Hand osteoarthritis is a common heterogeneous joint disorder with unclear molecular mechanisms and no disease-modifying *** this study,we performed single-cell RNA sequencing analysis to compare the cellular composition and subpopulationspecific gene expression between cartilage with macroscopically confirmed osteoarthritis(n=5)and cartilage without osteoarthritis(n=5)from the interphalangeal joints of five *** 105142 cells,we identified 13 subpopulations,including a novel subpopulation with inflammation-modulating potential annotated as inflammatory *** chondrocytes exhibited extensive alteration of gene expression patterns in osteoarthritic cartilage compared with nonosteoarthritic *** inflammatory chondrocytes and fibrocartilage chondrocytes showed a trend toward increased numbers in osteoarthritic *** these two subpopulations from osteoarthritic cartilage,the ferroptosis pathway was enriched,and expression of iron overload-related genes,e.g.,FTH1,was *** verify these findings,we conducted a Mendelian randomization study using UK Biobank and a population-based cross-sectional study using data collected from Xiangya Osteoarthritis *** predisposition toward higher expression of FTH1 mRNA significantly increased the risk of hand osteoarthritis(odds ratio=1.07,95%confidence interval:1.02–1.11)among participants(n=332668)in UK *** levels of serum ferritin(encoded by FTH1),a biomarker of body iron overload,were significantly associated with a high prevalence of hand osteoarthritis among participants(n=1241)of Xiangya Osteoarthritis Study(P-for-trend=0.037).In conclusion,our findings indicate that inflammatory and fibrocartilage chondrocytes are key subpopulations and that ferroptosis may be a key pathway in hand osteoarthritis,providing new insights into the pathophysiology and potential therapeutic targets of hand osteoarthritis.

Objectives: Bacillus cereus isolates from vegetables can cause diarrheal syndrome food poisoning. Lettuce, the most highly consumed ready-to-eat vegetable, poses a significant risk of B. cereus exposure. Hence, B. cereus monitoring and evaluation during cultivation are essential for preventing foodborne illnesses caused by this food-pathogen combination. However, the impact of different cultivation practices on B. cereus contamination in lettuce currently remains unclear. Furthermore, a validity indicator for the experimental models cannot be effectively validated under field conditions. Materials and Methods: Herein, we assessed the risk of B. cereus exposure using lettuce grown under two different cultivation farming systems in China (open-field versus greenhouse) using a quantitative microbial risk assessment model. Tukey's mean comparison method and kernel density estimation frequency distribution statistics were used for model validation using environmental samples and lettuce. Results: The mean concentration of B. cereus in open-field-grown lettuce was significantly higher than that in greenhouse-grown lettuce (P0.05), verifying the accuracy of the risk model. Conclusion: Soil solarization, complete manure treatment, usage of mulch films, and irrigation water quality should be considered in agricultural practices to control B. cereus concentrations in lettuce. Overall, we found that B. cereus was more abundant in open-field-grown lettuce, and we provide a risk evaluation framework that can guide farmers and policy-makers to control B. cereus contamination in farm-to-fork lettuce chains in China.

Evaluation of the performance of mold powder for continuous casting of plate slabs

The No. 3 slab caster,which mainly provides slabs to the 5000 mm plate mill at Baosteel, was put into production in December,2004. The size of the biggest slab produced by this caster is 2300 mm in width and 300 mm in thickness. The designed output of the caster is 2.3 Mt/a. Slab surface longitudinal crack defects,which were related to the heat flux of the mold, frequently occurred in the early stage of the startup of the caster. As mild cooling powder is beneficial to the uniformity of the shell of initial slabs ,the concentration of stress is reduced, and the longitudinal cracking on the surface is avoided. This study evaluates the performance of several kinds of powder, and the results show that mold powder of high basicity, high crystallization proportion and low heat flux is to the benefit of the reduction of the longitudinal cracks on the surface and the defects of slabs.

The ecological adaptability of cloned sheep to free-grazing in the Tengger Desert of Inner Mongolia,China

Since the birth of the first cloned sheep,somatic cell nuclear transfer technology has been successfully used to clone a variety of *** livestock have no apparent health risks,and the quality and safety of the cloned animal products are similar to non-cloned *** social behavior and environmental adaptability of postnatal cloned animals,especially when used for grassland farm production purposes,is *** the present study,the cloned Dorper sheep equipped with GPS location devices were free-grazed in a harsh natural environment similar to conditions commonly experienced by Mongolian *** main findings of this research were as follows.(1)Under free-grazing conditions,the cloned sheep showed excellent climatic and ecological *** extreme temperature conditions ranging from–30 to 40°C,the cloned sheep maintained acceptable body condition and behaved as other sheep.(2)The cloned sheep quickly adapted from a herd feeding strategy to the harsh environment and quickly exhibited a grazing regimen as other free-grazing sheep.(3)The cloned sheep exhibited free-grazing patterns and social behavior as other sheep.(4)The cloned sheep in the harsh environment thrived and produced healthy ***,the cloned Dorper sheep exhibited excellent ecological adaptation,which is an important consideration for breeding meat sheep by *** Dorper sheep readily adapted to the free-grazing conditions on the Mongolian plateau grassland,which attests to their ability to withstand harsh environmental conditions.

With the development of Chinese dairy industry cluster, the competition of raw milk resource is getting keen between the dairy processing enterprises in cluster. Because the investment of raw milk foundation has the characteristic of public goods, dairy processing enterprises in the cluster can all benefit from the output, whether it makes investment or not, so the investment of raw milk resource are not efficient in economics. Game equilibrium of Boxed pigs is the common result of enterprises competition in low cost cluster. In this paper, the boxed pigs’ game model is established to analyze the investment behavior of local and extraneous dairy processing enterprises. The analysis of the mode not only explains the reasons of boxed pigs’ equilibrium, but also gives guidance for the improvement of the game equilibrium. Finally, the case of Heilongjiang province dairy industry cluster is studied. It is also shown that establishing self-owned pastures and building long term contractual relations with scaled dairy farms or dairy production associations can improve the game result, so as to reach the cooperation equilibrium.

Multiplex PCR Assay Establishment for Detection of H5,H9 Subtypes Avian Influenza Virus and Duck Tembusu Virus

The paper aimed to establish a rapid multiplex PCR assay for detection of 1-15, H9 subtype avian influenza virus （VIA） and duck Tembusu virus （DT- MUV）. According to the HA gene sequences of I-L5 and H9 AIV and NS5 gene of DTMUV in C, enBank, three sets of primer specific to these three kinds of viruses were designed, respectively. The multiplex PCR for simultaneous detection of AIV （ HS, H9 ） and DTMUV was established, and was applied in clinical. The speci- ficity and sensitivity of the multiplex PCR were tested. The results showed that the assay could specifically amplify the gene fragments of I-IS AIV （380 bp）, H9 AIV （ 732 bp）, and DTMUV （250 bp ）, and were all negative for other duck viruses. This method showed the sensitivity of 533 pg/μL for H5 MV, 56 pg/μL for I-I9, and 6.6 ng/μL for DTMUV. Additionally, the detection results of 200 clinical samples indicated that this multiplex PCR method was a rapid, sensitive and svecific tool for clinical sample detection.
