A comparison of perioperative outcomes between extraperitoneal robotic single-port and multiport radical prostatectomy with the da Vinci Si Surgical System

To evaluate outcomes between extraperitoneal robotic single-port radical prostatectomy(epR-spRP)and extraperitoneal robotic multiport radical prostatectomy(epR-mpRP)performed with the da Vinci Si Surgical System,comparison was performed between 30 single-port(SP group)and 26 multiport(MP group)*** included operative time,estimated blood loss(EBL),hospital stay,peritoneal violation,pain scores,scar satisfaction,continence,and erectile *** median operation time and EBL were not different between the two *** the SP group,the median operation time of the first 10 patients was obviously longer than that of the latter 20 patients(P<0.001).The median postoperative hospital stay in the SP group was shorter than that in the MP group(P<0.001).The rate of peritoneal damage in the SP group was less than that in the MP group(P=0.017).The pain score and overall need for pain medications in the SP group were lower than those in the MP group(P<0.001 and P=0.015,respectively).Patients in the SP group were more satisfied with their scars than those in the MP group 3 months postoperatively(P=0.007).At 3 months,the cancer control,recovery of erectile function,and urinary continence rates were similar between the two *** is safe and feasible to perform epR-spRP using the da Vinci Si surgical ***,epR-spRP can be a treatment option for localized prostate *** epR-spRP still has a learning curve,it has advantages for postoperative pain and self-assessed *** the absence of the single-port robotic surgery platform,we can still provide minimally invasive surgery for patients.

Traditional Chinese medicines(TCMs),usually comprising hundreds of different compounds with distinctive physiochemical properties,have been practiced for thousands of years worldwide for the treatment of various human *** pharmacological properties have been clarified for many TCM in recent decades,while great efforts are needed to identify the active biomedical materials from TCM due to challenges such as technical barriers to screening,isolation,characterization,and optimization[1].Recent improvements in analytical tools,genome mining,and bioengineering technologies such as CRISPR-Cas systems are overcoming such challenges and offering new opportunities,promoting the integrative studies of TCM to systematically dig out the potential pharmaceutical compounds of TCMs[2-4].

Microvesicles Derived From Human Umbilical Cord Wharton's Jelly Mesenchymal Stem Cells Attenuate Bladder Tumor Cell Growth in Vitro and in Vivo

Several studies suggest that mesenchymal stem cells(MSCs) possess antitumor properties;however,the exact mechanisms remain unclear. Recently,microvesicles(MVs) are considered as a novel avenue intercellular communication,which may be a mediator in MSCs-related antitumor effect. In the present study,we evaluated whether MVs derived from human umbilical cord Wharton’s jelly mesenchymal stem cells(h WJMSCs) may inhibit bladder tumor T24 cells growth using cell culture and the BALB/c nu/nu mice xenograft model. CCK-8 assay and Ki-67 immunostaining were performed to estimate cell proliferation in vitro and in vivo. Flow cytometry and TUNEL assay were used to assess cell cycle and apoptosis. To study the conceivable mechanism by which h WJMSC-MVs attenuate bladder tumor T24 cells,we estimated the expression of Akt/p-Akt,p-p53,p21 and cleaved Caspase 3 by Western blot technique after exposing T24 cells to h WJMSC-MVs for 24,48 and 72 h. Our data indicated that h WJMSC-MVs can inhibit T24 cells proliferative viability via cell cycle arrest and induce apoptosis in T24 cells in vitro and in vivo. This study showed that h WJMSC-MVs down-regulated phosphorylation of Akt protein kinase and up-regulated cleaved Caspase 3 during the process of anti-proliferation and pro-apoptosis in T24 cells. These results demonstrate that h WJMSC-MVs play a vital role in h WJMSC-induced antitumor effect and may be a novel tool for cancer therapy as a new mechanism of cell-to-cell communication.

Several studies have shown that activation of unfolded protein response and endoplasmic reticulum(ER)stress plays a crucial role in severe cerebral ischemia/reperfusion *** occurs within hours after cerebral ischemia,but the relationship between ER stress and autophagy remains *** this study,we established experimental models using oxygen-glucose deprivation/reoxygenation in PC12 cells and primary neurons to simulate cerebral ischemia/reperfusion *** found that prolongation of oxygen-glucose deprivation activated the ER stress pathway protein kinase-like endoplasmic reticulum kinase(PERK)/eukaryotic translation initiation factor 2 subunit alpha(e IF2α)-activating transcription factor 4(ATF4)-C/EBP homologous protein(CHOP),increased neuronal apoptosis,and induced ***,inhibition of ER stress using inhibitors or by si RNA knockdown of the PERK gene significantly attenuated excessive autophagy and neuronal apoptosis,indicating an interaction between autophagy and ER stress and suggesting PERK as an essential target for regulating *** autophagy with chloroquine exacerbated ER stress-induced apoptosis,indicating that normal levels of autophagy play a protective role in neuronal injury following cerebral ischemia/reperfusion *** from this study indicate that cerebral ischemia/reperfusion injury can trigger neuronal ER stress and promote autophagy,and suggest that PERK is a possible target for inhibiting excessive autophagy in cerebral ischemia/reperfusion injury.

Metal bioaccumulation in plant leaves from an industrious area and the Botanical Garden in Beijing

The concentrations of Fe, Mn, Al, Zn, Pb, Ni, Cr, and As were measured in soils and leaves from 21 plant species growing on hills near the Beijing Steel Factory(BSF) and 17 plant species in the Beijing Botanical Garden(BBG). The results showed that soils from BSF were Zn contaminated according to the threshold of natural background of China. There was a metal contamination of the soils by Ni, and Cr in BSF comparing with those in BBG. The comparison between concentrations of metals in leaves from both sites indicated that, in general, accumulation of metals in the leaves of the same species was significantly different between the two sites. Even within the same locality each species accumulation of metals was significantly variable. The study aimed to screen landscape plants for the capacity to clean-up toxic metals in soils, and developed an overall metal accumulation index(MAI) for leaves and then categorized the MAI that can be applied broadly in the selection of species in polluted areas. To do this, the spectrum of MAI values were divided into four classes: strongly accumulated(SA or grade I), moderately accumulated(MA or grade II), intermediately accumulated(IA or grade III), and weakly accumulated(WA or grade IV). The results showed that elemental association between Fe, Al, Ni, and As was generally highly correlated with each other in the sampling sites. This may suggest their common biochemical characteristics. Generally, those species containing strong and moderate accumulation in both sites are considered including Vitex negundo, Broussonetia papyrifera, Ulmus pumila, and Rubia cordifolia. At BSF and other industrial sites with a similar ecosystem, strong and moderate accumulation species include Sophora japonica, Ampelopsis aconitifolia var. glabra, Platycladus orientalis, Wikstroemia chamaedaphne, Cleistogenes squarrosa, Grewia biloba, and in BBG, in addition Setaria viridis, Cotinus coggygria, Lespedeza floribunda, Rhamnus parvifolia, Lespedeza tomento

By the use of the 3/2 power law presented by Toba combined with the significant wave energy balance equation for wind wave, wind wave growth at a limited fetch is analytically investigated. The new wind wave growth relations (WWGRs) are analytically derived with sheltering coefficient and wind drag coefficient as parameters. The geometrical average of observational values of sheltering coefficient and the arithmetic average of observational values of wind drag coefficient are applied to determine the new WWGRs. Comparisons with existing empirical WWGRs are made.

Flexible fiber-shaped sodium dual-ion batteries(FSDIBS)as a proof of concept are fabricated by using the hierarchical ReS_(2) nanosheets anchored on the carbon nanotube(ReS_(2)@CNT)fiber as anode and graphite on the CNT as *** to large interlayer spacing and weak layer coupling force of the ReS_(2) nanosheets and the anion accommodation of the graphite combined with good flexibility of the CNT fiber,the FSDIBS demonstrate outstanding electrochemical performances with high working voltage and high specific volumetric energy density,durable cycling life,and good *** FSDIBS show a specific discharge capacity of 97.8 mAh cm^(−3) at a current density of 630 mA cm^(−3) and high specific energy density of 25.12 mWh cm^(−3)(based on the whole volume of the two electrodes)and superb stability with a capacity retention of 91.8%even after bending for 2100 ***,a series of ex situ/in situ characterizations are verified that the reversible shuttles of the Na^(+) cations and PF_(6)^(-) anions between the anode and cathode are simultaneously occurred during the charge/discharge process.

Ecological cruising control methods of vehicles have been extensively studied to further cut down energy consumption by optimizing vehicles’speed ***,most controllers cannot be put into practical application because of future terrain data requirements and excessive computational *** this paper,an eco-cruising strategy with real-time capability utilizing deep reinforcement learning is proposed for electric vehicles(EVs)propelled by in-wheel *** deep deterministic policy gradient algorithm is leveraged to continuously regulate the motor torque in response to road elevation *** comparing the proposed strategy to the energy economy benchmark optimized with dynamic programming(DP),and traditional constant speed(CS)strategy,its learning ability,optimality,and generalization performance are *** simulation results show that without a priori knowledge about the future trip,the proposed strategy provides 3.8%energy saving compared with the CS *** also yields a smaller gap than the globally optimal solution of *** testing on other driving cycles,the trained strategy reveals good generalization performance and impressive computational efficiency(about 2 ms per simulation step),making it practical and ***,the model-free characteristic of the proposed strategy makes it applicable for EVs with different powertrain topologies.
