Electroacupuncture activates AMPKa1 to improve learning and memory in the APP/PS1 mouse model of early Alzheimer's disease by regulating hippocampal mitochondrial dynamics

Objective: Studies have shown that electroacupuncture(EA) can alleviate cognitive impairments from Alzheimer's disease(AD) by regulating the expression of adenosine monophosphate-activated protein kinase(AMPK), but the specific mechanism involved remains to be elucidated. Therefore, this study explores the potential mechanism by which EA improves cognitive function from the perspective of mitochondrial ***: The four-month-old transgenic mice with amyloid precursor protein(APP)/presenilin 1(PS1)and AMPKa1-subunit conditional knockout(AMPKa1-cKO) were used for experiments. To evaluate the effects of EA treatment on cognitive function, the T-maze and Morris water maze were used. In addition,chemical exchange saturation transfer, thioflavin staining, transmission electron microscopy, mitochondrial membrane potential, and Western blotting were used to examine the potential mechanisms underlying the effects of EA on APP/PS1 ***: Both APP/PS1 mice and AMPKa1-cKO mice exhibited dysfunction in mitochondrial dynamics accompanied by learning and memory impairment. Inactivation of the AMPK/peroxisome proliferatoractivated receptor-γ coactivator-1a(PGC-1a) pathway increased pathological amyloid-β(Aβ) deposition and aggravated the dysfunction in mitochondrial dynamics. In addition, EA rescued learning and memory deficits in APP/PS1 mice by activating the AMPK/PGC-1a pathway, specifically by reducing pathological Aβ deposition, normalizing energy metabolism, protecting the structure and function of mitochondria,increasing the levels of mitochondrial fusion proteins, and downregulating the expression of fission proteins. However, the therapeutic effect of EA on cognition in APP/PS1 mice was hindered by ***: The regulation of hippocampal mitochondrial dynamics and reduction in Aβ deposition via the AMPK/PGC-1a pathway are critical for the ability of EA to ameliorate cognitive impairment in APP/PS1 mice.

Background: Chronic obstructive pulmonary disease(COPD), a common respiratory disease, can be effectively treated by traditional Chinese medicine(TCM). Qingfei Huatan, a TCM formula, has been reported to effectively alleviate the clinical symptoms of COPD patients. However, there is a lack of multi-centre,randomised, double-blind, controlled clinical trials documenting the clinical efficacy and safety of this formula in the treatment of acute exacerbation of COPD(AECOPD).Objective: This study evaluated the efficacy and safety of Qingfei Huatan formula in the treatment of AECOPD, thereby providing high-quality clinical ***, setting, participants and interventions: A total of 276 patients with AECOPD were included in this multi-centre, randomised, double-blind, placebo-controlled trial and were randomised into treatment and control groups at a ratio of 1:1. Patients in the treatment and control groups took Qingfei Huatan granules or simulated Qingfei Huatan granules twice a day, for 14 days, in addition to Western medicine treatment. All patients were followed up for 3 *** outcome measures: The primary outcome was time taken to symptom stabilisation. The secondary outcomes included duration of antibiotic use, clinical symptom and sign score, TCM syndrome score, dyspnoea score, and quality of life(QOL) score. Meanwhile, the safety of the formula was assessed through routine urine and stool tests, electrocardiograms, liver and kidney function tests, and the observation of adverse events throughout the ***: The time taken for effective stabilisation(P < 0.05) and obvious stabilisation(P < 0.01), and the duration of antibiotic use(P < 0.05) were significantly shorter in the treatment group than in the control group. On days 6, 9, 12 and 14 of treatment, clinical symptom and sign score decreased in both groups,particularly in the treatment group(P < 0.01). 气象灾害治理往往涉及多部门主体应急联动,当前治理环境复杂多变,如何提升气象灾害应急联动机制效能成为迫在眉睫的关键问题。基于气象灾害应急联动机制效能提升的质性研究,运用模糊集定性比较分析对我国2008-2022年间的21个案例进行组态分析。研究发现:(1)单一条件不构成气象灾害应急联动机制效能提升的必要条件;(2)高水平气象灾害应急联动机制效能提升路径可以归纳为“法制—资源双重驱动型”“法制—主体全面驱动型”和“法制—资源全面驱动型”3种类型;(3)在特定的客观禀赋条件下,信息共享和主体协调构成的条件配置可以与事件影响和主体协调构成的条件配置通过等效替代的方式提升气象灾害应急联动机制效能。

目的:研究BrdU标记猪脂肪干细胞(adipose-derived stem cells,ADSCs)的最佳剂量及时间,探讨其作为干细胞标记示踪方法的可行性.方法:自中华实验猪背部提取脂肪组织,采用贴壁法分离培养ADSCs,取第3代细胞以终浓度分别为10、15、20、25及30μmol/LBrdU进行标记;另一孔不含BrdU作为对照组,分别培养12、24、48、72及96h.免疫荧光法检测各组细胞BrdU标记率,找出BrdU的最佳标记方法,通过台盼蓝排斥试验、MTT及细胞凋亡检测,观察BrdU对ADSCs生长情况的影响.对第3代的ADSCs采用最佳标记方法后更换普通培养基继续培养,适时传代,连续检测第4、5、6、7、8代ADSCs的BrdU标记率.结果:原代培养的ADSCs的形态主要为长梭形,第3代ADSCs经BrdU标记后胞核呈红色荧光,随浓度的升高及时间的延长,BrdU阳性标记率逐渐升高,以终浓度20μmol/LBrdU标记48h后阳性率达90%以上,且连续传5代标记率仍达40%.MTT、台盼蓝排斥试验及细胞凋亡检测发现BrdU对ADSCs生长增殖基本无影响.结论:BrdU标记ADSCs的最佳剂量和时间为20μmol/L和48h,该方法标记率高,对细胞影响小,可用于动态研究ADSCs在移植体内生长、分化.

开发特殊性能红外材料是目前提升红外光学系统性能的关键,硫系玻璃作为组分-性能可调的红外材料无疑成为了热门选项.Se基与Te基玻璃涵盖中波、长波红外窗口,是最典型的红外器件应用材料之一,通过对Ge_(20)Se_(80–x)Te_(x)玻璃体系的结构与性能分析,阐述了Te含量对该玻璃体系结构与性能演变的规律.随着Te含量的增大,玻璃转变温度(Tg)受网络结构及平均键能的影响先升高后降低,密度与折射率近似线性的梯度增高,阿贝数逐渐增大,而维氏硬度几乎不随Te含量的变化而变化,断裂韧性随Te含量的增大而降低.针对平均配位数无法评价两种及以上同族元素组成的玻璃体系问题,成功建立了理论带隙-玻璃性能评价体系,并对Ge_(20)Se_(80–x)Te_(x)玻璃体系的密度、折射率、阿贝数、断裂韧性等参数与理论带隙建立了函数关系,该体系可用于快速评估玻璃组分与性能.

Highly efficient discovery of the covalent Minhibitors from crude Pu-erh tea by integrating biochemical and chemoproteomic approaches

The main proteases (Mpros) are hydrolases playing essential roles in the replication of β-coronaviruses including SARS-CoV-2. Herein， a highly efficient strategy was developed for discovering the Mproinactivators from crude plant extract integrating target-based biochemical assay and chemoproteomic approaches. Firstly， Pu-erh tea was found to potently suppress SARS-CoV-2 Mproin a time-dependent manner. Next， global chemical analysis coupling with peptide-modification profiling were used to identify the cysteine-modified constituents in Pu-erh tea. The results suggested that seven constituents in Pu-erh tea could modify SARS-CoV-2 Mpro， which turned out that epigallocatechin， gallocatechin and gallic acid were the most efficacious Mproinactivators. Further investigations demonstrated that epigallocatechin and gallocatechin could inactivate SARS-CoV-2 Mprovia blocking the formation of the homodimers. Collectively， this work proposed a novel and practical strategy for highly efficient discovery of time-dependent inhibitors of SARS-CoV-2 Mprofrom plant extracts， while three constituents in Pu-erh tea have emerged as robust SARS-CoV-2 Mproinactivators.
