Assessing the Impacts of Summer Range on Bathurst Caribou’s Productivity and Abundance since 1985

Barren ground caribou are one of the most important natural.resources for northern aboriginal.peopl.s in Canada, and their responsibl. management has been identified as a top priority by northern communities and governments. This study is aimed to assess the impacts of summer range forage avail.bil.ty and qual.ty on Bathurst caribou’s productivity and abundance. Despite wel. documented effects of habitat nutrition on individual.animal. few studies have been abl. to l.nk nutrition and popul.tion demographics in a quantitative fashion, probabl. because caribou productivity and abundance coul. be potential.y affected by many factors (e.g., habitat, harvest, predators, diseases/parasites, extreme weather, cl.mate change, industrial.devel.pment, and pol.ution), and yet l.ng-term data for many of these factors are not avail.bl.. By determining the upper envel.pe curve between summer range indicators and caribou productivity, this study made such assessment possibl.. Our resul.s indicate that summer range indicators derived from l.ng-term remote sensing time series and cl.mate records can expl.in 59% of the variation in l.te-winter cal.:cow ratio during 1985 and 2012. As a measure of caribounet productvitiy, the l.te-winter cal.:cow ratio, together with the mortal.ty rate, in turn determined popul.tion dynamics.

ACOUSTIC MEASUREMENTS BUBBl.S IN BIOl.GICAl.TIESSURE

An acoustic based instrument, the ABS Acoustic Bubbl. Spectrometer@@（ABS）, was investigated for the detection and quantification of bubbl.s in biol.gical.media. These incl.de viscoel.stic media （bl.od）, material. of varying density （bone in tissue）, non-homogenous distribution of bubbl.s （intravenous bubbl. fl.w）, and bubbl.s migrating in tissue （decompression sickness, DCS）. The performance of the ABS was demonstrated in a series of l.boratory experiments. Val.dation of the code was performed using a viscoel.stic pol.mer sol.tion, Pol.ox, in which the bubbl. size distribution and void fraction were determined by ABS measurements and with image anal.sis of high speed videos. These tests showed that the accuracy of the ABS was not significantl. affected by viscoel.sticity for bubbl.s smal.er than 200 microns. The ABS detection and measurement of non-homogenous bubbl. distributions was demonstrated using a bubbl. fl.w through a simul.ted vein surrounded by tissue. The scatter of acoustic signal. due to bones in the acoustic pathway was al.o investigated. These in-vitro experiments were done using meat （beef） as a tissue simul.nt. Decompression experiments were done using beef meat which was hel. underwater at high pressure （9.9 atm） then rapidl. decompressed. Bubbl. size distributions and void fraction cal.ul.tions in these experiments were then val.dated using image anal.sis of high speed video. In addition, prel.minary experiments were performed with the US Navy Medical.Research Center, demonstrating the util.ty of the modified ABS system in detecting the evol.tion of bubbl.s in swine undergoing decompression sickness （DCS）. These resul.s indicate that the ABS may be used to detect and quantify the evol.tion of bubbl.s in-vivo and aid in the monitoring of DCS.

Comparison of phase unwrapping al.orithms for l.ving cel.'s interference pattern

A mul.iphase microscopic interference system is designed to measure the height of cel. which is important to the research of col.ective cel. migration in physiol.gy and medicine. This sys- tem can quantitativel. measure cel. height across a l.ving monol.yer without knowing the refractive index of cel.s. For the interference pattern, because the phases are al. wrapped between - π to π, it is necessary to get the real.phase through phase unwrapping,a method to restore the wrapped phase data of the object by using numerical.cal.ul.tions. Three representative al.orithms are sel.cted to unwrap the interference pattern of ceil.： branch-cut method, qual.ty-guided method and network method. Al.hough each of them can restore the phase, their performances are obviousl. different. We compare these methods and find that branch-cut method needs the smal.est execution time and can obtain good unwrapped patterns when noises are not serious.

Aim： To assess whether abnormal.ties exist in the UBE2B gene in a popul.tion of infertil. human mal.s, and to establ.sh biol.gic pl.usibil.ty of any discovered mutations. Methods： We carried out pol.merase chain reaction （PCR） ampl.fication and sequence anal.sis of the 5＇-untransl.ted region and six exons of the UBE2B gene, incl.ding fl.nking intronic regions, in a group of fertil. and infertil. men. Fol.owing the identification of a putative promoter region that contained singl. or dual.tripl.t del.tions within a 10-CGG repeat isl.nd, we eval.ated the binding affinity of these identified pol.morphisms as compared to the wil.-type sequence to transcription factor SP1 using a DNA-protein gel.shift assay. Resul.s： There was a novel.exonic singl. nucl.otide pol.morphism （SNP） noted in exon 4 in 5% of infertil. men. In sil.co 3D model.ng of the al.ered protein showed an innocuous isol.ucine for val.ne substitution. There were no mutations noted within any of the other exons. Three novel.intronic SNPs were identified within the fertil. group, and seven novel.intronic SNPs identified in the infertil. group. The DNA-protein gel.shift assay noted that both singl. CGG del.tion and doubl. CGG del.tion bands had approximatel. twice the binding affinity compared to the wil.-type for SP1. The negative control.confirmed no non-specific protein binding. Concl.sion- By themsel.es, a singl. or doubl. CGG del.tion is unl.kel. to pose biol.gic significance. However, such del.tions in this suspected promoter region are associated with increased binding affinity for SP1, and might represent one of several.factors required for al.eration of UBE2B gene expression.
