Research on Condenser Deterioration Evolution Trend Based on ANP-EWM Fusion Health Degree

This study presents a proposed method for assessing the condition and predicting the future status of condensers operating in seawater over an extended period.The aim is to address the problems of scaling and corrosion,which lead to increased loss of cold resources.The method involves utilising a set of multivariate feature parameters associated with the condenser as input for evaluation and trend prediction.This methodology offers a precise means of determining the optimal timing for condenser cleaning,with the ultimate goal of improving its overall performance.The proposed approach involves the integration of the analytic network process with subjective expert experience and the entropy weightmethod with objective big data analysis to develop a fusion health degreemodel.The mathematical model is constructed quantitatively using the improved Mahalanobis distance.Furthermore,a comprehensive prediction model is developed by integrating the improved Informer model and Markov error correction.This model takes into account the health status of the equipment and several influencing factors,includingmultivariate feature characteristics.This model facilitates the objective examination and prediction of the progression of equipment deterioration trends.The present study involves the computation and verification of the field time series data,which serves to demonstrate the accuracy of the condenser health-related models proposed in this research.These models effectively depict the real condition and temporal variations of the equipment,thus offering a valuable method for determining the precise cleaning time required for the condenser.

Studies have suggested that the nucleus accumbens(NAc)is implicated in the pathophysiology of major depression;however,the regulatory strategy that targets the NAc to achieve an exclusive and outstanding anti-depression benefit has not been elucidated.Here,we identified a specific reduction of cyclic adenosine monophosphate(cAMP)in the subset of dopamine D1 receptor medium spiny neurons(D1-MSNs)in the NAc that promoted stress susceptibility,while the stimulation of cAMP production in NAc D1-MSNs efficiently rescued depression-like behaviors.Ketamine treatment enhanced cAMP both in D1-MSNs and dopamine D2 receptor medium spiny neurons(D2-MSNs)of depressed mice,however,the rapid antidepressant effect of ketamine solely depended on elevating cAMP in NAc D1-MSNs.We discovered that a higher dose of crocin markedly increased cAMP in the NAc and consistently relieved depression 24 h after oral administration,but not a lower dose.The fast onset property of crocin was verified through multicenter studies.Moreover,crocin specifically targeted at D1-MSN cAMP signaling in the NAc to relieve depression and had no effect on D2-MSN.These findings characterize a new strategy to achieve an exclusive and outstanding anti-depression benefit by elevating cAMP in D1-MSNs in the NAc,and provide a potential rapid antidepressant drug candidate,crocin.

Cancer immunotherapy has garnered promise in tumor progression, invasion, and metastasis through establishing durable and memorable immunological activity. However, low response rates, adverse side effects, and high costs compromise the additional benefits for patients treated with current chemical and biological agents. Chinese herbal medicines (CHMs) are a potential treasure trove of natural medicines and are gaining momentum in cancer immunomodulation with multi-component, multi-target, and multi-pathway characteristics. The active ingredient extracted from CHMs benefit generalized patients through modulating immune response mechanisms. Additionally, the introduction of nanotechnology has greatly improved the pharmacological qualities of active ingredients through increasing the hydrophilicity, stability, permeability, and targeting characteristics, further enhancing anti-cancer immunity. In this review, we summarize the mechanism of active ingredients for cancer immunomodulation, highlight nano-formulated deliveries of active ingredients for cancer immunotherapy, and provide insights into the future applications in the emerging field of nano-formulated active ingredients of CHMs.

SIRT1 activation synergizes with FXR agonism in hepatoprotection via governing nucleocytoplasmic shuttling and degradation of FXR

Farnesoid X receptor(FXR)is widely accepted as a promising target for various liver diseases;however,panels of ligands in drug development show limited clinical benefits,without a clear mechanism.Here,we reveal that acetylation initiates and orchestrates FXR nucleocytoplasmic shuttling and then enhances degradation by the cytosolic E3 ligase CHIP under conditions of liver injury,which represents the major culprit that limits the clinical benefits of FXR agonists against liver diseases.Upon inflammatory and apoptotic stimulation,enhanced FXR acetylation at K217,closed to the nuclear location signal,blocks its recognition by importin KPNA3,thereby preventing its nuclear import.Concomitantly,reduced phosphorylation at T442 within the nuclear export signals promotes its recognition by exportin CRM1,and thereby facilitating FXR export to the cytosol.Acetylation governs nucleocytoplasmic shuttling of FXR,resulting in enhanced cytosolic retention of FXR that is amenable to degradation by CHIP.SIRT1 activators reduce FXR acetylation and prevent its cytosolic degradation.More importantly,SIRT1 activators synergize with FXR agonists in combating acute and chronic liver injuries.In conclusion,these findings innovate a promising strategy to develop therapeutics against liver diseases by combining SIRT1 activators and FXR agonists.

Targeting tumor endothelial hyperglycolysis enhances immunotherapy through remodeling tumor microenvironment

Vascular abnormality is a hallmark of most solid tumors and facilitates immune evasion.Targeting the abnormal metabolism of tumor endothelial cells(TECs)may provide an opportunity to improve the outcome of immunotherapy.Here,in comparison to vascular endothelial cells from adjacent peritumoral tissues in patients with colorectal cancer(CRC),TECs presented enhanced glycolysis with higher glyceraldehyde-3-phosphate dehydrogenase(GAPDH)expression.Then an unbiased screening identified that osimertinib could modify the GAPDH and thus inhibit its activity in TECs.Low-dose osimertinib treatment caused tumor regression with vascular normalization and increased infiltration of immune effector cells in tumor,which was due to the reduced secretion of lactate from TECs by osimertinib through the inhibition of GAPDH.Moreover,osimertinib and anti-PD-1 blockade synergistically retarded tumor growth.This study provides a potential strategy to enhance immunotherapy by targeting the abnormal metabolism of TECs.

死亡问题不仅影响着布朗肖的批评写作,还渗透了他的叙述作品。布朗肖的死亡叙述呈现了一种特殊的死亡经验,是西方文学中招魂传统的延续和变奏。在布朗肖的叙述中,死亡经验有两种基本形式:复活与幸存。但《黑暗托马》和《死亡判决》讲述的复活本质上都是死亡的复活而非生命的复活,并且幸存也在死亡的判定和死亡的停止之间表现出模糊性。到了《我死亡的瞬间》,这样的幸存还被德里达解读为一种存留的方式,它把死亡变成了“死”。通过创造虚构的空间来回应死亡事件的幽灵维度,布朗肖的叙述承担了见证的使命。

Schisandra lignans ameliorate nonalcoholic steatohepatitis by regulating aberrant metabolism of phosphatidylethanolamines

Nonalcoholic steatohepatitis(NASH)is a spectrum of chronic liver disease characterized by hepatic lipid metabolism disorder.Recent reports emphasized the contribution of triglyceride and diglyceride accumulation to NASH,while the other lipids associated with the NASH pathogenesis remained unexplored.The specific purpose of our study was to explore a novel pathogenesis and treatment strategy of NASH via profiling the metabolic characteristics of lipids.Herein,multi-omics techniques based on LC—Q-TOF/MS,LC—MS/MS and MS imaging were developed and used to screen the action targets related to NASH progress and treatment.A methionine and choline deficient(MCD)diet-induced mouse model of NASH was then constructed,and Schisandra lignans extract(SLE)was applied to alleviate hepatic damage by regulating the lipid metabolism-related enzymes CES2A and CYP4A14.Hepatic lipidomics indicated that MCD-diet led to aberrant accumulation of phosphatidylethanolamines(PEs),and SLE could significantly reduce the accumulation of intrahepatic PEs.Notably,exogenous PE(18:0/18:1)was proved to significantly aggravate the mitochondrial damage and hepatocyte apoptosis.Supplementing PE(18:0/18:1)also deteriorated the NASH progress by up regulating intrahepatic proinflammatory and fibrotic factors,while PE synthase inhibitor exerted a prominent hepatoprotective role.The current work provides new insights into the relationship between PE metabolism and the pathogenesis of NASH.

The role of phosphatidylcholine 34:1 in the occurrence,development and treatment of ulcerative colitis

Lipid homeostasis is considered to be related to intestinal metabolic balance,while its role in the pathogenesis and treatment of ulcerative colitis(UC)remains largely unexplored.The present study aimed to identify the target lipids related to the occurrence,development and treatment of UC by comparing the lipidomics of UC patients,mice and colonic organoids with the corresponding healthy controls.Here,multi-dimensional lipidomics based on LC–QTOF/MS,LC–MS/MS and iMScope systems were constructed and used to decipher the alteration of lipidomic profiles.The results indicated that UC patients and mice were often accompanied by dysregulation of lipid homeostasis,in which triglycerides and phosphatidylcholines were significantly reduced.Notably,phosphatidylcholine 34:1(PC34:1)was characterized by high abundance and closely correlation with UC disease.Our results also revealed that down-regulation of PC synthase PCYT1 α and Pemt caused by UC modeling was the main factor leading to the reduction of PC34:1,and exogenous PC34:1 could greatly enhance the fumarate level via inhibiting the transformation of glutamate to N-acetylglutamate,thus exerting an anti-UC effect.Collectively,our study not only supplies common technologies and strategies for exploring lipid metabolism in mammals,but also provides opportunities for the discovery of therapeutic agents and biomarkers of UC.

[目的]农业社会化服务是解决小农户与大市场有效对接、加速融入现代农业产业体系生产体系经营体系、提升粮食安全保障能力的重要手段。对北大荒农业社会化服务能力对粮食安全影响进行评价分析,提出提升北大荒社会化服务水平和粮食安全保障能力、完善我国发展农业社会化服务政策建议。[方法]文章以北大荒农业社会化服务能力为研究对象,根据粮食安全本质要求和北大荒集团发展战略,基于粮食安全视角构建了北大荒农业社会化服务能力评价指标体系,主要由目标层、准则层(5项)、因子层(17项)、指标层(71项)构成,采用综合模糊评价模型对其评价分析。[结果](1)农业科技服务能力较强,主要体现在种业科研投入强度居于全国前列、农业科技进步贡献率较高、农技应用推广体系完备;(2)生产经营服务能力较强,主要体现在农业生产资料统一供应服务、标准化规模化数字化粮食生产能力和较强的粮食加工转化能力;(3)粮安保障服务能力较强,主要体现在金融保险服务总体完善、农机与基础设施服务健全、数字农服覆盖率高和航化作业服务能力较强、农业绿色发展服务方面突出、收入保障能力较强;(4)粮食收储分销能力中等偏上,但仓储能力和市场交易服务偏弱;(5)农业科技服务能力很强,但科技研发投入及研发人力资源建设偏弱;(6)品牌价值及认证服务水平中等偏上,但品牌认证服务能力一般。[结论]根据综合模糊评价结果,目前北大荒集团农业社会化服务能力处于较强水平,但在科技研发、仓储等基础设施和品牌认证服务能力等方面尚需改进提升。总体上,北大荒集团农业社会化服务对落实我国千亿斤(1斤=0.5 kg)粮食产能提升行动,促进农业节本增效、提质增效、营销增效,带动农民增收、农村经济组织增效,尤其对我国粮食主产区和重要农产品规模种植区发展农业社会化服务具有示范推广的实践价值。
