Serotonin regulates brain-derived neurotrophic factor expression in select brain regions during acute psychological stress

Previous studies suggest that serotonin （5-HT） might interact with brain-derived neurotrophic factor （BDNF） during the stress response. However, the relationship between 5-HT and BDNF expression under purely psychological stress is unclear. In this study, one hour before psychological stress exposure, the 5-HT1A receptor agonist 8-OH-DPAT or antagonist MDL73005, or the 5-HT2A receptor agonist DOI or antagonist ketanserin were administered to rats exposed to psychological stress. Immunohistochemistry and in situ hybridization revealed that after psychological stress, with the exception of the ventral tegmental area, BDNF protein and mRNA expression levels were higher in the 5-HT1A and the 5-HT2A receptor agonist groups compared with the solvent control no-stress or psychological stress group in the CA1 and CA3 of the hippocampus, prefrontal cortex, central amygdaloid nucleus, dorsomedial hypothalamic nucleus, dentate gyrus, shell of the nucleus accumbens and the midbrain periaqueductal gray. There was no significant difference between the two agonist groups. In contrast, after stress exposure, BDNF protein and mRNA expression levels were lower in the 5-HT1A and 5-HT2A receptor antagonist groups than in the solvent control non-stress group, with the exception of the ventral tegmental area. Our findings suggest that 5-HT regulates BDNF expression in a rat model of acute psychological stress.

HCV replication in PBMC and its influence on interferon therapy

AIM: To study hepatic virus C (HCV) RNA and HCV proteinexpression in peripheral blood mononuclear cells (PBMCs)of patients with HCV infection, and explore the relationshipbetween the HCV RNA in the PBMCs and response tointerferon (IFN) ***: Type-specific primers were designed and RT-nested PCR was used to detect the plus- and minus- strandsof HCV RNA in PBMCs of 54 patients with HCV infection;Indirect immunofluorescence assay was applied to identifyHCVNS5 protein expression in PBMCs; 6 month-, 3 MU-IFNregiment was administrated to observe the responses toIFN in 35 chronic hepatitis C patients with different HCVRNA status in ***: HCV plus strand RNA was found in 10 of 19(52.6 %) acute hepatitis C patients and 22 of 35 (62.9 %)chronic hepatitis C patients. HCV minus strand RNA wasdetected in 14 of 35 (40.0 %) chronic hepatitis C patients,but only one patient (5.3 %) with acute HCV infection wasfound to be minus HCV RNA positive. Though no HCV NS5protein expression was found in the examined 10 cases ofacute HCV infection, it was positive in 17 of 20 (85.0 %)chronic hepatitis C patients by indirect immunofluoresenceassay. There are significant differences of positive rate of theminus-strand and HCVNS5 protein between acute and chronichepatitis C groups(u=2.07, P＜0.05and u=4.43, P＜0.01respectively). The patients with minus-strand HCV RNAshowed a significantly lower 6-month sustained response (SR-6) to IFN compared to those without minus-strand HCVRNAin PBMCs (biologically 14.3 % vs 42.8 %, X2=4.12, P＜0.05and virologically 7.1% vs23.9 %, X2=4.24, P＜0.05).CONCLUSION: HCV is capable of infecting and replicatingin PBMCs, and HCVNS5 protein was expressed in *** patients with minus strand HCV RNA in PBMCs showeda significantly lower 6-month sustained response to IFN,suggesting that minus-strand HCV RNA in PBMCs may beone of the factors influencing response to IFN therapy.

Anti-HBV effect of TAT-HBV targeted ribonuclease

AIM: To prepare and purify TAT-HBV targeted ribonuclease fusion protein, evaluate its transduction activity and investigate its effect on HBV replication in 2.2.15 ***: The prokaryotic expression vector pTAT containing TR gene was used in transforming E. coli BL21(DE3) LysS and TR was expressed with the induction of IPTG. The TAT-TR fusion protein was purified using Ni-NTA-agrose and PD-10 desalting columns, and analyzed by SDSPAGE. Transduction efficiency of TAT-TR was detected with immunofluorescence assay and the concentration of HBeAg in the supernatant of the 2.2.15 cells was determined via solid-phase radioimmunoassay (spRIA). MTT assay was used to detect the cytotoxicity of ***: The SDS-PAGE showed that the TAT-TR fusion protein was purified successfully, and the purity of TAT-TR was 90 %. The visualization of TAT-TR by immunofiuorescence assay indicated its high efficiency in transducing 2.2.15 *** result suggests that TAT-TR could inhibit the replication of HBV effectively, it didn′t affect cell growth and had no ***: TAT-TR possesses a significant anti-HBV activity and the preparation of TAT-TR fusion protein has laid the foundation for the use of TR in the therapeutic trial of HBV infection.

Antidepressant Effects of Electroconvulsive Therapy Unrelated to the Brain＇s Functional Network Connectivity alterations at an Individual Level

Background： Electroconvulsive therapy （ECT） can alleviate the symptoms of treatment-resistant depression （TRD）. Functional network connectivity （FNC） is a newly developed method to investigate the brain＇s functional connectivity patterns. The first aim of this study was to investigate FNC alterations between TRD patients and healthy controls. The second aim was to explore the relationship between the ECT treatment response and pre-ECT treatment FNC alterations in individual TRD patients. Methods： This study included 82 TRD patients and 41 controls. Patients were screened at baseline and after 2 weeks of treatment with a combination of ECT and antidepressants. Group information guided-independent component analysis （G1G-ICA） was used to compute subject-specific functional networks （FNs）. Grassmann maniibld and step-wise forward component selection using support vector machines were adopted to perform the FNC measure and extract the functional networks＇ connectivity patterns （FCP）. Pearson＇s correlation analysis was used to calculate the correlations between the FCP and ECT response. Results： A total of 82 TRD patients in the ECT group were successfully treated. On an average, 8.50 ~ 2.00 ECT sessions were conducted. After ECT treatment, only 42 TRD patients had an improved response to ECT （the Hamilton scores reduction rate was more than 50%）, response rate 51%. 8 FNs （anterior and posterior default mode network, bilateral frontoparietal network, audio network, visual network, dorsal attention network, and sensorimotor network） were obtained using GIG-ICA. We did not found that FCPs were significantly different between TRD patients and healthy controls. Moreover, the baseline FCP was unrelated to the ECT treatment response. Conclusions： The FNC was not significantly different between the TRD patients and healthy controls, and the baseline FCP was unrelated to the ECT treatment response. These findings will necessitate that we modify the experimental scheme to explore

Thickness and sp3 bond modulation of（Ti/tetrahedral amorphous carbon）_n multilayer coatings

The durability of wear-resistant tetrahedral amorphous carbon（ta-C） coatings is intimately linked to their thickness and the concentration of *** enhance the lifespan of the coatings,we have prepared a multilayer coating consisting of alternating layers of Ti and ta-C,denoted as（Ti/ta-C）_n,using arc ion plating.A multi-layer structure in conjunction with stage cooling is employed to enhance the thickness and promote *** results indicate that the ID/IGvalue,representing the ratio of the intensity of the D peak to that of the G peak in Raman spectroscopy,decreases from 0.85 when no stage cooling is applied to 0.35 when stage cooling is employed,suggesting an enhancement in the fraction of sp3bonds within the *** obtained（Ti/ta-C）_n multilayer coating without peeling off has a maximum thickness of 1.14 μm,which is almost double that of the ta-C coating single layer *** the same time,the coating has a low coefficient of friction,down to 0.069,and maintains excellent friction properties with a wear rate of 3.4×10-6mm3·N-1·*** results conclusively show that the preparation technology can enhance both thickness and sp3bonding in ta-C coatings,significantly expanding their utility across tools,molds,and precision engineering components.

To the Editor: Auditory verbal hallucination (AVH) is defined as the hearing of voices in the absence of any external auditory stimulus.[1,2,3] Most AVH content is related to the experiencer, and these events usually cause serious distress. AVHs can occur in patients with a broad range of mental disorders and neurological diseases, such as schizophrenia, bipolar disorder (BP), major depressive disorder (MDD), post-traumatic stress disorder (PTSD), borderline personality disorder (BPD), and in healthy individuals.[2,3,4]
