Smartphone-Imaged HIV-1 Reverse-Transcription loop-Mediated Isothermal Ampliflcation(RT-lAMP) on a Chip from Whole Blood

Viral load measurements are an essential tool for the long-term clinical care of human immunodeficiency virus （HIV）-positive individuals. The gold standards in viral load instrumentation, however, are still too limited by their size, cost, and sophisticated operation for these measurements to be ubiquitous in remote settings with poor healthcare infrastructure, including parts of the world that are disproportionately affected by HIV infection. The challenge of developing a point-of-care platform capable of making viral load more accessible has been frequently approached but no solution has yet emerged that meets the practical requirements of low cost, portability, and ease-of-use. In this paper, we perform reverse-transcription loop-mediated isothermal amplification （RT-lAMP） on minimally processed HIV-spiked whole blood samples with a microfluidic and silicon microchip platform, and perform fluorescence measurements with a consumer smartphone. Our integrated assay shows amplification from as few as three viruses in a - 60 nl RT- lAMP droplet, corresponding to a whole blood concentration of 670 viruses per μl of whole blood. The technology contains greater power in a digital RT-lAMP approach that could be scaled up for the determination of viral load from a finger prick of blood in the clinical care of HIV-positive individuals. We demonstrate that all aspects of this viral load approach, from a drop of blood to imaging the RT-lAMP reaction, are compatible with lab-on-a-chip components and mobile instrumentation.

Purpose Jump-landing assessments provide a means to quantify an individual’s ability to attenuate ground reaction forces,generate lower limb explosive power and maintain joint *** order to identify risk factors that can be targeted through appropriate training interventions,it is necessary to establish which(scalar)objective kinetic,kinematic,and performance measures are most associated with lower-extremity *** Online searches of MEDlINE,SCOPUS,EBSCOHost,SPORTDiscus and PubMed databases were completed for all articles published before March 2020 in accordance with PRISMA *** 40 articles investigating nine jump-landing assessments were included in this *** 79%of studies using drop jump(n=14)observed an association with future injury,while only 8%of countermovement jump studies(n=13)observed an association with injury *** 57%of studies using unilateral assessments found associations with risk of injury(n=14).Studies using performance measures(jump height/distance)as outcome measure were only associated with injury risk in 30%of ***,those using kinetic and/or kinematic analyses(knee abduction moment,knee valgus angle,knee separation distance,peak ground reaction force)found associations with injury in 89%of *** The landing element of jump-landing assessments appears to be superior for identifying individuals at greater risk of injury;likely due to a closer representation of the injury ***,jump-landing assessments that involve attenuation of impact forces such as the drop jump appear most suited for this purpose but should involve assessment of frontal plane knee motion and ground reaction forces.

Stocking of lake Kinneret by exotic fish species (Mugilids: Mugil cephalus and liza ramada) started in 1958. The rational stocking of lake Kinneret with Mugilids was double functioned: To improve fishermen income and the water quality in the lake. Mugilids are chosen as introduction target because their market value is high, the fish cannot reproduce in the lake and fingerlings are low cost available. During 1960-2015, 56.2 million fingerlings were introduced into the lake approximately averaged one million per year of M. cephalus (MC) and l. ramada (lR). The study of the Mugilids food composition has indicated consumption of suspensoids, mostly detritial organic particles. Therefore no damage to water quality is suggested. The best fitness between recruitment to Dock on Side landings and stocking capacity was evaluated at 3 years interval. Differences were found between recruitments of MC and lR to commercial catches by individual averaged age and weight. The long term success of Mugilid introduction to lake Kinneret is confirmed.

Identification and differentiation of PDX1 β-cell progenitors within the human pancreatic epithelium

Synthesis and evaluation of a novel antibacterial dental resin composite with quaternary ammonium salts

The novel quaternary ammonium bromide (QAB)-containing oligomers were synthesized and applied for developing an antibacterial resin composite. Compressive strength (CS) and S. mutans (an oral bacteria strain) viability were used to evaluate the mechanical strength and antibacterial activity of the formed composites. All the QAB-modified resin composites showed significant antibacterial activity and mechanical strength reduction. Increasing chain length and loading significantly enhanced the antibacterial activity but dramatically reduced the CS as well. The 30-day aging study showed that the incorporation of the QAB accelerated the degradation of the composite, suggesting that the QAB may not be well suitable for development of antibacterial dental resin composites or at least the QAB loading should be well controlled, unlike its use in dental glass-ionomer cements. The work in this study is beneficial and valuable to those who are interested in studying antibacterial dental resin composites.

Drying conditions in Switzerland-indication from a 35-year landsat time-series analysis of vegetation water content estimates to support SDGs

Exacerbated by climate change,Europe has experienced series of hot and dry summer since the beginning of the 21st *** importance of land conditions became an international concern with a dedicated sustainable development goal(SDG),the SDG *** calls for developing and finding innovative solu-tions to follow and evaluate impacts of changing land condi-tions induced by various driving *** Switzerland,drought risk will significantly increase in the coming decades with severe consequences on agriculture,energy production and *** this paper,we used a 35-year satellite-derived annual and seasonal times-series of normalized difference water index(NDWI)to follow vegetation water content evolution at different spatial and temporal scales across Switzerland and related them to temperature and precipitation to investigate possible responses of changing climatic *** indicate that there is a small and slow drying tendency at the country scale with a NDWI mean decreasing slope of−0.22%/year for the 23%significant pixels across *** tendency is mostly visible below 2000 m above sea level(m.a.s.l.)and in all biogeographical *** Southern Alps regions appear to be more responsive to changing drying conditions with a significant and slight negative NDWI trend(−0.39%/year)over the last 35 ***,NDWI values are mostly a func-tion of temperature at elevations below the tree line rather than *** suggest that multi-annual and seasonal NDWI can be a valuable indicator to monitor vegetation water content at different scales,but other components such as land cover type and evapotranspiration should be considered to better characterize NDWI *** Earth Observations data can provide valuable complementary obser-vations for national statistics on the ecological state of vegeta-tion to support SDG 15 to monitor land affected by drying conditions.

High Performance liquid Chromatographic Identification and Estimation of Phthalates in Sewer Waste and a Receiving River in Ibadan City, Southwestern Nigeria

Medical wastes have been implicated in river pollution in developing countries and most often people depend on water from such rivers for sources of livelihood. Phthalates (endocrine disruptors) are major components in medical wastes and are commonly found contaminants in aquatic environment. Most sewage treatment facilities handling medical waste are inefficient due to overuse and poor maintenance and discharge directly into rivers. This study aimed to investigate the identity and estimates the concentration of phthalates in supposed treated medical wastes from a hospital sewer and water from a receiving river. Samples were randomly collected before and after treatment by the sewer plants, while samples were randomly collected along the course of the river starting from point of discharge. Control samples were taken from upstream about 500 m along the river course. The samples were extracted by liquid-liquid chroma- tographic process using dichloromethane, after which they were cleaned up in a column of silica gel using hexane as the mobile solvent. The cleaned extracts were analyzed by HPlC. The concentrations (μg/l) of dimethyl-, diethyl-, diphenyl-, dibutyl- and di-(2-ethyl)hexyl phthalates ranged from 62.81 ± 18.53;4.74 ± 3.57;2.05 ± 1.80;11.40 ± 5.58 to 141.92 ± 35.8 respectively in the sewer waste. The receiving river had a concentration (μg/l) of 9.17 ± 14.02;0.18 ± 0.31;0.48 ± 0.84;2.84 ± 1.21;61.72 ± 38.35 respectively for dimethyl-, diethyl-, diphenyl-, dibutyl- and di-(2-ethyl) hexyl phthalates. These concentrations were higher than control and far exceeded the USEPA limits of 3 μg/l recom- mended for phthalates in water. Contaminants of aquatic environment by untreated wastes from hospitals has serious implications on public health and environment as human risks for phthalate esters downstream are high and this calls for urgent need to develop strategy to build incentives for compliance in treatment and discharge of wastes into river waters.

Assessment of Groundwater Quality and Saline Intrusions in Coastal Aquifers of lagos Metropolis, Nigeria

As a result of immense industrialisation and high population growth, groundwater is heavily relied on in lagos metropolis to serve as an alternative source of water where surface water is seriously polluted. The continued reliance on ground water has resulted in its decline in quantity and quality. In this study, the coastal aquifers of lagos metropolis were selected for an assessment of its groundwater quality and impact of saline intrusion. Water samples collected along the coastal region were subjected to various physicochemical analyses. Results obtained were compared with permissible values for drinking water stated by Federal Environmental Protection Agency (FEPA) and World Health Organization (WHO). The results revealed that all the water samples were significantly hard (range 522.14 – 1233.34mg/l). The salinity was delineated by conductivity measurements. Three samples had specific conductance above the stated limits for fresh water. The samples however met the stipulated limits for drinking water for the other tested parameters.

A Novel Antibacterial Dental Resin Composite

This study reports the synthesis and evaluation of a novel furanone-containing antibacterial resin composite. Compres-sive strength (CS) and S. mutans viability were used to evaluate the mechanical strength and antibacterial activity of the composites. With 5% to 30% addition of the furanone derivative, the composite showed no change in CS but a significant antibacterial activity with a 16% - 68% reduction in the S. mutans viability. Further, the antibacterial activity of the modified composite was not affected by human saliva. The aging study implies that the modified composite may have a long-lasting antibacterial function. Within the limitations of this study, it appears that this experimental resin composite may potentially be developed into a clinically attractive dental restorative due to its high mechanical strength and antibacterial function.
