Lactiplantibacillus plantarum DPUL-S164 regulate AhR signaling to ameliorate DSS-induced intestinal barrier damage by producing indole-3-lactic acid in a tryptophan-rich diet

The aim of this paper was to study the effect of combination of Lactobacillus strains and tryptophan-rich diet on the intestinal barrier function of Balb/c mice exposed to a cocktail of antibiotics (ABX)and dextran sodium sulfate (DSS). Several Lactobacillus strains isolated from the healthy human fecal sample was found to utilize tryptophan(Trp) to produce indole derivatives. The results of Trp metabolism indicated that the ability of Lactobacillus to metabolize Trp to produce Indole-3-lactic Acid (ILA)， indole-3-carboxaldehyde (I3A)， and indole-3-acetic acid (IAA)varies in vitro and in vivo. The effect of Lactobacillus with high-yielding indole derivatives on disease activity index (DAI)， colon length， and intestinal permeability was significantly better than that of Lactobacillus with low-yielding indole derivatives in a high Trp diet. And Lactobacillus combined with Trp intervention also had a certain regulatory effect on the intestinal flora of male Balb/C mice. Among them， Lactiplantibacillus plantarum (L. plantarum) DPUL-S164 produced more ILA both in vivo and in vitro， and the combination of L. plantarum DPUL-S164 and Trp significantly decreased the expression level of the serum pro-inflammatory cytokine interleukin (IL)-6 and increased the expression level of the anti-inflammatory cytokine IL-10， significantly improved the number of goblet cells in the mouse mucous layer and increased mucin and tight junction protein expression. Furthermore， L. plantarum DPUL-S164 combined with Trp intervention activated the aryl hydrocarbon receptors (AhR) signaling pathway. Furthermore， we found that the expression of colonic tight junction protein was positively correlated with the expression of colonic AhR， and the expression of AhR was positively correlated with the concentrations of ILA and I3A in vivo. Therefore， we conclude that the ILA as AhR ligand produced by L. plantarum DPUL-S164 regulated the AhR pathway， thus up-regulating the expression of the tight junction protein and protecting the integrity of the epi

Alloy 690TT tube samples with different scratch depths were repaired by grinding treatments using abra-sive papers of two different particle sizes.The microstructure and stress corrosion cracking(SCC)behavior were studied in detail.During grinding,the plastic accumulation zone vulnerable to SCC was removed.Meanwhile,some residual slip steps remained in the scratched area.Corrosion tests lasting 1000,2000,3000,and 4000 h show that the sensitivity and risk of SCC in the scratched area are decreased by grind-ing.Treatment using abrasive particles of a smaller size is more effective.Nevertheless,deep scratches remained hazardous even after the grinding.

Enhanced direct gaseous CO_(2) fixation into higher bio-succinic acid production and selectivity

Utilizing CO_(2)for bio-succinic acid production is an attractive approach to achieve carbon capture and recycling(CCR)with simultaneous production of a useful platform chemical.Actinobacillus succinogenes and Basfia succiniciproducens were selected and investigated as microbial catalysts.Firstly,the type and concentration of inorganic carbon concentration and glucose concentration were evaluated.6 g C/L MgCO_(3)and 24 g C/L glucose were found to be the optimal basic operational conditions,with succinic acid production and carbon yield of over 30 g/L and over 40%,respectively.Then,for maximum gaseous CO_(2)fixation,carbonate was replaced with CO_(2)at different ratios.The“less carbonate more CO_(2)”condition of the inorganic carbon source was set as carbonate:CO_(2)=1:9(based on the mass of carbon).This condition presented the highest availability of CO_(2)by well-balanced chemical reaction equilibrium and phase equilibrium,showing the best performance with regarding CO_(2)fixation(about 15 mg C/(L·hr)),with suppressed lactic acid accumulation.According to key enzymes analysis,the ratio of phosphoenolpyruvate carboxykinase to lactic dehydrogenase was enhanced at high ratios of gaseous CO_(2),which could promote glucose conversion through the succinic acid path.To further increase gaseous CO_(2)fixation and succinic acid production and selectivity,stepwise CO_(2)addition was evaluated.50%-65%increase in inorganic carbon utilization was obtained coupled with 20%-30%increase in succinic acid selectivity.This was due to the promotion of the succinic acid branch of the glucose metabolism,while suppressing the pyruvate branch,along with the inhibition on the conversion from glucose to lactic acid.

Influence of soaking Malus domeri(Bois)Chev.leaves on gut microbiota and metabolites of long-living elderly individuals in hezhou city,Guangxi,China

To investigate the effects of drinking the soaking of Malus domeri(Bois)Chev.leaves on gut microbiota and metabolites of long-living elderly individuals in hezhou city,Guangxi,China.It has been reported that longevity is closely related to metabolism and the gut microbiota.The 16S rRNA sequencing and liquid chromatography-mass spectrometry(LC-MS)were used to analysis fecal samples and explore the factors affecting longevity in the region.Interestingly,we discovered,that elderly individuals who had been drinking the soaking of M.domeri(Bois)Chev.leaves for a long time exhibited higher diversity of the gut microbiota than without drinking the soaking,notably.The proportions of Ruminococcaceae and Prevotella were decreased in those who did not drink this soaking.In addition,a total of 106 metabolites were characterized,and the people of long-lived people(>90 years old)and elderly people(hev.leaves significantly altered the gut microbiota and upregulated levels of haplopine,farnesol,genipic acid,momordicinin,2-hydroxyestrone,hydroxyphenyllactic acid,caffeic acid,sophoraflavanone B,and soyasaponin I.We preliminarily determined that M.domeri(Bois)Chev.leaves consumption may be an important factor affecting longevity in this area.

Lateral earth pressure of granular backfills on retaining walls with expanded polystyrene geofoam inclusions under limited surcharge loading

Existing studies have focused on the behavior of the retaining wall equipped with expanded polystyrene(EPS)geofoam inclusions under semi-infinite surcharge loading rather than limited surcharge loading.In this paper,the failure mode and the earth pressure acting on the rigid retaining wall with EPS geofoam inclusions and granular backfills(henceforth referred to as EPS-wall),under limited surcharge loading are investigated through two-and three-dimensional model tests.The testing results show that different from the sliding of almost all the backfill in the EPS-wall under semi-infinite surcharge loading,only an approximately triangular backfill slides in the wall under limited surcharge loading.The distribution of the lateral earth pressure on the EPS-wall under limited surcharge loading is non-linear,and the distribution changes from the increase of the wall depth to the decrease with the increase of the limited surcharge loading.An approach based on the force equilibrium of a differential element is developed to predict the lateral earth pressure behind the EPS-wall subjected to limited surcharge loading,and its performance was fully validated by the three-dimensional model tests.

Radar Signal Intra-Pulse Modulation Recognition Based on Deep Residual Network

In view of low recognition rate of complex radar intra-pulse modulation signal type by traditional methods under low signal-to-noise ratio(SNR),the paper proposes an automatic recog-nition method of complex radar intra-pulse modulation signal type based on deep residual network.The basic principle of the recognition method is to obtain the transformation relationship between the time and frequency of complex radar intra-pulse modulation signal through short-time Fourier transform(STFT),and then design an appropriate deep residual network to extract the features of the time-frequency map and complete a variety of complex intra-pulse modulation signal type recognition.In addition,in order to improve the generalization ability of the proposed method,label smoothing and L2 regularization are introduced.The simulation results show that the proposed method has a recognition accuracy of more than 95%for complex radar intra-pulse modulation sig-nal types under low SNR(2 dB).

Fullerene-derived boron-doped defective nanocarbon for highly selective H_(2)O_(2) electrosynthesis

Electrochemical production of hydrogen peroxide(H_(2)O_(2))via the two-electron(2e-)pathway of oxygen reduction reaction(ORR)supplies an auspicious alternative to the current industrial anthraquinone process.Nonetheless,it still lacks efficient electrocatalysts to achieve high ORR activity together with 2e-selectivity simultaneously.herein,a boron-doped defective nanocarbon(B-DC)electrocatalyst is synthesized by using fullerene frameworks as the precursor and boric oxide as the boron source.The obtained B-DC materials have a hierarchical porous structure,befitting boron dopants,and abundant topological pentagon defects,exhibiting a high ORR onset potential of 0.78 V and a dominated 2e-selectivity(over 95%).Remarkably,when B-DC electrocatalyst is employed in a real device,it achieves a high H_(2)O_(2) yield rate(247 mg·L^(-1)·h^(-1)),quantitative Faraday efficiency(~100%),and ultrafast organic pollutant degradation rate.The theoretical calculation reveals that the synergistic effect of topological pentagon defects and the incorporation of boron dopants promote the activation of the O_(2) molecule and facilitates the desorption of oxygen intermediate.This finding will be very helpful for the comprehension of the synergistic effect of topological defects and heteroatom dopants for boosting the electrocatalytic performance of nanocarbon toward H_(2)O_(2) production.

Bailing capsule(百令胶囊) alleviates autoimmune thyroiditis via regulating peroxisome proliferator-activated receptor signaling pathway: a multi-omics analysis

OBJECTIVE: To investigate the efficacy and potential mechanism of Bailing capsule (百令胶囊， BL) anti-autoimmune thyroiditis (AIT). METHODS: Based on the AIT rat model， the effect of BL in alleviating AIT was evaluated by detecting serum thyroid index free triiodothyronine (FT3)， free thyroxine (FT4)， thyroid-stimulating hormone (TSH)， thyroglobulin antibody (TGAb)， thyroid peroxidase antibody (TPOAb)， and inflammatory factors Interferon-gamma (IFN-γ)， Interleukin-4， -10， and -12 (IL-4， IL-10， and IL-12) as well as thyroid tissue hematoxylin-eosin (he) staining and ultrastructure observation. The mechanism of BL was explored by combining transcriptome and proteome analysis， and further verified by Western blot (WB). RESULTS: BL effectively reduced serum FT3， FT4， TGAb， and TPOAb levels in AIT rats， restored TSH balance， inhibited the release of pro-inflammatory cytokines IFN-γ and IL-12， promoted the production of anti-inflammatory cytokines IL-4 and IL-10， and significantly reduced IFN-γ/IL-4 and IL-12/IL-10， improved thyroid follicular structure， and protected thyroid tissue from injury. Kyoto Encyclopedia of Genes and Genomes and protein interaction network analysis showed that BL affected the expression of fatty acid-binding protein 4， acyl-CoA synthetase long-chain family member 1， and acyl-CoA dehydrogenase long chain to regulate the peroxisome proliferator-activated receptor signaling pathway， thereby inhibiting the fatty acid metabolism and the inflammatory state of AIT rats. CONCLUSIONS: BL could effectively reduce thyroid inflammation in AIT model rats. The possible BL mechanism was to regulate the peroxisome proliferator-activated receptor signaling pathway and inhibit fatty acid metabolism. This study suggested that BL has the potential to be used in clinical treatment of AIT.

Improvement of Lactiplantibacillus plantarum MWFLp-182 on oxidative deficits induced by in 2，2'-azobis(2-methylpropionamidine) dihydrochloride and the relating key gene analysis

Lactiplantibacillus plantarum MWFLp-182 was originally isolated from the feces of long-living elderly individuals in hezhou city， Guangxi， China. L. plantarum MWFLp-182 showed favorable gastric and intestinal tolerance compared with Lactiplantibacillus rhamnosus GG (LGG). L. plantarum MWFLp-182 was further examined for its antioxidant and anti-inflammatory activity in ABAP-treated HT-29 cells. Importantly， it enhanced the expression of anti-inflammatory factor (interleukin-10 (IL-10))， antioxidant cytokines (superoxide dismutase (SOD)， glutathione peroxidase (GPx)， nuclear factor erythroid 2-related factor (Nrf2)， and hemeoxygenase 1(HO-1))， B-cell lymphoma-2 (Bcl-2)， and tight junction (Zonula occludens protein 1(ZO-1)， Claudin-1， and Occludin) proteins and genes， and reduced reactive oxygen species (ROS). The complete genome of L. plantarum MWFLp-182 contained a single circular chromosome that was 3 257 196 bp long， with a G + C content of 44.49%， and a single circular plasmid that was 53 560 bp long. The genes tpx， trxA， trxB， npr， nrdH， dps， recA， gpx， gshA and arsC in this strain were related to antioxidant function; the key genes involved in antioxidant function were further investigated. In this study， we identified a probiotic candidate with antioxidant properties.

随着网络信息技术在战争中的使用,以非直接接触的、非暴力手段为主的非传统军事对抗形式在现代战争中运用越来越多。由此,现代战争表现出非常鲜明的混合战争特点,战争不再仅仅局限于实体战场中以军事手段展开的对抗与冲突,在实体战场之外出现了许多与实体战场同时展开的非传统军事对抗与冲突,对战争进程产生着重要影响。在这种综合运用军事手段与非军事手段的混合战争新形态中,网络舆论战成为不可忽视的重要因素。网络舆论战在政治、经济、外交、军事、信息等多个战场空间中扮演着至关重要的角色,已经成为混合战争不可或缺的重要组成部分。网络舆论战结合了传统的心理战和现代的信息战,是混合战争中使用非传统军事手段的典型表现形式,其主要特征表现为:空间的虚拟性、攻击主体的匿名性和参与主体的广泛性。网络舆论战通过控制社交媒体、传播虚假信息、使用网络情报战等手段,力图重塑意识形态、操纵战场信息、左右民意归属,极为深刻地影响着整个战争的进程。当前,充分把握网络舆论战的特征和影响已经成为深刻认识混合战争的重要问题。
