Establishment of human cerebral organoid systems to model early neural development and assess the central neurotoxicity of environmental toxins

Human brain development is a complex process,and animal models often have significant limitations.To address this,researchers have developed pluripotent stem cell-derived three-dimensional structures,known as brain-like organoids,to more accurately model early human brain development and disease.To enable more consistent and intuitive reproduction of early brain development,in this study,we incorporated forebrain organoid culture technology into the traditional unguided method of brain organoid culture.This involved embedding organoids in matrigel for only 7 days during the rapid expansion phase of the neural epithelium and then removing them from the matrigel for further cultivation,resulting in a new type of human brain organoid system.This cerebral organoid system replicated the temporospatial characteristics of early human brain development,including neuroepithelium derivation,neural progenitor cell production and maintenance,neuron differentiation and migration,and cortical layer patterning and formation,providing more consistent and reproducible organoids for developmental modeling and toxicology testing.As a proof of concept,we applied the heavy metal cadmium to this newly improved organoid system to test whether it could be used to evaluate the neurotoxicity of environmental toxins.Brain organoids exposed to cadmium for 7 or 14 days manifested severe damage and abnormalities in their neurodevelopmental patterns,including bursts of cortical cell death and premature differentiation.Cadmium exposure caused progressive depletion of neural progenitor cells and loss of organoid integrity,accompanied by compensatory cell proliferation at ectopic locations.The convenience,flexibility,and controllability of this newly developed organoid platform make it a powerful and affordable alternative to animal models for use in neurodevelopmental,neurological,and neurotoxicological studies.

Solid particle size characterization by a high-frequency collision response in pneumatic particulate flow

A novel triaxial vibration method is developed for the real-time characterization of the solid particle size distribution(PsD)in pneumatic particulate flow,which is critical for chemical industry.In this work,the particle-wall collision and friction behaviours were analysed by the time-domain statistical and timefrequency joint methods to narrow the high-frequency response range by the initial experiment of free fall for a single particle,interparticle,and multiple particles.Subsequently,verification experiments of PSD characterization in pneumatic flow were performed.First,the quantitative triaxial energy response model that considers the particle size,shape,and mass factors were established.Second,a good agreement of the particle number identification was found between the triaxial vibration energy and mean particle size of 150-550μm.Moreover,the performance with the best accuracy was focused on a range of 42-43 kHz in the x-axis and z-axis and 36.8-38.8 kHz in the y-axis.Finally,the individual particle energy was inversely analysed by the triaxial vibration response within the optimized frequency bands,and the PSD was characterized in real-time by a low error rate,that is,5.2% from the XZ-axis direction of sand(42-43 kHz)and 5.6% from the XYZ-axis of glass(30.9-33.9 kHz,46.2-47.2 kHz,38.3-41.3 kHz for each axis response).Therefore,this research complements the existing approaches for PsD characterization in particulate multiphase flow.

Increased long-distance and homo-trans interactions related to H3K27me3 in Arabidopsis hybrids

In plants,the genome structure of hybrids changes compared with their parents,but the effects of these changes in hybrids remain elusive.Comparing reciprocal crosses between Col×C24 and C24×Col in Arabidopsis using high-throughput chromosome conformation capture assay(Hi-C)analysis,we found that hybrid three-dimensional(3D)chromatin organization had more long-distance interactions relative to parents,and this was mainly located in promoter regions and enriched in genes with heterosis-related pathways.The interactions between euchromatin and heterochromatin were increased,and the compartment strength decreased in hybrids.In compartment domain(CD)boundaries,the distal interactions were more in hybrids than their parents.In the hybrids of CURLY LEAF(clf)mutants clfCol×clfC24and clfC24×clfCol,the heterosis phenotype was damaged,and the long-distance interactions in hybrids were fewer than in their parents with lower H3K27me3.ChIP-seq data revealed higher levels of H3K27me3 in the region adjacent to the CD boundary and the same interactional homo-trans sites in the wild-type(WT)hybrids,which may have led to more long-distance interactions.In addition,the differentially expressed genes(DEGs)located in the boundaries of CDs and loop regions changed obviously in WT,and the functional enrichment for DEGs was different between WT and clf in the longdistance interactions and loop regions.Our findings may therefore propose a new epigenetic explanation of heterosis in the Arabidopsis hybrids and provide new insights into crop breeding and yield increase.

Is clustered seismicity an indicator of regional stress?Insights from earthquake sequences in Yongning-Luguhu faulted basin,Southwest China

Using hypocenter relocation,moment tensor inversion,stress field inversion,and fault slip tendency analysis,this study systematically investigated three M5.5-5.8 earthquake sequences that occurred after 2000 in the Yongning-Luguhu faulted basin in the middle of the Lijiang-Xiaojinhe fault zone within the Sichuan-Yunnan block,Southwest China.Our results show that since the 2008 Wenchuan earthquake,the tectonic stress pattern in this area may have changed and that b-values estimated for the earthquake sequences show evidence of an increasing trend in stress in the study area.Seismicity in the small-scale faulted basin adjacent to the large-scale fault zone is a possible indicator of regional stress.We also note that the aftershocks of the M5.7 earthquake sequence in 2012 and the M5.5 earthquake sequence in 2022 show relatively clear fluid diffusion-triggering characteristics.Overpressure of deep fluids is still the main factor driving seismic activity in the region,and we propose that the background tectonic stresses have not yet reached critical levels.

Advanced charge transfer technology for highly efficient and long-lived TADF-type organic afterglow with near-infrared light-excitable property

Heavy atom effects and n-π*transitions have been frequently reported to enhance room-temperature organic phosphorescence efficiency but lead to shortage of phosphorescence lifetimes.Unlike these reported studies,we conceive the incorporation of advanced charge transfer(CT)technology to boost room-temperature organic afterglow efficiency and simultaneously maintain afterglow lifetimes.Here we design difluoroboronβ-diketonate(BF2bdk)CT compounds with moderate singlet-triplet splitting energy(ΔEST)of around 0.4 e V,and relatively large spin-orbit coupling matrix elements(SOCME(S_(1)-T_(1)),1–10 cm^(-1))to achieve efficient intersystem crossing(ISC)and moderate rates of reverse intersystem crossing(kRISC,1–10 s^(-1)).The advanced CT technology,which includes multiple electron-donating groups and orthogonal donor-acceptor arrangement,have been found to narrowΔESTand enhance both ISC and RISC.Meanwhile,the organic matrices suppress nonradiative decay of BF2bdk’s T1states by their rigid microenvironment.Consequently,thermally activated delayed fluorescence(TADF)-type organic afterglow materials can be achieved with afterglow efficiency up to 83.0%,long lifetimes of 433 ms,excellent processablility,as well as advanced anti-counterfeiting and information encryption.Furthermore,with the aid of up-conversion materials and through radiative energy transfer,TADF-type afterglow materials with aqueous dispersity and near-infrared light-excitable property have been achieved,which paves the way for biomedical applications.

Changes in milk fat globule membrane proteins along lactation stage of Laoshan dairy goat

The milk fat globule membrane(MFGM)is a complex structure with numerous functions,and its composition is affected by many factors.There have been few systematic investigations on goat MFGM proteome profiling during lactation.Individual milk samples from 15 healthy dairy goats were obtained at six lactation time points for investigation of the MFGM proteome using both data-independent acquisition(DIA)and data-dependent acquisition(DDA)proteomics techniques combined with multivariate statistical analysis.Using the DIA method,890 variably abundant MFGM proteins were discovered throughout the lactation cycle.From 1 to 240 d,butyrophilin subfamily 1 member A1,lipoprotein lipase,perilipin-2,and adipose triglyceride lipase were upregulated,while APOE,complement C3,clusterin,and IgG were downregulated.Furthermore,from 1 to 90 d,annexin A1,annexin A2,and antithrombin-ll were downregulated,then upregulated by d 240.Albumin had a high degree of connectedness,indicating that it was a key protein,according to protein-protein interaction research.Overall,our findings gave new insights into the biological features of MFGM protein in goat milk throughout lactation,which may aid in the creation of specialized MFGM products and infant formula.

Mechanical properties and interfacial characteristics of 6061 Al alloy plates fabricated by hot-roll bonding

This work aims to investigate the mechanical properties and interfacial characteristics of 6061 Al alloy plates fabricated by hotroll bonding (HRB) based on friction stir welding. The results showed that ultimate tensile strength and total elongation of the hot-rolled and aged joints increased with the packaging vacuum, and the tensile specimens fractured at the matrix after exceeding 1 Pa. Non-equilibrium grain boundaries were formed at the hot-rolled interface, and a large amount of Mg2Si particles were linearly precipitated along the interfacial grain boundaries (IGBs). During subsequent heat treatment, Mg2Si particles dissolved back into the matrix, and Al2O3 film remaining at the interface eventually evolved into MgO. In addition, the local IGBs underwent staged elimination during HRB, which facilitated the interface healing due to the fusion of grains at the interface. This process was achieved by the dissociation, emission, and annihilation of dislocations on the IGBs.

Endowing flexible and adaptable fiber devices with light-emitting capabilities has the potential to revolutionize the current design philosophy of intelligent,wearable interactive devices.However,significant challenges remain in developing fiber devices when it comes to achieving uniform and customizable light effects while utilizing lightweight hardware.Here,we introduce a mass-produced,wearable,and interactive photochromic fiber that provides uniform multicolored light control.We designed independent waveguides inside the fiber to maintain total internal reflection of light as it traverses the fiber.The impact of excessive light leakage on the overall illuminance can be reduced by utilizing the saturable absorption effect of fluorescent materials to ensure light emission uniformity along the transmission direction.In addition,we coupled various fluorescent composite materials inside the fiber to achieve artificially controllable spectral radiation of multiple color systems in a single fiber.We prepared fibers on mass-produced kilometer-long using the thermal drawing method.The fibers can be directly integrated into daily wearable devices or clothing in various patterns and combined with other signal input components to control and display patterns as needed.This work provides a new perspective and inspiration to the existing field of fiber display interaction,paving the way for future human–machine integration.

In-situ Horizontal Extrusion Test of Herbaceous Root-Soil with Different Root Types

The influence of different types of roots on the soil is complex and still remains unclear.Four in-situ extrusion tests were conducted on two types of root systems,namely fibrous and tap root system,for three plants,Eleusine indica,Potentilla anserine,and Artemisia argyi,according to the classification in Botany,and the thrust-displacement curves and failure patterns of different samples were analysed by comparison to fill the aforementioned gap.Results reveal that the roots can reduce the characteristics of soil brittleness and enhance its capability to resist large deformation,and different root types contribute different effects to the strain-hardening behavior of the root-soil mass.The contribution of the fibrous root system to strength is limited,whilst the tap root system substantially enhances strength and stiffness.Results of failure patterns show that fibrous and tap root systems affect soil solidification and surface cracking reduction.However,the effect of the tap root system depends on the composition of lateral and tap roots:long and rich lateral roots are effective for resisting the creation of cracks,but thick tap roots with few and thin lateral roots may lead to several surface cracks.

随着基础教育评价改革的全面深化,过程性评价受到越来越多的重视,然而有关过程性评价对学生学业成绩的实际影响如何仍存在不同看法。为深入探究过程性评价对学习效果的影响,采用元分析方法对已有63项相关主题的实验和准实验研究进行系统定量分析。结果显示,随机效应模型得到的合并效应值为0.964,表明过程性评价对提高学生学业成绩具有显著的正面影响;此外,不同国别、学科、学段和评价方式对学业成绩的影响存在显著差异,不同实验时长的影响差异不显著。今后应注重过程性评价在课堂教学中的有效实施策略,加强对教师和学生评价素养的培养,以推动过程性评价在教育实践中的广泛应用。
