Development and validation of a novel non-invasive test for diagnosing nonalcoholic fatty liver disease in Chinese children

With the exploding prevalence of obesity,many children are at risk of developing nonalcoholic fatty liver *** anthropometric and laboratory parameters,our study aimed to develop a model to quantitatively evaluate liver fat content(LFC)in children with *** A well-characterized cohort of 181 children between 5 and 16 years of age were recruited to the study in the Endocrinology Department as the derivation *** external validation cohort comprised 77 *** assessment of liver fat content was performed using proton magnetic resonance *** and laboratory metrics were measured in all subjects.B-ultrasound examination was carried out in the external validation *** Kruskal-Wallis test,Spearman bivariate correlation analyses,univariable linear regressions and multivariable linear regression were used to build the optimal predictive *** The model was based on indicators including alanine aminotransferase,homeostasis model assessment of insulin resistance,triglycerides,waist circumference and Tanner *** adjusted R^(2)of the model was 0.589,which presented high sensitivity and specificity both in internal[sensitivity of 0.824,specificity of 0.900,area under curve(AUC)of 0.900 with a 95%confidence interval:0.783–1.000]and external validation(sensitivity of 0.918 and specificity of 0.821,AUC of 0.901 with a 95%confidence interval:0.818–0.984).Conclusions Our model based on five clinical indicators was simple,non-invasive,and inexpensive;it had high sensitivity and specificity in predicting LFC in ***,it may be useful for identifying children with obesity who are at risk for developing nonalcoholic fatty liver disease.

Background Alanine aminotransferase(ALT)is widely used to screen patients with hepatic ***,the current reference ranges(<50 U/L)were developed by laboratories and have not been validated in populations with a large number of healthy *** This study collected venous blood and anthropometric data from a total of 13,287 healthy children aged 3 months to 18 years who underwent routine physical examinations in the Department of Pediatric *** applied the least mean square algorithm to establish age-and sex-related reference percentiles of serum levels of *** validation,we recruited 4276 children and adolescents with obesity/overweight who underwent evaluation and metabolic tests in the *** receiver operating characteristic curves,we determined age-and sex-specific upper limit percentiles of liver enzymes for fatty liver *** This study revealed a significant correlation between serum transaminase levels and age and sex(P<0.01).These transaminase levels exhibited age-and sex-specific *** individuals in the non-alcoholic fatty liver disease(NAFLD)cohort,elevated ALT levels displayed a positive association with clinical markers of disease severity,including homeostatic model assessment of insulin resistance,waist-hip ratio,and serum uric acid levels(P<0.01).According to the receiver operating characteristic curves,ALT levels at the 92.58th percentile for boys and the 92.07th percentile for girls yielded the highest accuracy and *** This study provides age-and sex-specific reference ranges for ALT,aspartate aminotransferase,andγ-glutamyltransferase in Chinese children and adolescents,making it the largest population study to ***,the study establishes a precise upper limit for ALT levels,facilitating their use in NAFLD screening.

A rabbit model of pediatric nonalcoholic steatohepatitis: The role of adiponectin

AIM： To create a rabbit model of pediatric nonalcoholic steatohepatitis （NASH） and to evaluate the role of adiponectin in the process. METHODS： Thirty-two specific pathogen-free male New Zealand rabbits were divided randomly into three groups：（1） the normal control group （n = 10） was fed with standard diet for 12 wk; （2） the model group A （n = 11）; and （3） model group B （n = 11） were fed with a highfat diet （standard diet ＋ 10% lard ＋ 2% cholesterol） for 8 and 12 wk, respectively. Hepatic histological changes were observed and biochemical parameters as well as serum levels of adiponectin, interleukin （IL）-6, IL-10 and tumor necrosis factor （TNF）-α were measured. RESULTS： Typical histological hepatic lesions of NASH were observed in both model groups described as liver steatosis, liver inflammatory infiltration, cytologic ballooning, perisinusoidal fibrosis and overall fibrosis. Compared with the normal control group, there were significant increases in model groups A and B in weight gain （1097.2 ± 72.3, 1360.5± 107.6 vs 928.0 ±58.1, P 〈 0.05, P 〈 0.01）, liver weight （93.81±6.64, 104.6±4.42 vs 54.4±1.71, P 〈 0.01）, Lg （ALT）（1.9±0.29, 1.84± 0.28 vs 1.60±0.17, P 〈 0.01）, and Lg （TG）（1.03 ±0.24, 1.16 ±0.33 vs 0.00 ±0.16, P 〈 0.01）. Weight gain was much more in model group B than in model group A （1360.5± 107.6 vs 1097.2 ±72.3, P 〈 0.05）. But, there was no significant difference between the two groups concerning the other indexes. Pro-inflammatory cytokines （IL-6 and TNF-α） increased in model group B compared with that of control and model group A （IL-6：1.86±0.21 vs 1.41 ±0.33, 1.38± 0.42, P 〈 0.01; TNF-α： 1.18±0.07 vs 0.66 ±0.08, 0.86 ±0.43, P 〈 0.01, P 〈 0.05）, whereas serum adiponectin and IL-10 decreased in model groups compared with that in the control （adiponectin： A： 21.87±4.84 and B： 21.48 ±4.60 vs 27.36 ±7.29, P 〈 0.05. FTO gene polymorphisms and obesity risk in Chinese population: a meta-analysis

Evaluation of Auto-Ignition Characteristics of Diesel Surrogate Fuels with the Same Derived Cetane Numbers at Changed Thermodynamic Environments

The ignition tendency of diesel fuels is highly sensitive to ambient conditions and fuel *** this study,the ignition characteristics of different diesel surrogate fuels with the same derived cetane numbers(DCN)were measured and compared in varied thermodynamic and oxidizing *** combustion pressures,heat release rates,ignition delays,and combustion delays of the test fuels were *** experimental results showed that the diesel surrogate fuels with the same DCNs exhibit similar ignition propensity at standard DCN test ***,for the test conditions of high cetane fuels,high ambient temperatures,and suficient oxygen concentrations,surrogate fuels with the same DCN have similar ignition behaviors,and using the DCN to evaluate fuel ignition tendency is ***,for the test conditions of low cetane fuels,low ambient temperatures,and reduced oxygen concentrations,different ignition behaviors are observed for the surrogate fuels with the same DCN,so at these conditions using DCN as the evaluation index for fuel ignition tendency may lead to higher uncertainty.

Regulated polarization degree of upconversion luminescence and multiple anti-counterfeit applications

Polarization upconversion luminescence(PUCL)of lanthanide ions(Ln^(3+))has been widely used in single particle tracking,microfluidics detection,three-dimensional displays,and so ***,no effective strategy has been developed for modulating *** we report a strategy to regulate PUCL in Ho^(3+)-doped NaYF4single nanorods based on the number of upconversion *** constructing a multiphoton upconversion system for Ho^(3+),we regulate the degree of polarization(DOP)of PUCL from 0.590 for two-photon luminescence to 0.929 for three-photon upconversion luminescence(UCL).Furthermore,our strategy is verified from cross-relaxation between Ho^(3+)and Yb^(3+),excitation wavelength,excitation power density,and local site *** this regulation strategy of PUCL has also been achieved in Tm^(3+),with the DOP ranging from 0.233 for two-photon luminescence to 0.925 for four-photon UCL Besides,multi-dimensional anti-counterfeiting display has been explored with *** work provides an effective strategy for regulating PUCL and also provides more opportunities for the development of polarization display optical encoding,anti-counterfeiting,and integrated optical devices.
