The wheat midge, sitodiplosis mosellana, is a serious pest of wheat worldwide. In North America, management of s. mosellana in spring wheat relies on the timely application of pesticides, based on midge adults levels caught in pheromone traps or seen via field scouting during wheat heading. In this context, biopesticides can be an effective alternative to pesticides for controlling s. mosellana within an Integrated Pest Management program. A field study using insect pathogenic fungus Beauveria bassiana GHA, nematode steinernema Jeltiae with Barricade polymer gel 1%, pyrethrin, combined formulations of B. bassiana GHA and pyrethrin, Jasmonic acid (JA) and chlorpyrifos (chemical check) was performed to determine to which extent they affect midge larval populations, kernel damage levels, grain yield, and quality, and the impacts on adult parasitoid Macroglenes penetrans populations. The results indicated that biopesticides JA and s. Jeltiae were the most effective in reducing larval populations and kernel damage levels, and produced a higher spring wheat yield when compared to the water control at both study locations (East Valier and North Valier, Montana, UsA). Increased test weight in wheat had been recorded with two previous biopesticides at East Valier but not for North Valier, when compared over water control. These results were comparable in efficacy to the chlorpyrifos. This study also suggested that B. bassiana and pyrethrin may work synergistically, as exemplified by lower total larval populations and kernel damage levels when applied together. This study did not demonstrate the effect of any treatments on M. penetrans populations.

AIM: To find out the outcome of laser photocoagulation in clinically significant macular edema (CsME) by optical coherence tomography (OCT). · METHODs: It was a prospective, non-controlled, case series study enrolling 81 eyes of 64 patients with CsME between August 2008 and January 2010. All patients received modified grid photocoagulation with frequency doubled Nd: YAG laser. Each patient was evaluated in terms of best-corrected visual acuity (BCVA) and regression or progression of maculopathy after laser therapy at 1, 3 and 6 months. spearman’s correlation test was used to show the correlation between BCVA and total macular volume (TMV). Analysis of variance (ANOVA) was used to compare among groups and independent t-test was used to compare in each group. · REsULTs: There is high correlation between BCVA and TMV (P ≤0.001). BCVA improved in 50.6%, remained static in 39.5 % and deteriorated in 9.9% patients after 6 month of treatment. The Baseline TMV (mean and sD) were 9.26±1.83, 10.4±2.38, 11.5±3.05, 8.89±0.75 and 9.47±1.98 mm3 for different OCT patterns, sT (sponge like thickening), CMO (cystoid macular edema), sFD (subfoveal detachment), VMIA (Vitreo macular interface abnormality) and average TMV respectively (P =0.04). After 6 months of laser treatment, the mean TMV decreased from 9.47±1.98mm3 to 8.77±1.31mm3(P =0.01). In sT there was significant decrease in TMV, P =0.01, Further within these groups at 6 months, they were significantly different, P =0.01. · CONCLUsION: OCT showed the different morphological variant of CsME while the response of treatment is different. TMV decreased the most and hence showed the improvement in vision after 6 months of laser treatment. In the era of Anti vascular endothelial growth factors (VEGFs), efficacy of laser seems to be in shadow but it is still first line of treatment in developing nation like Nepal where antiVEGFs may not be easily available and affordable.

1.产EsBL菌与非产EsBL菌中整合子的分布及其相互关系背景近年来,人们发现整合子的水平传播是导致耐药性快速转移的主要原因。整合子是通过位点特异性重组而将开放阅读框集合在一起的一种基因元件,整合子能通过确保基因的准确表达而将这些开放阅读框转变成功能性的基因。EsBL基因常与其他耐药基因共同存在于同一质粒上,导致细菌较强的耐药性,对临床的抗感染治疗带来了巨大的挑战。当前,有关EsBL基因的研究已相当普遍,但是对产EsBL菌中整合子的分布情况的研究还相对较少。目的本实验以中南大学湘雅三医院临床分离的大肠埃希菌及肺炎克雷伯菌为研究对象,对产EsBL菌与非产EsBL菌中的整合子进行研究。本研究的目的是研究整合子与EsBL基因的分布情况与细菌耐药的关系,以及在产EsBL菌株与非产EsBL菌株中整合子的分布的不同导致的细菌耐药性的差异,同时分析在含有整合子的细菌中EsBL基因对细菌耐药性的影响。方法收集中南大学湘雅三医院2009年6月到2009年12月的大肠埃希菌和肺炎克雷伯菌共172株,采用PCR对Ⅰ类整合酶基因进行检测,对整合子的可变区进行测序并用Alu Ⅰ酶进行限制性片段长度多态性分析。结果产EsBL菌株中Ⅰ类整合子阳性率为69%,非产EsBL菌株中Ⅰ类整合子阳性率为40.2%,差异有统计学意义,PsBL菌株中,整合子阳性菌株对12种抗菌药的耐药率高于整合子阴性菌株;在非产EsBL菌株中,整合子阳性菌株仅有8种抗菌药的耐药率高于整合子阴性菌株。在对整合子和EsBL基因之间的关系进行研究的另一实验结果显示：在整合子阳性菌株中,产EsBL菌株有9种抗菌药的耐药率高于非产EsBL菌株;在整合子阴性菌株中,产EsBL菌株有8种抗菌药的耐药率高于非产EsBL菌株。在整合子阳性菌株中,有84%的菌株可检测到可变区,仍有15.9%菌株则无法测得。在整合子的可变区中未发现EsBL基因,整合子与EsBL基因的共同存在情况仍有待于进一步研究探讨。我们发现在整合子存在多种耐药基因盒,而drfA17和aadA7同时存在的情况较多(41.4%)。有36.2%的菌株携带多个整合子。结论 EsBL基因与Ⅰ类整合子在细菌中可以同时存在,并且它们的同时存在提高了细菌的多重耐药水平。 2.在抗生素选择性压力下的整合酶表达. 背景在选择性压力存在的情况下,细菌能够通过遗传变异诱发一系列的反应来获得选择优势。这些反应包括sOs反应、一般的压力反应、热休克反应和严紧反应,这些反应都能够影响易错聚合酶的调节功能。在不同的选择性压力下,细菌对抗生素的耐药基因一旦产生,就可以通过整合子的位点特异性重组机制在不同的菌株之间进行传播,从而导致新的耐药菌株的产生。亚抑菌浓度的抗生素可以诱发细菌的耐药性,另外也可以通过类似sOs反应和一般压力反应的遗传保护机制来诱导。当诱导因素存在时,细菌能够通过改变自身基因的表达来适应这些压力。目的整合酶基因在细菌耐药性基因水平转移的过程中具有重要作用,本研究旨在分析在亚抑菌浓度的作用下,不同抗生素对整合子中整合酶表达的的影响。方法在已收集并对可变区进行测序的Ⅰ类整合子阳性的细菌中,选出两株大肠埃希菌(E9和E22)作为研究对象。我们选择三种抗生素作为诱导药物,三种抗生素分别为甲氧苄啶(一种化疗药物)、链霉素(氨基糖苷类)及左氧氟沙星(人工合成的氟喹诺酮类)。分别测得三种抗生素对两株大肠埃希菌的MIC。用含有低于MIC的抗生素浓度的LB培养基连续地培养细菌4个月,同时用不含抗生素的LB培养基培养的细菌作为阴性对照。为了给细菌提供充足的营养,每48小时更换一次培养基。在细菌生长的过程中,每一个月留一次细菌进行鉴定。最后对不同4批细菌的整合酶表达进行定量测定。测定时以细菌的16s rDNA为内参,采用ΔΔCt法计算出整合酶的相对表达量。结果对于两株实验菌株,左氧氟沙星组整合酶表达升高最多。通过4个月的培养,E9的整合酶的表达量升高了77.7倍,E22的表达量则升高了18.25倍。结果说明,在左氧氟沙星的选择性压力下,细菌整合子中整合酶的表达水平明显提高。而在甲氧苄啶组,两株细菌的整合酶表达水平均无明显改变。在链霉素组,两株细菌的整合酶表达水平有一定升高,但升高程度低于左氧氟沙星组,其中E9整合酶的表达量升高了13.73倍,E22整合酶的表达量升高了3.7倍。而在两株细菌的阴性对照组中,整合酶的表达量变化缺少规律性,但改变量均较小。其中,E9整合酶的表达量升高了4.2倍,E22整合酶的表达量升高了1.9倍。结论在长期的选择性压力作用下,整合子中整合酶能被不同程度的诱导,而诱导的程度则与抗生素的种类和菌株本身有关。

目的分析铜绿假单胞菌中Ⅰ类整合子可变区的长度对Ⅰ类整合酶基因(intⅠ1)表达水平的影响,以探讨整合酶表达的调控机制。方法将25株临床分离的Ⅰ类整合子阳性铜绿假单胞菌按可变区的长度分为A、B组(A组:1000～2000bp;B组:>2000bp),提取细菌的总RNA并逆转录为cDNA,用实时荧光定量PCR法测定两组细菌intⅠ1mRNA的相对表达量,并比较具有不同长度可变区的细菌中intⅠ1基因的平均表达水平。结果两组细菌中intⅠ1mRNA的相对表达量差异有统计学意义(PsrRNA的Ct值均值及⊿Ct值分别为:17.25、10.34、6.91,B组细菌intⅠ1和16srRNA的Ct值均值及⊿Ct值分别为:18.91、9.47、9.45,⊿⊿Ct为5.81;A组细菌的intⅠ1mRNA平均表达水平是B组的5.81倍。结论在临床分离的Ⅰ类整合子阳性的铜绿假单胞菌中,短可变区组整合酶表达水平较高,而长可变区组整合酶表达水平较低,提示Ⅰ类整合子中整合酶的表达水平可能受整合子可变区基因盒数目的影响,从而影响整合子继续捕获基因盒的能力。

The middle and upper atmosphere of Earth displays many large-scale oscillations in several parameters. Of these oscillations, Atmospheric Oscillation (AO) and semi-annual Oscillation (sAO) are prominent ones. In this work, we have analyzed the Very Low Frequency/Low Frequency (VLF/LF) data from two of the transmitters of frequency 45.9 kHz at NsY, sicily, Italy and 37.5 kHz at Grindavik, Iceland. The selected Trasmitter-Receiver Great Circle Path (TRGCP) is middle latitude which is marine in case of Grindavik TRGCP and terrestrial in case of NsY TRGCP. The VLF/LF signals are bounced back and forth from D-layer of ionosphere (altitude at ~65 km during day and ~95 km during night) in Earth-ionospheric waveguide. This infers the presence of atmospheric oscillations as a consequence of change in ionization and recombination rates. Many works related to AO and sAO are mostly done only for equatorial region of the ionosphere and authors have reported the elegant dominancy of AO and sAO in the VLF/LF amplitude pattern over years. To our surprise, in our work it is seen that not only AO and sAO but also other oscillations are necessary to model the oscillation pattern of middle latitude ionosphere.
