Tranylcypromine upregulates Sestrin 2 expression to ameliorate NLRP3-related noise-induced hearing loss

Noise-induced hearing loss is the primary non-genetic factor contributing to auditory ***,there are currently no effective pharmacological interventions for patients with noise-induced hearing ***,we present evidence suggesting that the lysine-specific demethylase 1 inhibitor–tranylcypromine is an otoprotective agent that could be used to treat noise-induced hearing loss,and elucidate its underlying regulatory *** established a mouse model of permanent threshold shift hearing loss by exposing the mice to white broadband noise at a sound pressure level of 120 d B for 4 *** found that tranylcypromine treatment led to the upregulation of Sestrin2(SESN2)and activation of the autophagy markers light chain 3B and lysosome-associated membrane glycoprotein 1 in the cochleae of mice treated with *** noise exposure group treated with tranylcypromine showed significantly lower average auditory brainstem response hearing thresholds at click,4,8,and 16 k Hz frequencies compared with the noise exposure group treated with *** findings indicate that tranylcypromine treatment resulted in increased SESN2,light chain 3B,and lysosome-associated membrane glycoprotein 1 expression after noise exposure,leading to a reduction in levels of 4-hydroxynonenal and cleaved caspase-3,thereby reducing noise-induced hair cell ***,immunoblot analysis demonstrated that treatment with tranylcypromine upregulated SESN2 expression via the autophagy *** treatment also reduced the production of NOD-like receptor family pyrin domaincontaining 3(NLRP3)*** conclusion,our results showed that tranylcypromine treatment ameliorated cochlear inflammation by promoting the expression of SESN2,which induced autophagy,thereby restricting NLRP3-related inflammasome signaling,alleviating cochlear hair cell loss,and protecting hearing *** findings suggest that inhibiting lysine-specific demethylase 1 is a

Influence of sintering temperature on the phases and photoelectric characteristics of BiOCl/ZnO composite powders

Zinc oxide is a typical functional oxide that has been widely researched for various industry applications due to its peculiar physical characteristics. However, to achieve its potential in promising applications, much work has been diligently performed to improve the physical properties of ZnO. In this work, an aqueous suspension route was used to prepare BiOCl/ZnO composite powders, and sintering processes were applied to investigate the influence of sintering temperature on the phase evolutions, microstructures, and photoelectric characteristics of BiOCl/ZnO composite powders. The results indicated that the photoelectric properties mainly depend on the relevant content of BiOCl in the composite powders and the sintering temperature. The photoelectric measurements in K_2SO_4 solutions show that the photoelectric properties of the samples with the appropriate BiOCl content(0.3 mol% and 2.0 mol%) are better than those of ZnO and commercial TiO_2(P_(25)) powders, but the photoelectric measurements in NaOH solutions indicate that the photoelectric characteristics of the as-sintered samples are only better than those of P_(25).

Interannual variability of boreal summer intraseasonal oscillation over the northwestern Pacific influenced by the Pacific Meridional Mode

During the boreal summer,intraseasonal oscillations exhibit significant interannual variations in intensity over two key regions:the central-western equatorial Pacific(5°S-5°N,150°E-150°W)and the subtropical Northwestern Pacific(10°-20°N,130°E-175°W).The former is well-documented and considered to be influenced by the ENSO,while the latter has received comparatively less attention and is likely influenced by the Pacific Meridional Mode(PMM),as suggested by partial correlation analysis *** elucidate the physical processes responsible for the enhanced(weakened)intraseasonal convection over the subtropical northwestern Pacific during warm(cold)PMM years,the authors employed a moisture budget *** findings reveal that during warm PMM years,there is an increase in summer-mean moisture over the subtropical northwestern *** increase interacts with intensified vertical motion perturbations in the region,leading to greater vertical moisture advection in the lower troposphere and consequently resulting in convective *** a process is pivotal in amplifying intraseasonal convection *** observational findings were further verified by model experiments forced by PMM-like sea surface temperature patterns.

目的探讨丹酚酸B(SalB)可能通过调控Lcn2改善非酒精性脂肪肝的作用。方法体内实验,8周龄♂C57BL/6J小鼠用普通维持饲料喂养,作为对照组,8周龄ApoE-/-小鼠用高脂饲料喂养,随机分为模型组和SalB组。在饲养8周后,SalB组小鼠用SalB灌胃,低剂量为15 mg·kg^(-1)·d^(-1),高剂量为30 mg·kg^(-1)·d^(-1),对照组和模型组用等剂量生理盐水灌胃。肝脏转录组测序结果揭示模型组和SalB组的差异表达基因,HE染色、油红O染色观察肝组织的病理变化,试剂盒检测血清和肝组织的脂质、氧化应激和炎症指标,RT-qPCR检测Lcn2、SREBP-1C以及脂质合成相关酶的mRNA水平。体外实验,用棕榈酸(PA)诱导L02和LX-2细胞24 h建立NAFLD模型,用SalB(30μmol·L^(-1))和PA(0.2 mmol·L^(-1))共孵育检测SalB体外改善NAFLD的情况。试剂盒检测L02细胞TC、TG的含量,细胞油红O染色试剂盒检测L02细胞的脂质积累,RT-qPCR检测LX-2细胞Lcn2、SREBP-1C以及脂质合成相关酶基因的mRNA水平。结果模型组血清、肝脏的生化指标异常,肝组织脂质、炎症和氧化应激水平升高,出现大面积脂肪空泡和大量的脂质沉积,PA诱导的L02细胞也出现大量的脂质积累,在SalB干预后均有明显改善。转录组测序结果表明SalB可以调控肝脏脂质代谢和炎症。肝脏和LX-2细胞mRNA水平显示模型组Lcn2、SREBP-1C以及脂质合成相关酶基因表达明显上调,SalB干预后均能降低这些基因的表达。结论丹酚酸B可以明显改善非酒精性脂肪肝,其机制可能是通过下调Lcn2、SREBP-1C和脂质合成酶的表达。

Patterned silver nanowire(AgNW)networks have been widely used as transparent electrodes in many optoelectrical ***,obvious patterning visibility and poor thermostability of AgNW are still limiting its practical ***,we report self-assembled monolayer(SAM)modulated Plateau-Rayleigh instability(PRI)of AgNW,which allows invisible patterning and superior stability of the AgNW *** opposite effects of different SAMs on the PRI are identified:the alkanethiol SAMs activate surface atom diffusion while the mercaptobenzoheterocyclic(MBH)SAMs suppress the *** degradation temperature of the AgNWs can be therefore,for the first time,tuned in the range of 193-381℃,so that the AgNW network can be patterned via PRI with a tiny optical difference between the insulative and conductive regions,i.e.,patterning ***,the MBH SAMs provide AgNW with excellent durability under thermal annealing and oxidation,which enhances the maximum heating temperature of the AgNW transparent heater by over 120℃.Beyond the micro-patterning,we consider that the developed SAM strategy can be extended to other metal nanowires for stability improvement and has huge potential in nanoengineering of one-dimensional metal materials.

Analysis of Heat Dissipation in LED with Various Adhesives

Light-emitting diodes (LEDs) are semiconducting, light producing units, with the advantages of small size, long life, low voltage, reduced energy consumption, and high vibration resistance. Unfortunately, most LED output power is converted into heat, with little converted into optical energy. If the heat is not transferred outward from the unit, the LED will overheat and its light emission efficiency, stability, and lifespan will be reduced. Improving LED heat dissipation has become a hot topic. This paper used the finite element method to discuss the temperature change in LED heat sources, using epoxy and Al2O3 adhesives with varying heat transfer coefficients.
