NeCAB family of neuronal calcium-binding proteins in health and disease

The N-terminal eF-hand calcium-binding proteins 1–3(NeCAB1–3) constitute a family of predominantly neuronal proteins characterized by the presence of at least one eF-hand calcium-binding domain and a functionally less well characterized C-terminal antibiotic biosynthesis monooxygenase domain. All three family members were initially discovered due to their interactions with other proteins. NeCAB1 associates with synaptotagmin-1, a critical neuronal protein involved in membrane trafficking and synaptic vesicle exocytosis. NeCAB2 interacts with predominantly striatal G-protein-coupled receptors, while NeCAB3 partners with amyloid-β A4 precursor protein-binding family A members 2 and 3, key regulators of amyloid-β production. This demonstrates the capacity of the family for interactions with various classes of proteins. NeCAB proteins exhibit distinct subcellular localizations: NeCAB1 is found in the nucleus and cytosol, NeCAB2 resides in endosomes and the plasma membrane, and NeCAB3 is present in the endoplasmic reticulum and Golgi apparatus. The antibiotic biosynthesis monooxygenase domain, an evolutionarily ancient component, is akin to atypical heme oxygenases in prokaryotes but is not wellcharacterized in vertebrates. Prokaryotic antibiotic biosynthesis monooxygenase domains typically form dimers, suggesting that calcium-mediated conformational changes in NeCAB proteins may induce antibiotic biosynthesis monooxygenase domain dimerization, potentially activating some enzymatic properties. However, the substrate for this enzymatic activity remains uncertain. Alternatively, calcium-mediated conformational changes might influence protein interactions or the subcellular localization of NeCAB proteins by controlling the availability of protein–protein interaction domains situated between the eF hands and the antibiotic biosynthesis monooxygenase domain. The interaction of regular quasi-monochromatic waves with a weakly submerged rectangular shelf is studied by means of CFD simulations. The fundamental incident wave frequency is kept constant for the full set of simulated cases, while the incident wave amplitude is made increase progressively, so that the interaction with the shelf is dominated by almost inviscid non-linear flow for the smallest and by breaking for the highest incident waves. A parameter identification（PI） procedure is used to adapt a reduced model to the highly resolved time-space matrix of wave elevations obtained from the numerical simulations, on the weather and lee side respectively. In particular the wave number and the frequency of the component waves in the reduced model are left uncoupled, thus computed by the PI independently. The comparison of simulated data with experiments generally shows a very good agreement. Free/locked, incident/reflected, first/higher order wave components are quantified accurately by the PI and the energy transfer to super-harmonics is clearly evidenced. Moreover the results of the PI show clearly a very large increase in the phase speed of the higher order free waves on the lee side of the shelf, with increasing deviation from the linear behavior with increasing incident wave amplitude.

The finite element method was used to simulate the crack propagation in a pipeline in the presence of soil backfill. Since typical values for soil modulus are known, a spring element can be used to simulate the effects of backfilled soil on crack propagation. This consists of the relationship between the soil property and the stiffness of the spring element, and the effect of backfill depth on the crack propagation in the pipes. By equating crack driving force, G, in the presence of soil, to the dynamic fracture toughness of the pipeline material, a theoretical predictive capability was obtained for the gas transmission and distribution pipelines. Numerical calculations had also been carried out for the cracked pipe with backfill and without backfill. (edited author abstract) 9 Refs.

Water-rich clay to sand suspensions show a shear rate dependent flow behavior and knowledge of the appropriate rheological model is relevant for sedimentological, industrial and hydraulic studies. We present experimental rheological measurements of water-rich(40 to 60 wt%) clay to silt(population A) and silt to sand(population B) suspensions mixed in different proportions. The data evidence a shear rate dependent shear thinning-shear thickening transition. At lower shear rates, the suspensions organize in chains of particles, whereas at higher shear rates, these chains disrupt so increasing the viscosity. The viscosity, consistency and yield stress decrease as the A+B fraction decreases as the content of B particles increases. This behavior reflects the competing effects of the lubrication and frictional processes as a function of particle size and water content. Transitional flows form by the incorporation of small amounts of the finer fraction while ‘oceanic floods’ form at the estuary of rivers and the submarine debris-flows increase their velocity by incorporating water. The critical Reynolds number of the studied suspensions is ～2000±100 suggesting that the grainsize plays a major role in the laminar to turbulent transition. our results have implications for the modeling of sediment flows and the hazard related to floods.

This paper proposes an online adaptive approximate solution for the infinite-horizon optimal tracking control problem of continuous-time nonlinear systems with unknown *** requirement of the complete knowledge of system dynamics is avoided by employing an adaptive identifier in conjunction with a novel adaptive law, such that the estimated identifier weights converge to a small neighborhood of their ideal *** adaptive steady-state controller is developed to maintain the desired tracking performance at the steady-state, and an adaptive optimal controller is designed to stabilize the tracking error dynamics in an optimal manner. For this purpose, a critic neural network(NN) is utilized to approximate the optimal value function of the Hamilton-Jacobi-Bellman(HJB) equation,which is used in the construction of the optimal controller. The learning of two NNs, i.e., the identifier NN and the critic NN, is continuous and simultaneous by means of a novel adaptive law design methodology based on the parameter estimation *** of the whole system consisting of the identifier NN,the critic NN and the optimal tracking control is guaranteed using Lyapunov theory; convergence to a near-optimal control law is proved. Simulation results exemplify the effectiveness of the proposed method.
