Proteomic and metabolomic profiling underlines the stage- and time-dependent effects of high temperature on grape berry metabolism

Climate change scenarios predict an increase in mean air temperatures and in the frequency,intensity,and length of extreme temperature events in many wine-growing regions *** elevated temperature has detrimental effects on berry growth and composition,it threatens the economic and environmental sustainability of wine *** Cabernet Sauvignon fruit-bearing cuttings,we investigated the effects of high temperature(HT)on grapevine berries through a label-free shotgun proteomic analysis coupled to a complementary metabolomic *** the 2,279 proteins identified,592 differentially abundant proteins were found in berries exposed to *** gene ontology categories“stress,”“protein,”“secondary metabolism,”and“cell wall”were predominantly altered under *** temperatures strongly impaired carbohydrate and energy metabolism,and the effects depended on the stage of development and duration of *** amounts correlated poorly with protein expression levels in HT berries,highlighting the value of proteomic studies in the context of heat ***,this work reveals that HT alters key proteins driving berry development and ***,we provide a list of differentially abundant proteins that can be considered as potential markers for developing or selecting grape varieties that are better adapted to warmer climates or extreme heat waves.

Grapevine is a perennial crop often cultivated by grafting a scion cultivar on a suitable *** influence scions,particularly with regard to water uptake and ***,one of the possibilities to adapt viticulture to the extended drought stress periods is to select rootstocks conferring increased tolerance to ***,the molecular mechanisms associated with the ability of rootstock/scion combination to influence grape berry metabolism under drought stress are still poorly *** transcriptomic changes induced by drought stress in grape berries(*** noir)from vines grafted on either 110R(drought-tolerant)or 125AA(drought-sensitive)rootstock were *** experiments were conducted in the vineyard for two years and two grape berry developmental stages(50%and 100%veraison).The genome-wide microarray approach showed that water stress strongly impacts gene expression in the berries,through ontology categories that cover cell wall metabolism,primary and secondary metabolism,signaling,stress,and hormones,and that some of these effects strongly depend on the rootstock ***,under drought stress,berries from vines grafted on 110R displayed a different transcriptional response compared to 125AA-concerning genes related to jasmonate(JA),phenylpropanoid metabolism,and pathogenesis-related *** data also suggest a link between Ja.a.d secondary metabolism in water-stressed ***,genes related to secondary metabolism and JA are more induced and/or less repressed by drought stress in the berries grafted on the drought-sensitive rootstock *** rootstock-dependent gene expression changes are relevant for berry composition and sensory properties.

Evaluation of Hemostatic and Antihemolytic Effects of Aqueous Extract of Garcinia kola (Clusiaceae) Fresh Seeds

Objectives: Garcinia kola seeds (EAgk) (Clusiaceae) contain coagulant components use to pretreat aquaculture wastewater in Nigeria. This work was undertaken to assess the effects of EAgk on blood coagulation because of its contents. Methods: Five groups of four rats of both sexes (200 ± 5 g) received orally distilled water, phytomenadione (15 mg/kg b.w.) and garcinia kola seeds extract (250, 500 and 1000 mg/kg b.w.) for four days and after this period, bleeding time was measured by tail hemorrhage model. Activated partial thromboplastin time (aPTT) and platelet count were determined by coagulometer and hematological analyzer respectively. Blood red cells protection of EAgk was measured by the methods of 2,2’-Azobis(2-methylpropionamidine) dihydrochloride (AAPH)-induced hemolysis and inhibition of 2,2-diphenyl-1-picryl hydrazyl (dPPH) radical and hydrogen peroxide. Results: EAgk at 1000 mg/kg b.w. induced a significant decrease in bleeding time from 420 ± 27.3 s to 157.5 ± 18.9 s. The platelet count of group of rat treated with EAgk (1000 mg/kg b.w.) increased significantly (p 0.05, n = 4). EAgk inhibited hemolysis, dPPH radical and hydrogen peroxide with IC50 values of 1.02 ± 0.2;3.2 × 10?2 ± 0.12 and 1.01 ± 0.5 mg/mg respectively. Conclusions: EAgk exhibits hemostatic effects, which could justify the presence of coagulant components in the extract. Erythrocytes’ protective activity of Garcinia kola seeds may be due to free radicals and hydrogen peroxide inhibition.

Fish is rich in essential nutrients but can remain a potential source of food poisoning. A study was, therefore, carried out with the general objective of highlighting the probable biological and chemical dangers associated with the consumption of fish and freshwater fruits from the Guéssabo river. The present study was conducted on 50 carp fish, 50 mackerel fish, 50 frogs and 50 mollusks caught in the Guéssabo River. Microorganisms were enumerated by conventional tests and heavy metals were investigated by Inductively Coupled Plasma Optical Emission Spectrometry. The enumeration showed high levels of thermotolerant Coliforms (2.5 × 104), Enterobacteriaceae (3.4 × 104), S. aureus (2.3 × 103) and Yeast/Molds (2.1 × 104), in all samples the standards were not in conformity with the required standards. High levels of Al (7230 μg/kg) and Cd (21.57 μg/kg) were observed in carp fish and these values are above the standard. In conclusion, the fish caught in the Guéssabo River could be a health risk factor for the consumer.

An AZ41 magnesium alloy in the hot-rolled condition without further thermomechanical processing to modify its microstructure was investigated to establish its suitability for use within a superplastic forming process and to establish optimum forming *** was assessed using elevated temperature tensile testing and hot gas bulging,across a range of strain rates(1×10^(−1)−1×10^(−3)s^(−1))and temperatures(350−450℃).Circle grid analysis with gom.Aramis cameras was used to understand peak strains and material thinning in relation to industrial forming *** forming EBSd and STEM analysis was conducted to understand the mechanisms responsible for the materials formability,with dynamic recrystallization being clearly *** elongation of 520%was achieved at 450℃ and 1×10^(−3)s^(−1);industrially relevant elongation was achieved at 1×10^(−2) s^(−1) at both 450℃(195%)and 400℃(170%).

Spatial and Temporal Patterns of In Situ Sea Surface Temperatures within the Gulf of Mexico from 1901-2010

This work examines the spatial and temporal patterns of seasonal sea surface temperatures (SSTs) across the Gulf of Mexico (gom. for the period 1901-2010. The Extended Reconstructed Sea Surface Temperature, version 4 (ERSST.v4), dataset was selected for this study over other reconstructions because of its 2° × 2° grid cell spatial resolution, its recent update to adjust for known biases in SST observations, and its ability to be compared to other in situ studies of gom.SSTs. The monthly ERSST.v4 data were averaged seasonally for each year and grid cell in the gom. Seasonal SST trends were then calculated for each grid cell with varying start dates (e.g., 1901-2010, 1911- 2010) to account for nonlinear SST changes over the study period. Results indicate that the gom.SSTs closely resemble those of global annual temperature trends: SSTs warmed from 1901 to ~1940, followed by a period with little trend or a slight cooling until the mid-1970s, and then a warming afterwards through 2010. The spatial patterns and magnitudes of SST changes, however, varied by season and location within the gom. The spatial patterns involved gradients with latitudinal and/or longitudinal components: a southwest-northeast (warmer-cooler) gradient in winter, an east-west (warmer-cooler) gradient in spring and fall, and a northwest-southeast (warmer-cooler) gradient in summer. The magnitude of SST changes tended to be largest in summer, followed by spring, fall, and winter. The long-term (1901-2010) SST trends were significant throughout the gom.in summer and fall, but only significant towards the southwestern gom.in winter and spring. These results have implications in discussion of climate change and its impacts on tropical activity in the gom.Basin.

Predict sandstone distribution by integrated study of deformed shale using borehole image and seismic data,a case study from northern Gulf of Mexico
