AIM: To investigate the effects of interferon-alpha (IFN-α) to restrain the growth and invasive potential of hepatocellular carcinoma (HCC) induced by hepatitis B virus (HBV) X protein. METHODS: The pcDNA3.1-HBx plasmid was transfected into Chang cells by Lipofectamine in vitro, and Chang/HBx was co-cultured with IFN-α. Cell survival growth curve and clonogenicity assay were used to test the growth potential of Chang/pcDNA3.1, Chang/HBx and IFN-α-Chang/HBx in vitro. Growth assay in nude mice was used to detect the growth potential of Chang/ pcDNA3.1, Chang/HBx and IFN-α-Chang/HBx in vivo. Wound healing and transwell migration assays were used to detect the invasive ability of Chang/pcDNA3.1, Chang/HBx and IFN-α-Chang/HBx. RESULTS: Compared with CCL13 cells transfected with pcDNA3.1, CCL13 with stable expression of hepatitis B virus X protein showed the characteristics of malignant cells with high capability of growth and invasion by detecting their growth curves, colony forming efficiency, wound healing , transwell migration assays and growth assays in nude mice. Its capability of growth and invasion could be controlled by IFN-α. CONCLUSION: IFN-α can restrain the growth and invasive potential of HCC cells induced by HBx protein, which has provided an experimental basis for IFN-α therapy of HCC.

AIM： To explore the possibility of reversing multi-drug resistance （MDR） to HepG2/mdr1 in vitro and in vivo with RNA interference （RNAi）. METHODS： HepG2/mdr1 was obtained by cloning the whole gene mdrl into HepG2 cells, shRNA targeting sequence was designed to be homologous to the P-gp encoding MDR1 mRNA consensus sequence, pSUPER- shRNA/mdrl was constructed using the enzyme- digested technique. HepG2/mdrl cells were transfected with vectors of pSUPER-shRNA/mdrl to measure their efficacy by real-time PCR for mdrl mRNA, flow cytometry （FCM） for P-gp expression, and Rhodamine efflux, MTT method for HepG2/mdrl function, respectively. In vivo, mice tumors were treated by injecting pSUPER-shRNA/mdrl in situ and into intra- abdominal cavity. Tumors were collected to create cell suspension and cryosections after chemothearpy with adiramycin and mytomycin. The cell suspension was incubated in RPMI-1640 supplemented with G418 to screen stable cells for appreciating the reversal of MDR. Cryosections were treated with immunohistochemistry technique to show the effectiveness of transfection and the expression of ***： pSUPER-shRNA/mdrl was successfully constructed, which was confirmed by sequencing. The MDR phenotype of HepG2/mdr1 was decreased significantly in vitro transfection. HepG2/mdr1 showing its MDR was reversed notably in P-gp expression （11.0% vs 98.2%, P 〈 0.01）. Real-time PCR showed that mRNA/mdrl was lower in test groups than in control groups （18.73± 1.33 vs 68.03±2.21, P 〈 0.001）. Compared with HepG2, the sensitivity of HepG2/ mdrl and HepG2/mdr1-dsRNA cells to ADM was decreased by 1.64 times and 15.6 times, respectively. The accumulation of DNR in positive groups was decreased evidently. In vivo, the p-gp expression in positive groups was significantly lower than that in control groups （65.1% vs 94.1%, P 〈 0.05）. The tumor suppressing rate in test groups was 57.8%. After chemotherapy, the growth rate in test groups was lower than that in control groups （

As the bed depth increases,sintering yield increases,but the productivity *** reveal the reasons for the decrease in productivity and explore targeted solutions,the bed resistance of mixtures,wet zone,and combustion zone was analyzed in the *** results showed that the decreased porosity of mixture resulted in the increased bed resistance by 160.56%when the bed depth increased from 600 to 1000 *** improving porosity of 1%by adding loosening bars with optimized size and distribution,the bed resistance decreased,and the productivity increased by 5%.The increase in bed depth increased the thickness of the wet zone from 120 to 680 mm and the resistance from 1.56 to 8.83 *** using a three-stage intensive mixer and pre-adding water for granulation,the moisture of mixture was reduced by 0.6%,and the sintering productivity increased by 4%.Besides,the high bed resistance is mainly caused by the increase in the thickness of the combustion zone from 31.9 to 132.7 mm,and the bed resistance increased from 0.70 to 5.62 *** bed resistance of the combustion zone at 900 mm was increased by 90.51%compared to 700 *** optimization of the distribution of coke breeze,the thickness of combustion zone at the lower layer decreased from 132.7 to 106.84 mm and permeability improved significantly.

Transition metal-based compounds can serve as pre-catalysts to obtain genuine oxygen evolution reaction（OER） electrocatalysts in the form of oxyhydroxides through electrochemical ***,the role and existence form of leached oxygen anions are still ***,we selected iron selenite-wrapped hydrated nickel molybdate（denoted as NiMoO/FeSeO） as a pre-catalyst to study the oxyanion *** is surprising to find that SeO2-exists in the catalyst in the form of intercalation,which is different from previous studies that suggest that anions are doped with residual elements after electrochemical activation,or adsorbed on the catalyst *** experiment and theoretical calculations show that the existence of SeO42-intercalation effectively adjusts the electronic structure of NiFeOOH,promotes intramolecular electron transfer and O-O release,and thus lowers the reaction energy *** expected,the synthesized NiFeOOH-SeO only needs 202 and 285 mV to attain 100 and 1000 mA cm-2in 1 M ***,the anion exchange membrane water electrolyzer（AEMWE） consisting of NiFeOOHSeO anode and Pt/C cathode can reach 1 A cm-2at 1.70 V and no significant attenuation within 300 *** findings provide insights into the mechanism,by which the intercalated oxyanions enhance the OER performance of NiFeOOH,thereby facilitating large-scale hydrogen production through AEMWE.

To accurately investigate the evolution characteristics and generation mechanism of retained oil, the study analyzed organic-rich lacustrine shale samples from the Paleogene Kongdian Formation in Cangdong Sag, Bohai Bay Basin. This analysis involves Rock-Eval pyrolysis, pyrolysis simulation experiments,Gas Chromatograph Mass Spectrometer（GC-MS）, and reactive molecular dynamics simulations（Reax FF）. The results revealed the retained oil primarily consisted of n-alkanes with carbon numbers ranging from C14to C36. The generation of retained oil occurred through three stages. A slow growth stage of production rate was observed before reaching the peak of oil production in Stage I. Stage II involved a rapid increase in oil retention, with C12-C17and C24-C32serving as the primary components,increasing continuously during the pyrolysis process. The generation process involved the cleavage of weak bonds, including bridging bonds（hydroxyl, oxy, peroxy, imino, amino, and nitro）, ether bonds, and acid amides in the first stage（Ro= 0.50%-0.75%）. The carbon chains in aromatic ring structures with heteroatomic functional groups breaks in the second stage（Ro= 0.75%-1.20%）. In the third stage（Ro= 1.20%-2.50%）, the ring structures underwent ring-opening reactions to synthesize iso-short-chain olefins and radicals, while further breakdown of aliphatic chains occurred. By coupling pyrolysis simulation experiments and molecular simulation technology, the evolution characteristics and bond breaking mechanism of retained oil in three stages were revealed, providing a reference for the formation and evolution mechanism of retained oil.

In this paper,a type of reinforcing structure for composite shell with single and through hole is *** experimental tests for the composite shells without hole,with single hole and reinforced structure,with through hole and reinforced structure subjected to hydrostatic pressure were carried out by the designed experimental test *** mechanical responses of the composite shells under hydrostatic pressure are obtained by the high-speed camera and strain *** results show that the entire deformation process of the shell can be divided into three:uniform compression,"buckling mode formation"and ***"buckling mode formation"process is captured and reported for the first *** the composite shell with single hole,the proposed reinforcing structure has a significant reinforcement effect on the shell and the buckling capacity of the shell is not weaker than the complete composite *** the composite shell with through hole,sealing effect can be achieved by the proposed reinforcing structure,but the buckling capacity of the shell after reinforcement can only reach 77%of the original buckling capacity.
