Transcription factors CncC and Maf connect the molecular network between pesticide resistance and resurgence of pest mites

Pesticide resistance and resurgence are serious problems often occurring simultaneously in the field.In our long-term study of a fenpropathrin-resistant strain of Tetranychus cinnabaribus,enhancement of detoxification and modified fecundity mechanisms were both observed.Here we investigate the network across these two mechanisms and find a key node between resistance and resurgence.We show that the ecdysone pathway is involved in regulating the fecundity of T.cinnabaribus.The concentration change of ecdysone is consistent with the fecundity curve;the concentration of ecdysone is higher in the fenpropathrin-resistant strain which has stronger fecundity.The enhancement of ecdysone is due to overexpression of two P450 genes(CYP314A1 and CYP315A1)in the ecdysone synthesis pathway.Silencing expression of these CYP genes resulted in lower concentration of ecdysone,reduced expression of vitellogenin,and reduced fecundity of T.cinnabaribus.The expression of CYP315A1 is regulated by transcription factors Cap-n-collar isoform C(CncC)and Musculoaponeurotic fibrosarcoma protein(Maf),which are involved in regulating other P450 genes functioning in detoxification of fenpropathrin in T.cinnabaribus.A similar regulation is established in citrus pest mite Panonychus citri showing that the CncC pathway regulates expression of PcCYP315Al,which affects mite fecundity.Transcription factors are activated to upregulate detoxification genes facilitating pesticide resistance,while the“one to multiple”regulation mode of transcription factors simultaneously increases expression of metabolic enzyme genes in hormone pathways and alters the physiology of pests.This is an important response of arthropods to pesticides which leads to resistance and population resurgence.

Identification of the key chitinase genes in Tetranychus cinnabarinus (Boisduval) based on the expression and sequence characteristic analysis

Chitin is an important content in the exoskeletons of arthropods, and its hydrolyzation is catalyzed by chitinases during the process of molting, thus, the chitinases are considered as ideal target to interfere the growth of arthropods. This study intends to clarify the characteristic of the chitinases during the development of Tetranychus cinnabarinus, and screen out important genes as potential control targets. The results showed that the total enzyme concentration of chitinases was significantly higher in larva, the first and second nymph than that in egg and adult. Base on the transcriptome data, six unigenes encoding chitinases were identified and their expression patterns in different developmental stages were detected. The expressions of TcCHIT1 and TcCHITIO showed high abundance during the molting process and their expression change during the developmental stages was consistent with the enzyme concentration. The full-length of these two genes were further cloned, and the structural characteristics of their proteins were analyzed by constructing the three-dimensional structure model. The results provide basic information to understand the characteristic of chitinases in T. cinnabarinus and might be considered as target for control.

Functional analysis of UG7207D3 associated with abamectin resistance in Tetranychus cinnabarinus(Boisduval)

Uridine diphosphate(UDP)-glycosyltransferases(UGTS)are widely distributed within living organisms and share roles in biotransformation of various lipophilic endo-and xenobiotics with activated UDP sugars.In this study,it was found that the activity of UGTs in abamectin-resistant(AbR)strain was significantly higher(2.3-fod)than that in susceptible strain(SS)of Tetramychus cinnabarinus.Further analysis showed that 5-nitrouracil,the inhibitor ofUGTs,could enhance the lethal effect of abamectin on mites.From the previous microarray results,we found an UGT gene(UGT201D3)overexpressed in AbR strain.Quantitative PCR analysis showed that UGT20ID3 was highly expressed and more inducible with abamectin exposure in the AbR strain.After silencing the tran-scription of UGT201D3,the activity of UGTs was decreased and the susceptibility to abamectin was increased in AbR strain whereas it was not in sS.Furthermore,UGT201D3 gene was then succefully expressed in Escherichia coli.The recombinant UGT20ID3 exhibited a-naphthol activity(2.810.43 nmol mg protein/min),and the enzyme activity could be inhibited by abamectin(inhibitory concentration at 50%:57.50±3.54 μmol/L).High-performance liquid chromatography analysis demonstrated that the recombinant UGT201D3 could efctively deplete abamectin(15.77%±3.72%)incubating with 150 ug protein for 6 h.These results provided direct evidence that UGT201D3 was involved in abamectin resistance in T cinnabarinus.

Retrieval of Land-surface Temperature from AMSR2 Data Using a Deep Dynamic Learning Neural Network

It is more difficult to retrieve land surface temperature(LST) from passive microwave remote sensing data than from thermal remote sensing data, because the emissivities in the passive microwave band can change more easily than those in the thermal infrared band. Thus, it is very difficult to build a stable relationship. Passive microwave band emissivities are greatly influenced by the soil moisture, which varies with time. This makes it difficult to develop a general physical algorithm. This paper proposes a method to utilize multiple-satellite, sensors and resolution coupled with a deep dynamic learning neural network to retrieve the land surface temperature from images acquired by the Advanced Microwave Scanning Radiometer 2(AMSR2), a sensor that is similar to the Advanced Microwave Scanning Radiometer Earth Observing System(AMSR-E). The AMSR-E and MODIS sensors are located aboard the Aqua satellite. The MODIS LST product is used as the ground truth data to overcome the difficulties in obtaining large scale land surface temperature data. The mean and standard deviation of the retrieval error are approximately 1.4° and 1.9° when five frequencies(ten channels, 10.7, 18.7, 23.8, 36.5, 89 V/H GHz) are used. This method can effectively eliminate the influences of the soil moisture, roughness, atmosphere and various other factors. An analysis of the application of this method to the retrieval of land surface temperature from AMSR2 data indicates that the method is feasible. The accuracy is approximately 1.8° through a comparison between the retrieval results with ground measurement data from meteorological stations.

Transgenic cotton expressing C YP392A4 double-stranded RNA decreases the reproductive ability of Tetranychus cinnabarinus

As a polyphagous pest, Tetranychus cinnabarinus has the ability to overcome the defense of various hosts, and causes severe losses to various economically important crops. Since the interaction between pest and host plants is a valuable clue to investigate potential ways for pest management, we intend to identify the key genes of T. cinnabarinus for its adaption on cotton, then, with RNA interference （RNAi） and transgenic technology, construct a transgenic cotton strain to interfere with this process, and evaluate the effect of this method on the management of the mites. The difference of gene expression of T. cinnabarinus was analyzed when it was transferred to a new host （from cowpea to cotton） through high-throughput sequencing, and a number of differentially expressed genes involved in detoxification, digestion and specific processes during the development were classified. From them, a P450 gene CYP392A4 with high abundance and prominent over-expression on the cotton was selected as a candidate. With transgenic technology, cotton plants expressing double-stranded RNA of CYP392A4 were constructed. Feeding experiments showed that it can decrease the expression of the target gene, result in the reduction of reproductive ability of the mites, and the population of T. cinnabarinus showed an apparent fitness cost on the transgenic cotton. These results provide a new approach to restrict the development of mite population on the host. It is also a useful attempt to control piercing sucking pests through RNAi and transgenic technology.

Design of 50 MeV proton microbeam based on cyclotron accelerator

High-energy proton microbeam facilities are powerful tools in space science,biology and cancer therapy studies.The primary limitations of the 50 MeV proton microbeam system are the poor beam quality provided by the cyclotron and the problem of intense scattering in the slit position.Here,we present an optical design for a cyclotron-based 50 MeV high-energy proton microbeam system with a micron-sized resolution.The microbeam system,which has an Oxford triplet lens configuration,has relatively small spherical aberrations and is insensitive to changes in the beam divergence angle and momentum spread.In addition,the energy filtration included in the system can reduce the beam momentum spread from 1 to 0.02%.The effects of lens parasitic aberrations and the lens fringe field on the beam spot resolution are also discussed.In addition,owing to the severe scattering of 50 MeV protons in slit materials,a slit system model based on the Geant4 toolkit enables the quantitative analysis of scattered protons and secondary particles.For the slit system settings under a 10-micron final beam spot,very few scattered protons can enter the quadrupole lens system and affect the focusing performance of the microbeam system,but the secondary radiation of neutrons and gamma rays generated at the collimation system should be considered for the 50 MeV proton microbeam.These data demonstrate that a 50 MeV proton microbeam system with a micron-sized beam spot based on a cyclotron is feasible.

Influence of various stressors on the expression of core genes of the small interfering RNA pathway in the oriental fruit fly, Bactrocera dorsalis

RNA interference （RNAi）-based technology has emerged as a potential tool for controlling insect pests, however, previous studies found that the efficiency of RNAi in Bactrocera dorsalis was variable. In nature, insects often meet various challenges, such as pathogen infections, extreme temperatures, lack of nutrition and heavy metals. To better understand the association of the stressors with efficiency of RNAi, in the current study we tested the expression of three core genes, dicer2 （Bddcr2）, r2d2 （Bdr2d2） and argonaute2 （Bdago2）, of the small interfering RNA （siRNA） pathway of B. dorsalis upon various stressors. Our results showed that all three genes were upregulated by the infection of invertebrate iridescent virus 6, which suggested a function of the siRNA pathway against viral infection. The loading of FeC13 could also increase the expression of Bddcr2. The treatments ofEscherichia coli, extremely high （40~C） and low （0~C） temperatures, as well as starvation, could negatively influence the expression of Bddcr2 and/or Bdago2. In total, our results showed that various stressors could influence the expression of core components ofB. dorsalis siRNA pathway. This highlights further speculation on the RNAi efficiency upon these stressors. Considering the complexity and variation of RNAi efficiency in different conditions, these results provide initial aspects in possible environmental stressors to influence the activity of the siRNA pathway, but the real impact of RNAi efficiency posed by these stressors requires further studies.

Mouse embryonic stem cells(mESCs)cycle in and out of a transient 2-cell(2C)-like totipotent state,driven by a com-plex genetic circuit involves both the coding and repetitive sections of the genome.While a vast array of regulators,including the multi-functional protein Rif1,has been reported to influence the switch of fate potential,how they act in concert to achieve this cellular plasticity remains elusive.Here,by modularizing the known totipotency regulatory factors,we identify an unprecedented functional connection between Rif1 and the non-canonical polycomb repres-sive complex PRC1.6.Downregulation of the expression of either Rif1 or PRC1.6 subunits imposes similar impacts on the transcriptome of mESCs.The LacO-LacI induced ectopic colocalization assay detects a specific interaction between Rif1 and Pcgf6,bolstering the intactness of the PRC1.6 complex.Chromatin immunoprecipitation followed by sequencing(ChIP-seq)analysis further reveals that Rif1 is required for the accurate targeting of Pcgf6 to a group of genomic loci encompassing many genes involved in the regulation of the 2C-like state.Depletion of Rif1 or Pcgf6 not only activates 2C genes such as Zscan4 and Zfp352,but also derepresses a group of the endogenous retroviral element MERVL,a key marker for totipotency.Collectively,our findings discover that Rif1 can serve as a novel auxiliary component in the PRC1.6 complex to restrain the genetic circuit underlying totipotent fate potential,shedding new mechanistic insights into its function in regulating the cellular plasticity of embryonic stem cells.

Identification and Expression of a b-actin Gene from Liposcelis bostrychophila Badonnel(Psocoptera:Liposcelididae)

A β-actin gene, Libβ-actinl, from the psocid, Liposcelis bostrychophila, was isolated, sequenced, and expressed in Escherichia coll. The cDNA sequence was 1 281 bp in length and contained an open reading frame of 1 131 bp encoding 376 amino acids with a predicted molecular weight of 41.82 kDa. According to a B lastN search, the coding region shared the highest identity （97%） with Pediculus hurnanus actin 5C, while the deduced amino acid sequence was completely identical to a mutant of Drosophila melanogaster actin 5C. Comparison of the nucleotide and deduced amino acid sequences confirmed the high similarity between Libβ-actinl and homologs in other insect species. The 3＇ untranslated region （3＇ UTR） of the Libβ-actinl mRNA had a high A＋U content （approximately 75%） and contained three repeats of the AUUUUUA and AUUUA motifs, which may play a role in regulating mRNA decay. The expression of Libβ-actinl was further analyzed in insecticide induced and control psocids. The results indicated that there was no significant difference in expression of Libβ-actinl between the induced and control groups, suggesting that Libβ-actinl may be an appropriate internal control for the gene expression profiling in this insect. Furthermore, Libβ-actinl was also heterologously expressed in Escherichia coli, which provided a basis to investigate the physiological functions of actin genes in the psocid.
