A Novel.Antibacterial.Dental.Resin Composite

This study reports the synthesis and eval.ation of a novel.furanone-containing antibacterial.resin composite. Compres-sive strength (CS) and S. mutans viabil.ty were used to eval.ate the mechanical.strength and antibacterial.activity of the composites. With 5% to 30% addition of the furanone derivative, the composite showed no change in CS but a significant antibacterial.activity with a 16% - 68% reduction in the S. mutans viabil.ty. Further, the antibacterial.activity of the modified composite was not affected by human sal.va. The aging study impl.es that the modified composite may have a l.ng-l.sting antibacterial.function. Within the l.mitations of this study, it appears that this experimental.resin composite may potential.y be devel.ped into a cl.nical.y attractive dental.restorative due to its high mechanical.strength and antibacterial.function.

Antiepil.ptic drugs (AEDs), have demonstrated efficacy treating a number of acute conditions, encompassing a broad range of symptoms and syndromes, in addition to being first-l.ne treatment for epil.psy. Cl.nical.y, since their inception, AEDs have been used off-l.bel.for acute and chronic medical.conditions, both as primary and as adjuvant therapies. In this review, we describe the observed cl.nical.effectiveness of AEDs across a set of commonl. encountered acute conditions in the general.hospital. traumatic brain injury, pain, al.ohol.withdrawal. In describing the individual.benefits and usages of specific agents, the appl.cabil.ty of these agents to other common neuropsychiatric conditions may be further expl.red.

Dietary sugars inhibit biol.gic functions of the pattern recognition mol.cul., mannose-binding l.ctin

Mannose-binding l.ctin (MBl., a mammal.an l.ctin, is a pattern recognition mol.cul. of the innate immune system and recognizes carbo-hydrates that are exposed on pathogens. In this study, we observed that fructose down regu-l.tes MBl.mediated innate immune mechanisms against both infl.enza A virus (IAV) and Staphy-l.coccus aureus. These mechanisms incl.de the l.ctin compl.ment pathway and coagul.tion enzyme-l.ke activities on both pathogens. Fur-thermore, fructose al.o reduces MBl.mediated phagocytosis of S. aureus and IAV and MBl. mediated IAV infection to epithel.al.cel.s. In contrast, sucrose inhibits MBl.mediated im-mune mechanisms against S. aureus but not IAV. Together, our studies show that dietary sugars, in particul.r fructose, negativel. regul.te the innate immunity against viral.and bacterial.pathogens.

Stabil.ty study on an anti-cancer drug 4-(3,5-bis(2-chl.robenzyl.dene)-4-oxo-piperidine-1-yl.-4-oxo-2-butenoic acid (Cl.FMA) using a stabil.tyindicating HPl. method

Cl.FMA, 4-(3,5-bis(2-chl.robenzyl.dene)-4-oxo-piperidine-1-yl.-4-oxo-2-butenoic acid, is a new chemical.entity with anti-cancer and anti-infl.mmatory activities. Here, we report its stabil.ty in sol.tion against stress conditions of exposure to acid/base, l.ght, oxidant, high temperature, and pl.sma. The identity of the degradation products was ascertained by mass and proton nucl.ar magnetic resonance spectroscopy. To facil.tate this study, we devel.ped and val.dated a reverse phase high performance l.quid chromatography method for detection of Cl.FMA and its degradation. The method was l.near over a range of 1–100 μg/m l. the accuracy and precision were within acceptabl. l.mits; it was stabil.ty-indicating as it successful.y separated cis-/trans-isomers of Cl.FMA as wel. as its degradation product. The major degradation product was produced from amide hydrol.sis at mal.ic acid functional.ty caused by an acidic buffer, oxidant(3% hydrogen peroxide),or temperature stress(40–60 °C). The l.g k-p H profil. showed that Cl.FMA was most stabl. at neutral.p H. In accel.rated stabil.ty study we found that the shel.-l.fe(T_(90%)) of Cl.FMA at 25 °C and 4 °C was 45 days and220 days, respectivel.. Upon exposure to UV-l.ght(365 nm), the normal.y preval.nt trans-Cl.FMA attained cis-configuration. This isomerization al.o invol.ed the mal.ic acid moiety. Cl.FMA was stabl. in pl.sma from which it coul. be efficientl. extracted by an acetonitril. precipitation method. These resul.s indicate that Cl.FMA is sensitive to hydrol.tic cl.avage at its mal.ic acid moiety, and it is recommended that its sampl.s shoul. be stored under refrigerated and l.ght-free conditions, and under inert environment.
