Influenza virus(IFV) infection is a major cause of severe respiratory illness that affects almost every country in the world. IFV infections result in respiratory illness and even acute lung injury and death, but the underlying mechanisms responsible for IFV pathogenesis remain to be fully elucidated. In this study, we found that the basic fibroblast growth factor2(FGF2) level was markedly increased in H1N1 virus-infected humans and mice. FGF2, which is predominately derived from epithelial cells, recruits and activates neutrophils via the FGFR2-PI3K-AKT-NFκB signaling pathway. FGF2 depletion or knockout exacerbated influenza-associated disease by impairing neutrophil recruitment and activation. More importantly, administration of recombinant FGF2 protein significantly alleviated the severity of IFV-induced lung injury and promoted the survival of IFV-infected animals that was mediated by the recruited neutrophils. These findings demonstrate that FGF2 plays a critical role in preventing IFV-induced lung injury and suggest FGF2 as a promising potential therapeutic target during IFV infection.

Introduction:In order to enhance the effectiveness of resource allocation,regions must tailor their responses to their specific epidemiological and economic ***:Utilizing Spectrum software,we projected the cost-effectiveness of 10 chosen HIV interventions in six cities in eastern China from 2019 to *** assessed three scenarios—Base,Achievable,and Idealized—for each *** analysis included the projected number of HIV infections and deaths averted,as well as the incremental costeffectiveness ratios for each intervention in the six ***:In Shijiazhuang,Wuxi,Yantai,and Zhenjiang,cities with initially low antiretroviral therapy(ART)coverage,ART showed significant effectiveness,especially for ***,in Foshan and Ningbo,where ART coverage was notably high,oral pre-exposure prophylaxis(PrEP)for men who have sex with men(MSM)proved effective in the Idealized *** outreach,ART for males,and ART for females demonstrated cost-effectiveness across all six cities in both Achievable and Idealized scenarios at the predefined thresholds for each ***:Maintaining an appropriate coverage rate for outreach to MSM can lead to *** cities with low ART coverage,scaling up ART remains a crucial *** regions with high ART coverage,consideration may be given to the utilization of oral PrEP for MSM individuals,requiring budget allocation.

糖尿病肾病(DKD)是糖尿病最常见、最严重的慢性微血管并发症之一,是导致终末期肾病(ESRD)的重要原因。DKD的详细病因及发病机制尚不清楚,目前尚无有效的治疗方法,主要通过有效控制血糖、血压及脂代谢紊乱、降低尿蛋白等因素,延缓DKD进程。阿魏酸(FA)是川芎、当归等中药的有效成分之一,具有抗血小板聚集、促进血小板解聚、抗血栓、抗氧化、抗自由基等多方面的生物活性。阿魏酸哌嗪(PF)是通过FA人工合成的衍生物,大量的体内外实验表明PF可以通过影响肾脏损伤的潜在机制延缓DKD进展。本文就PF治疗DKD的研究现状及相关作用机制作一综述。

In face of the everlasting battle toward COVID-19 and the rapid evolution of SARS-CoV-2,no specific and effective drugs for treating this disease have been reported until ***-converting enzyme 2(ACE2),a receptor of SARS-CoV-2,mediates the virus infection by binding to spike *** ACE2 is expressed in the lung,kidney,and intestine,its expressing levels are rather low,especially in the *** the great infectivity of COVID-19,we speculate that SARS-CoV-2 may depend on other routes to facilitate its ***,we first discover an interaction between host cell receptor CD147 and SARS-CoV-2 spike *** loss of CD147 or blocking CD147 in Vero E6 and BEAS-2B cell lines by anti-CD147 antibody,Meplazumab,inhibits SARSCoV-2 *** of human CD147 allows virus entry into non-susceptible BHK-21 cells,which can be neutralized by CD147 extracellular *** loads are detectable in the lungs of human CD147(hCD147)mice infected with SARS-CoV-2,but not in those of virus-infected wild type ***,virions are observed in lymphocytes of lung tissue from a COVID-19 *** T cells with a property of ACE2 natural deficiency can be infected with SARS-CoV-2 pseudovirus in a dosedependent manner,which is specifically inhibited by ***,CD147 mediates virus entering host cells by ***,our study reveals a novel virus entry route,CD147-spike protein,which provides an important target for developing specific and effective drug against COVID-19.

目的探究中性粒细胞在蓖麻毒素(Ricin Toxin,RT)致毒过程中的作用,寻找解毒的有效策略。方法采用10×Genomics单细胞转录组测序技术对中毒小鼠外周血单个核细胞(PBMCs)进行转录组测序及分析,并通过流式细胞术测定目的细胞亚群。结果经降维聚类、差异基因、拟时序分析结果显示CD177^(-)CD121b^(+)CCRL2^(+)中性粒细胞亚群为RT致毒过程关键亚群,随后通过流式细胞术测定显示该亚群在中毒后6小时在中性粒细胞中的比例显著升高。同时利用CCRL2抗体治疗中毒小鼠其保护率达60%。结论本研究首次发现在RT中毒后中性粒CD177^(-)CD121b^(+)CCRL2^(+)细胞亚群呈现显著升高,同时CCRL2抗体能够有效保护RT中毒小鼠,为RT致毒机制研究提供新的思路,为其治疗提供新的策略。
