Background/objecti.e: A nati.nwi.e survey on the resi.tance to fi.st li.e anti.tuberculosi. (anti.TB) drugs was conducted i. Ghana from 2007-2008 by Noguchi.Memori.l i.sti.ute for Medi.al Research i. collaborati.n wi.h the Nati.nal Tuberculosi. Control Programme. We ai.ed to characteri.e mycobacteri.l speci.s causi.g pulmonary tuberculosi. (PTB) and determi.e the resi.tance pattern to fi.st li.e anti.TB drugs among newly di.gnosed and previ.usly treated PTB pati.nts i. Ghana. Methods: Two sputum samples from consented new smear posi.i.e PTB pati.nts who had never been treated for TB or had been on anti.TB treatment for less than a month and pati.nts who had been treated for TB previ.usly for more than a month i. selected di.gnosti. centres nati.nwi.e were collected for culture, i.enti.i.ati.n and drug suscepti.i.i.y test. Culture posi.i.e i.olates were tested agai.st streptomyci. (S), i.oni.zi. (H), ri.ampi.i. (R) and ethambutol (E) usi.g the si.pli.i.d proporti.n method and li.e probe assay (LPA). The LPA was performed i. mi.-2017. Results: Among 410 samples, 345 posi.i.e cultures were obtai.ed and i.enti.i.d as Mycobacteri.m tuberculosi. complex (MTBC). Of the 345 i.olates, 133 were further di.ferenti.ted by GenoType MTBC®as M. tuberculosi., 126 (94.7%) and M. afri.anum 7 (5.3%). The overall drug resi.tance patterns were as follows: 43/345 (12.5%), 6/345 (1.7%), 9/345 (2.6%) and 71/345 (20.6%) were resi.tant to H, R, E and S respecti.ely and 5/345 (1.4%) were multi.drug resi.tant (MDR). Conclusi.n: The results i.di.ate hi.h levels of resi.tance to S and H among new and previ.usly treated TB pati.nts. We recommend adequate survei.lance systems i.cludi.g peri.di. nati.nal anti.TB drug resi.tance surveys.

Background: To better understand the extent of the magni.ude of tuberculosi. (TB) and Human i.munodefi.i.ncy Vi.us (Hi.) co-i.fecti.n i. Ghana, a baseli.e study was conducted to establi.h the nati.nal prevalence of the dual i.fecti.n. The study ai.ed to determi.e the most prevalent Hi. serotype (Hi.-1 or Hi.-2) i. TB pati.nts (new and old cases);genotype mycobacteri.l speci.s causi.g TB/Hi. co-i.fecti.n and determi.e thei. drug suscepti.i.i.y patterns. Methods: Sputum and dri.d blood samples were collected from 503 TB pati.nts from 67 health faci.i.i.s nati.nwi.e between December 2007 and November 2008. All samples were processed for mycobacteri.l and Hi. testi.g usi.g conventi.nal and molecular methods. Results: A total of 517 pai.ed sputum samples were recei.ed from 517 pati.nts. A total 503 pati.nts [335 (66.6%) males;168 (33.4%) females] had at least one culture posi.i.e sample. Majori.y (93.0%) of the pati.nts were new cases whi.e 7.0% were old cases. All 503 TB i.olates were Mycobacteri.m tuberculosi. complex. Of 503 blood samples, 74 were posi.i.e for Hi. (14.7%), compri.i.g 71 (14.1%) and 3 (0.6%) for Hi.-1 and Hi.-1 & 2 respecti.ely;none was posi.i.e for Hi.-2 alone. The seroprevalence of Hi. i. newly di.gnosed TB pati.nts and those already on treatment, was 69/468 (14.7%) and 5/35 (14.3%) respecti.ely (p > 0.05). Di.ferenti.ti.n of i.olates from TB/Hi. co-i.fected pati.nts showed that 70/74 (94.6%) were Mycobacteri.m tuberculosi. whi.e 4/74 (5.4%) were Mycobacteri.m afri.anum. Monoresi.tance to i.oni.zi. and ri.ampi.i. were 4/74 (5.4%) and 1/74 (1.4%) respecti.ely;resi.tance to both drugs (multi.drug resi.tant-MDR) was not observed. Si.ty ni.e (93.2%) i.olates were suscepti.le to both drugs. Conclusi.n: The prevalence of Hi. i.fecti.n i. TB pati.nts was 14.7%. Background: The di.gnosi. and treatment of acti.e tuberculosi. and the detecti.n/management of latent tuberculosi. i.fecti.n (LTBi. cases are the two mai. strategi.s for the TB control, parti.ularly i. endemi. countri.s. Tuberculi. ski. test (TST) and i.terferon Gamma Release Assays (i.RAs) are tools for detecti.n of LTBi. The objecti.e of thi. study was to evaluate the performance of the TST and Quanti.ERON-TB Gold Plus® (QTF-Plus) and to i.enti.y a threshold for TST i. best agreement wi.h QTF-Plus for LTBi.detecti.n i. a hi.h TB burden setti.g. Methods: i. July 2020, a cross-secti.nal analyti.al study was performed for QFT-Plus usi.g blood samples and TST i. 101 i.di.i.uals wi.h a hi.h ri.k of TB li.i.g i. Bobo-Di.ulasso, Burki.a Faso. A crude compari.on between both tests was done and recei.er operati.g characteri.ti. curve was generated to determi.e TST’s threshold. TST sensi.i.i.y, speci.i.i.y, predi.ti.e values and accuracy were calculated. Adjusted agreement between TST and QFT-Plus was evaluated. Results: Wi.h the mi.i.um threshold of posi.i.i.y set at 5 mm for TST, the overall agreement between the latter and QFT-Plus was poor wi.h a Kappa coeffi.i.nt (κ) rated at 0.319 (95% Ci. 0.131 - 0.508). Thi. cut-off yi.lded a sensi.i.i.y of 94.12% (95% Ci. 88.53 - 99.71), and very poor speci.i.i.y of 36.4% (95% i.: 25.0 - 47.80). However, an adjusted cut-off set at 11 mm gave a better speci.i.i.y of 72.73% (95% Ci. 62.1 - 83.30) of TST and i.proved the PPV (86%). Moreover, concordance between both tests was i.proved wi.h κ at 0.56 (95% Ci. 0.385 - 0.728) and 80.20% of accuracy. Factors associ.ted wi.h di.cordance between TST (11 mm) and QFT-Plus results were BCG vacci.ati.n, OR = 7.53 (95% Ci. 1.43 - 139.25), p = 0.05 and chroni. cough, OR = 5.07 (95% Ci. 1.27 - 20.43), p = 0.01. Conclusi.ns: Thi. study showed that usi.g a mi.i.al cut-off of 11mm for TST si.ni.i.antly i.proved the concordance between QTF-Plus (i.RA) and TST. Usi.g the cut-off TST of
