.2‰–1.4‰)values,with corresponding negative Δ^(36)s between1.2‰to0.2‰,which resemble the isotopic

Adsorption is a unit operation widely used for the tertiary treatment of the most diverse effluents,whose mechanism is based on removing recalcitrant compounds from the organic and inorganic origin.In this process,choosing a suitable adsorbent is a fundamental point.This review article focuses on the adsorbents with natural geological origin:minerals,clays,geopolymers,and even wastes resulted from mining activity.Therefore,over 450 articles and research papers were explored.These materials’main sources are described,and their characteristics,composition,and intrinsic properties are related to adsorption.Herein,we discuss the effects of several process parameters,such as p H,temperature,pollutant,and adsorbent concentration.Furthermore,equilibrium,kinetics,and thermodynamic aspects are also addressed,and relevant regeneration prospects and final disposal.Finally,some suggestions and perspectives on applying these adsorbents in wastewater treatment are presented as future trends.
