Huanglian-Renshen-Decoction Maintains Isletβ-Cell Identity in T2DM Mice through Regulating GLP-1 and GLP-1R in Both Islet and Intestine

Objective: To elucidate the effect of Huanglian-Renshen-Decoction(HRD) on ameliorating type 2 diabetes mellitus by maintaining islet β-cell identity through regulating paracrine and endocrine glucagon-like peptide-1(GLP-1)/GLP-1 receptor(GLP-1R) in both islet and intestine. Methods: The db/db mice were divided into the model(distilled water), low-dose HRD(LHRD, 3 g/kg), high-dose HRD(HHRD, 6 g/kg), and liraglutide(400 μg/kg) groups using a random number table, 8 mice in each group. The db/m mice were used as the control group(n=8, distilled water). The entire treatment of mice lasted for 6 weeks. Blood insulin, glucose, and GLP-1levels were quantified using enzyme-linked immunosorbent assay kits. The proliferation and apoptosis factors of islet cells were determined by immunohistochemistry(IHC) and immunofluorescence(IF) staining. Then,GLP-1, GLP-1R, prohormone convertase 1/3(PC1/3), PC2, v-maf musculoaponeurotic fibrosarcoma oncogene homologue A(MafA), and pancreatic and duodenal homeobox 1(PDX1) were detected by Western blot, IHC,IF, and real-time quantitative polymerase chain reaction, respectively. Results: HRD reduced the weight and blood glucose of the db/db mice, and improved insulin sensitivity at the same time(P<0.05 or P<0.01). HRD also promoted mice to secrete more insulin and less glucagon(P<0.05 or P<0.01). Moreover, it also increased the number of islet β cell and decreased islet α cell mass(P<0.01). After HRD treatment, the levels of GLP-1,GLP-1R, PC1/3, PC2, MafA, and PDX1 in the pancreas and intestine significantly increased(P<0.05 or P<0.01).Conclusion: HRD can maintain the normal function and identity of islet β cell, and the underlying mechanism is related to promoting the paracrine and endocrine activation of GLP-1 in pancreas and intestine.

基于深圳北站广场向下增建城际铁路地下车站结构的实际工程背景,场地土中地下车站附带上部已建结构的地震反应特征、相互作用机理及其破坏机制成为结构抗震性能评价的关键问题。鉴于此,采用整体动力时程分析方法建立了土-地下结构-上部结构静、动力耦合非线性动力相互作用的三维精细化数值模型,系统地研究了地下车站附带上部结构、单建地下车站结构、单建地上结构的地震响应规律及其损伤演变过程。结果表明:相比单建地下车站结构,由于受上部结构影响,增建地下车站结构的水平侧向力及其层间位移角均显著增大,且顶、底板地震损伤面积及程度明显加剧;相比单建地上结构,由于受地下车站结构影响,增建地下车站结构的上部结构的变形协调性显著降低,其地震损伤较单建地上结构严重,尤其在中柱与二层楼板连接处。研究成果可为既有上部结构向下增层拓建的科学设计、防震减灾提供支撑和借鉴。

Electroacupuncture enhances the mitophagy of granulosa cells in premature ovarian insufficiency model mice by inactivating the hippo-yes-associated protein/transcriptional co-activator with postsynaptic density protein,drosophila disc large tumor suppresso

OBJECTIVE: To investigate the potential mechanism of electroacupuncture（EA） in alleviating premature ovarian insufficiency（POI） and to provide a theoretical basis for EA treatment of POI. METHODS: For this purpose, a POI mice model was developed by injecting 12 mg/kg busulfan and 120 mg/kg cyclophosphamide intraperitoneally to induce POI. It was then proceeded by EA intervention at Guanyuan（CV4） acupoint on the second day following modeling. Similarly, apoptosis in ovarian granulosa cells was detected by terminal deoxynucleotidyl transferase d UTP nick end labeling staining, while enzyme-linked immunosorbent assay was employed for measuring serum folliclestimulating hormone（FSH）, luteinizing hormone（LH）, estrogen（E2）, and anti-müllerian hormone（AMH） levels. Moreover, transmission electron microscopy（TEM） was employed for examining mitochondrial morphology, while autophagy and hippo-yes-associated protein/transcriptional co-activator with postsynaptic density protein, drosophila disc large tumor suppressor, and zonula occludens-1 protein binding motif（YAP/TAZ） pathway related protein levels in ovarian tissue were detected via Western blotting. RESULTS: Analysis of serum levels of various hormones indicated that serum FSH and LH were reduced in EA compared to the POI group, while E2and AMH levels were found to be elevated in EA compared to the POI group. The EA was found to inhibit apoptosis in granulosa cells in POI model mice, in addition to improved mitochondrial damage and significantly improved mitophagy. Pathway analysis revealed that EA was involved in activating the hippo-YAP/TAZ pathway, followed by reversing EA effects on granulosa cell apoptosis and mitophagy with the use of verteporfin, an autophagy and YAP-T-cell factor/enhancer of split and activator of transcription domain family member interaction inhibitor. Transcription coactivator YAP1 promotes CCND1/CDK6 expression, stimulating cell proliferation in cloned cattle placentas

Somatic cell nuclear transfer(SCNT) has been successfully employed across various mammalian species,yet cloned animals consistently exhibit low pregnancy rates, primarily due to placental abnormalities such as hyperplasia and hypertrophy. This study investigated the involvement of the Hippo signaling pathway in aberrant placentaldevelopmentinSCNT-inducedbovine pregnancies. SCNT-derived cattle exhibited placental hypertrophy, including enlarged abdominal circumference and altered placental cotyledon morphology. RNA sequencing analysis indicated significant dysregulation of Hippo signaling pathway genes in SCNT placentas. Coexpression of YAP1 and CCND1 was observed in cloned blastocysts, placental tissues, and bovine placental mesenchymal stem cells(bPMSCs). Manipulation of YAP1expression demonstrated the capacity to regulate bPMSC proliferation. Experimental assays confirmed the direct binding of YAP1 to CCND1, which subsequently promoted CCND1 expression in bPMSCs. Furthermore, inhibition of CDK6, a downstream target of CCND1, attenuated SCNT bPMSC proliferation. This study identified YAP1 as a key regulatory component within the Hippo signaling pathway that drives placental hyperplasia in cloned cattle through up-regulation of CCND1-CDK6 expression, facilitating cell cycle progression. These findings offer potential avenues for enhancing cloning efficiency, with implications for evolutionary biology and the conservation of valuable germplasm resources.

High concentration of Ti3+ in C-TiOx nanosheets boosting efficient photodegradation

Liver transplantation using an otherwise-wasted partial liver resection graft

Liver transplantation represents a complex surgical procedure and serves as a curative treatment for patients presenting an acute or chronic end-stage liver disease, or carefully selected liver malignancy. A significant gap still exists between the number of available donor organs and potential recipients. The use of an otherwise-wasted resected liver lobe from patients with benign liver tumors is a new, albeit small, option to alleviate the allograft shortage. This review provides evidence that resected liver lobes may be used successfully in liver transplantation.

Research on the deformation mechanism of flexural toppling deep-seated landslides based on large centrifuge physical model testing: A case study of the Genie landslide in the Tibetan Plateau

Flexural toppling is a widespread failure mode in natural and excavated anti-inclined rock slopes of the eastern Tibetan Plateau， which can further evolve into large deep-seated landslides， posing significant threats to the construction and operation of important infrastructure. In this study， we took the large deep-seated Genie landslide as a case study and established a centrifuge physical model to analyze the deformation mechanism and evolutionary process of flexural toppling deep-seated landslides under the effect of gravity. The results indicate that the geomechanical mode of landslide instability is characterized by flexural toppling-shearing-sliding， with the deformation process could be divided into three stages: initial slow deformation stage marked by crack generation， rapid deformation stage characterized by the expansion and fracturing of cracks， and a low-speed deformation stage characterized by creep. Notably， the sliding surfaces do not adhere strictly to predefined bending zones but develop cracks along the bending parts， which gradually expand and penetrate deeper， eventually forming multiple potential sliding surfaces. The analysis of the entire process deformation evolution of flexural toppling deep-seated landslides suggests that the rapid deformation stage is the optimal time to take action for landslide prevention and control. The study demonstrates that the shear strength also plays a key factor in the evolution of flexural toppling. The research reveals the deformation mechanism and evolutionary process of large deep-seated landslides in deep-cut canyon areas， providing valuable reference for the prediction， assessment， and prevention of such slopes and landslides.
