Comparison of total immunoglobulin A levels in different samples in Leghorn and broiler chickens

Objective: To standardize an ELISA protocol to quantify total immunoglobulin A(IgA)from different biological ***: Two independent experiments were conducted. In Experiment 1, total IgA levels were quantified from the lachrymal fluid, tracheal swab, and cloacal swab at various time points from Days 30 to 89 in white Leghorn chickens. Experiment 2 was conducted to evaluate the effect of 50 or 500 ppb of aflatoxin b1(AFb1) on total IgA quantified in samples from the lachrymal fluid, tracheal swab, gut content and cloacal swab in broiler chickens at 21 days of ***: Lachrymal fluid contained the highest level of IgA; however, the sampling procedure was time consuming and stressful to the bird, and the sample volume depends largely on the size of the chicken. Cloacal swabs also contained a high concentration of IgA; this sampling procedure was faster than lachrymal fluid sampling and was not affected by the age of the bird. Tracheal sampling was more difficult than cloacal sampling; the age of the bird limited the sampling, and the IgA concentration was the lowest detected at all sampling ages. 500 ppb of AFb1 significantly reduced total IgA concentration in the gut content compared with control or 50 ppb of AFb1 treated ***, a significant reduction in total IgA was also observed in those chickens that received 50 ppb of AFb1 in gut rinse when compared with cloacal ***: The results of this study suggest that cloacal swab is an easy and reliable way to evaluate mucosal IgA concentration in both Leghorn and broiler chickens.

The concept of an integrated "lab on a chip" has long been a goal for the micro-electro-mechanical-systems(MEMS) *** would entail the integration of not only the sampling and analysis of various functions,but also the ability to transmit this information off the chip to a central *** paper describes the initial steps in the fabrication of a "lab on a chip" which would continually analyze blood sampled via microneedles using techniques such as nano plasmonics,specifically,concentrations of *** analysis could then be transmitted off the chip using digital signal *** paper describes the analysis and optimization of the microneedle shape and size and the fabrication of the resulting needles in silicon using deep reactive ion etching(DRIE).The paper also describes the opportunities for fabrication of such needles in alternative materials and describes the issues that still have to be overcome before such an integrated device is realized.

AIM To characterize antiviral therapy eligibility among hepatitis b virus(HbV)-infected adults at a university hospital in *** Hepatitis b surface antigen-positive adults(n = 160) who were h IV-negative and referred to the hospital after a routine or clinically-driven HbV test were enrolled. Alanine Aminotransferase(ALT),Aspartate Aminotransferase(AST),platelet count,hepatitis b e-antigen,and HbV DNA were measured. Liver fibrosis/cirrhosis was assessed by physical examination,AST-to-platelet ratio index,and transient elastography. In antiviral therapy-na?ve individuals,we described hbV stages and antiviral therapy eligibility per World health Organization(WhO) and by hbV test(routine vs clinical). Elevated ALT was > 19 in women and > 30 U/L in men. Among treatmentexperienced individuals,we described medication side effects,adherence,and viral *** The median age was 33 years,71.9% were men,and 30.9% were diagnosed with HbV through a clinicallydriven test with the remainder identified via routine testing(at the blood bank,community events,etc.). Among 120 treatment-na?ve individuals,2.5% were categorized as immune tolerant,11.7% were immune active,35.6% were inactive carriers,and 46.7% had an indeterminate phenotype. Per WhO guidelines,13(10.8%) were eligible for immediate antiviral therapy. The odds of eligibility were eight times higher for those diagnosed at clinical vs routine settings(adjusted odds ratio,8.33; 95%CI: 2.26-29.41). Among 40 treatmentexperienced hbV patients,virtually all took tenofovir,and a history of mild side effects was reported in 20%. Though reported adherence was good,12 of 29(41.4%) had HbV DNA > 20 IU/m L. CONCLUSION Approximately one in ten HbV-monoinfected Zambians were eligible for antivirals. Many had indeterminate phenotype and needed clinical follow-up.

Rapid Genomic Evolution of a Non-virulent Coxsackievirus b3 in Selenium-deficient Mice

Keshan disease, an endemic cardiomyopathy in China, can be prevented with selenium (Se) supplementation. However, the seasonal and annual nature of the disease suggests that an infectious co-factor is required along with a deficiency in Se. Using a murine model of coxsackievirus b3 (CVb3) -induced myocarditis, Se-deficient mice were shown to be more susceptible to the cardiopathologic effects of the virus. In addition, a normall benign strain of CVb3 becomes virulent in Se-deficient mice. This change in virulence was shown to be due to point mutations in the viral genorne. Although the mechanism of the viral mutation is not known, the oxidative stress status of the Se-deficient host may play a role, either by directly affecting the virus and/or affecting host immune defenses

构造控制的热液白云岩（HTD）储层相和相关的淋滤灰岩都是北美洲重要的油气产层，而在全球范围的勘探中也日益得到重视。这些储层分布在密歇根盆地、阿巴拉契亚盆地以及加拿大东部和美国其他盆地奥陶系（局部为志留系和泥盆系）的多个构造带、西加拿大沉积盆地的泥盆系和密西西比系地层、大西洋裂谷边缘的侏罗系以及阿拉伯湾和其他地区的侏罗系-白垩系层位。热液白云石化系指埋藏条件下的白云石化，通常深度不大，作用流体（一般都是真正的盐水）的温度和压力均高于附近主岩地层的环境条件。主岩地层通常为灰岩。热液白云岩储层相成因证据的获得，需要综合使用埋藏-热作用史图、流体包裹体温度数据和就位时间的制约因素。热液白云岩储层相是热液矿床系列的组成部分，这一系列还包括了沉积-喷发型铅锌矿和以热液白云岩为主岩的密西西比河谷型硫化物矿床。以上三类矿床都明显受控于扭张和／或走滑（扭）断层，而流体流动一般均集中在扭张和膨胀的构造部位以及上盘。扭断层负花状构造上方的扭张凹陷是热液白云岩储层相的有利布井位置。交代型和孔隙充填型的鞍状白云岩都是热液白云岩储层相的典型特征。在许多储层中，基质交代白云岩和鞍状白云岩差不多是在同期形成的，而且具有相同的流体和相同的温度条件。对于白云石化的横向延伸范围、所形成白云岩的结构、孔隙类型和孔隙体积而言，原始主岩相均有重大影响。角砾岩、条带状组构、剪切微裂缝和其他岩石特征均记录了此类储层的短期剪切应力和孔隙流体压力瞬时变化，在活动断层附近更是如此。高温热液的间歇性作用（脉动）可能使主灰岩的干酪根发生蚀变，这一过程可称为“强制成熟作用”。基底高地、下伏砂岩（和／或碳酸盐岩？）含水层（很可能超压）以及上覆和内部的页岩封闭层和弱透水层，也都可以限制或影响HTD的就位。虽然仍存在许多问题和不确定性，尤其是在镁和盐水的来源以及质量平衡等方面，但对HTD成藏层带的认识和积极探索仍在继续发展。三维地震成像和地震异常成图技术的频繁使用，再加上对构造凹陷线性走向带的斜向水平钻井，都有助于降低这种储层的勘探开发风险。
