Comparative proteomic analysis of plasma exosomes reveals the functional contribution of N-acetyl-alpha-glucosaminidase to Parkinson’s disease

Parkinson’s disease is the second most common progressive neurodegenerative disorder,and few reliable biomarkers are available to track disease *** proteins,DNA,mRNA,and lipids carried by exosomes reflect intracellular changes,and thus can serve as biomarkers for a variety of *** this study,we investigated alterations in the protein content of plasma exosomes derived from patients with Parkinson’s disease and the potential therapeutic roles of these proteins in Parkinson’s *** a tandem mass tag-based quantitative proteomics approach,we characterized the proteomes of plasma exosomes derived from individual patients,identified exosomal protein signatures specific to patients with Parkinson’s disease,and identified N-acetyl-alpha-glucosaminidase as a differentially expressed protein.N-acetyl-alpha-glucosaminidase expression levels in exosomes from the plasma of patients and healthy controls were validated by enzyme-linked immunosorbent assay and western *** results demonstrated that the exosomal N-acetyl-alpha-glucosaminidase concentration was not only lower in Parkinson’s disease,but also decreased with increasing Hoehn-Yahr stage,suggesting that N-acetyl-alpha-glucosaminidase could be used to rapidly evaluate Parkinson’s disease ***,western blot and immunohistochemistry analysis showed that N-acetyl-alpha-glucosaminidase levels were markedly reduced both in cells treated with 1-methyl-4-phenylpyridinium and cells overexpressingα-synuclein compared with control ***,N-acetyl-alpha-glucosaminidase overexpression significantly increased cell viability and inhibitedα-synuclein expression in 1-methyl-4-phenylpyridinium-treated *** together,our findings demonstrate for the first time that exosomal N-acetyl-alpha-glucosaminidase may serve as a biomarker for Parkinson’s disease diagnosis,and that N-acetyl-alpha-glucosaminidase may reduceα-synuclein expression and 1-methyl-4-phenylpyridinium-induced n

New scoring system for the evaluation obstructive degrees based on computed tomography for obstructive colorectal cancer

BACKGROUND The degree of obstruction plays an important role in decision-making for obstructive colorectal cancer(OCRC).The existing assessment still relies on the colorectal obstruction scoring system(CROSS)which is based on a comprehensive analysis of patients’complaints and eating *** data collection relies on subjective descriptions and lacks objective ***,a scoring system for the evaluation of computed tomography-based obstructive degree(CTOD)is urgently required for *** To explore the relationship between CTOD and CROSS and to determine whether CTOD could affect the short-term and long-term *** Of 173 patients were *** was obtained using k-means,the ratio of proximal to distal obstruction,and the proportion of nonparenchymal areas at the site of *** was integrated with the CROSS to analyze the effect of emergency intervention on ***-term and long-term outcomes were compared between the *** CTOD severe obstruction(CTOD grade 3)was an independent risk factor[odds ratio(OR)=3.390,95%confidence interval(CI):1.340-8.570,P=0.010]via multivariate analysis of short-term outcomes,while CROSS grade was *** the CTOD-CROSS grade system,for the non-severe obstructive(CTOD 1-2 to CROSS 1-4)group,the complication rate of emergency interventions was significantly higher than that of non-emergency interventions(71.4%vs 41.8%,P=0.040).The postoperative pneumonia rate was higher in the emergency intervention group than in the non-severe obstructive group(35.7%vs 8.9%,P=0.020).However,CTOD grade was not an independent risk factor of overall survival and progression-free *** CTOD was useful in preoperative decision-making to avoid unnecessary emergency interventions and complications.

Design,Synthesis and Pharmacological Evaluation of Novel 4-Phenoxyquinoline Derivatives as VEGFR2 Kinase Inhibitors for Tumor Treatment

Vascular endothelial growth factor 2(VEGFR2)plays a vital role in regulating of tumor metastasis and angiogenesis,which has emerged as one of the effective targets for clinical tumor ***,a series of novel facilely accessible 4-phenoxyquinoline derivatives was prepared and assessed for their antitumor activity against three human tumor cell lines(SGC-7901,HepG2 and A549).Among these compounds,6a,6b and 6c show strong antitumor activity on HepG2 cells[the drug concentration of eliminating half of tumor cells(IC_(50))=9.33,1.84,8.54μmol/L].Notably,compound 6b shows potent selective inhibitory activity against VEGFR2 kinase with an IC_(50) value of 4.66 nmol/*** excellent anti-angiogenesis capability of compound 6b was confirmed by tube formation and chick chorioallantoic membrane(CAM)*** vivo studies confirmed that compound 6b was able to inhibit tumor growth in HepG2 xenografts of BALB/c nude mice without obvious side or toxic *** results demonstrated that compound 6b exhibited remarkable anti-angiogenesis and tumor growth inhibitory effects with less toxicity in vitro and in vivo *** findings highlighted the potential of compound 6b as a promising VEGFR2 kinase inhibitor for the development of antitumor drugs.

Functional ternary salt construction enabling an in-situ Li3N/LiF-enriched interface for ultra-stable all-solid-state lithium metal batteries

Poly（ethylene oxide）-based polymer all-solid-state lithium metal batteries（ASSLBs） have received widespread attention due to their low cost,good process ability,and high energy ***,the growth of Li dendrites within polymer solid-state electrolytes damages the reversibility of Li anodes and still impedes their widespread *** efficient strategy is to construct a superior solid electrolyte ***,a stable interface enriched with Li3N and LiF is in-situ formed between Li anode and a terna ry salt *** terna ry salt electrolyte is innovatively designed by introducing lithium bis（trifluoromethane sulfonyl）imide（LiTFSI）,lithium bis（fluorosulfonyl）imide（LiFSI）,and LiNO3to poly（ethylene oxide） *** characterization indicates that LiNO3and LiFSI contribute to forming a Li3N-LiF-enriched interface and meanwhile LiTFSI ensures excellent ***,among various Li compound components,Li3N has high ionic conductivity,which is beneficial for reducing overpotential,while LiF has high interfacial energy which can enhance nucleation energy and suppress the formation of Li *** experimental results show that ASSLBs coupled with LiFePO4cathode display extremely excellent cycle stability approximately 2200 cycles at 2 C,with a final and corresponding discharge specific capacity of 96.7 mA h g-1. Additionally,a schematic illustration of the working mechanism for the Li3N-LiF interface is proposed.

Separation of the ionospheric and thermospheric contributions in the data from the Triple Ionosophere Photo Meter onboard the Feng-Yun 3E satellite

The Triple Ionosphere Photo Meter(TRIPM)is a far ultraviolet nadir-view photometer that is onboard the secondgeneration,polar-orbiting Chinese meteorological satellite Feng-Yun 3E(FY-3E),which was launched on July 5,*** has three channels,and each is used to measure different types of emissions:(1)135.6 nm airglow produced by the recombination of the O+ions and electrons in the absence of sunlight(nighttime channel);(2)135.6 nm airglow from the OI when sunlight is present(daytime/dusk-dawn 135.6 nm channel);and(3)Airglow in the N_(2)LBH of the TRIPM(daytime/dusk-dawn N_(2)LBH channel).FY-3E is a satellite operating in the early morning ***,the data are mostly distributed on the dawndusk side or at times when the solar zenith angle is relatively *** this study,combined with simulation calculations,the data are *** further explore the application value of the data products,a model is used to separate the ionospheric radiative recombination contribution from the thermosphere photoelectron impact excitation contribution in the data from the daytime/dusk-dawn 135.6 nm *** approach not only improves the accuracy of O/N_(2)derived from the 135.6 nm channel and N_(2)LBH channel but also provides the ionospheric 135.6 nm radiance at the same time as the *** results show that the 135.6 nm radiance originating from the combination of O+and electrons extracted from the measurement data exhibits similar equatorial anomaly peak structure characteristics and seasonal variations as the corresponding GPS total electron content(TEC)*** the other hand,the separated 135.6 nm radiance generated from the thermosphere photoelectron collision excitation does not contain radiative recombination contributions;thus,the ionosphere influence is greatly avoided when the O/N_(2)is obtained.

CMOS direct conversion X-ray detector coupled with fluorinated liquid

X-ray detectors show potential applications in medical imaging,materials science,and nuclear *** achieve high detection efficiency and spatial resolution,many conventional semiconductor materials,such as amorphous selenium,cadmium telluride zinc,and perovskites,have been utilized in direct conversion X-ray ***,these semiconductor materials are susceptible to temperature-induced performance degradation,crystallization,delamination,uneven lattice growth,radiation damage,and high dark *** study explores a new approach by coupling an FC40 electronic fluorinated liquid with a specialized high-resolution and high-readout-speed complementary metal-oxide-semiconductor(CMOS)pixel array,specifically the Topmetal II−chip,to fabricate a direct conversion X-ray *** fluorinated liquid FC40(molecular formula:C_(21)F_(48)N_(2))is an electronic medium that is minimally affected by temperature and displays no issues with uniform *** exhibits a low dark current and minimal radiation damage and enables customizable thickness in X-ray *** addresses the limitations inherent in conventional semiconductor-based *** this study,simple X-ray detector imaging tests were conducted,demonstrating the excellent coupling capability between FC40 electronic fluorinated liquid and CMOS chips by the X-ray detector.A spatial resolution of 4.0 lp/mm was measured using a striped line par card,and a relatively clear image of a cockroach was displayed in the digital radiography imaging *** test results indicated the feasibility of fabricating an X-ray detector by combining FC40 electronic fluorinated liquid and CMOS *** to the absence of issues related to chip-material coupling,a high spatial resolution could be achieved by reducing the chip pixel *** method presents a new avenue for studies on novel liquid-based direct conversion X-ray detectors.

A Mn-porphyrinic metal–organic framework immobilizing glucose oxidase for combined photodynamic/chemodynamic/starvation therapy

The combination of photodynamic therapy（PDT） and chemodynamic therapy（CDT） in tumor treatment has attracted considerable ***,tumor hypoxia and glutathione（GSH） overproduction in the tumor tissue restricted the progress of their ***,a Mn-porphyrinic metal-organic framework（Mn-TCPP） was constructed by the one-pot method and further used for immobilizing glucose oxidase（GOx） to obtain GOx@*** would rapidly exhaust endogenous glucose into hydrogen peroxide（H2O2） and gluconic acid,thus shutting off the energy supply of tumor cells for starvation ***-TCPP catalyzed H2O2to produce oxygen,regulating the hypoxic tumor microenvironment and in turn improving1O2generation under laser ***,Mn-TCPP can reduce reactive oxygen species consumption owing to the redox reaction between Mn3+and GSH,thus greatly enhancing ***,benefiting from GOx-mediated starvation therapy,Mn2+produced by Mn3+reduction can react with sufficient intracellular H2O2to generate ·OH with high cytotoxicity through a Fenton-like *** treatment by GOx@Mn-TCPP under laser irradiation in vitro,the cell viability of 4T1 and A549 tumor cells reached to 20%,reflecting GOx@Mn-TCPP could give full play to the advantages of PDT/CDT/starvation *** results in vivo demonstrated that GOx@Mn-TCPP mediated synergistic cascade therapy could significantly inhibit tumor growth and improve the therapeutic effect.
