Evaluation of acute and subacute toxicity of whole-plant aqueous extract of Vernonia mespilifolia Less. in Wistar rats

Objective： This study investigated the acute and subacute toxicity of whole-plant aqueous extract of Vernonia mespilifolia Less. （AEVM） in rats for evaluating its safety profile. Methods： AEVM for the acute （2000 and 5000 mg/kg） and subacute （200, 400 and 600 mg/kg） toxicity studies was administered orally to rats according the guidelines 425 and 407 of Organization for Economic Cooperation and Development, respectively. Food and water intake as well as body and organ weight of animals were recorded. Signs of toxicity were assessed, and hematological, biochemical and histopathological analyses were performed. Results： In the acute toxicity study, a single dose of the aqueous extract at 2000 or 5000 mg/kg caused no mortality in the animals, suggesting that the median lethal dose is greater than 5000 mg/kg. In the subacute toxicity study, administration of the extract for 28 d, at all doses, caused no significant changes in the body weights or organ weights of rats in the treated groups when compared with the control group. In addition, hematological and biochemical parameters also revealed no toxic effects of the extract on rats. Histological sections of the heart, liver and kidney from test animals showed no signs of degen- eration. Conclusion： These results showed that AEVM at dosage levels up to 600 mg/kg is nontoxic and could also offer protection on some body tissues. AEVM could, therefore, be considered safe.

Abdominal pain and bowel habits alterations are common symptoms in the general population. The investigation to differentiate organic from functional bowel disorders represents a considerable burden both for patients and public health service. The selection of patients who should undergo endoscopic and/or radiological procedures is one of the key points of the diagnostic process, which should avoid the abuse of invasive and expensive tests as well as the underestimation of potentially harmful diseases. Over the cominci years,dinidans and researchers will be challenged to develop strategies to increase the patient＇s compliance and to reduce the economic and social costs of the intestinal diseases.

The Mesozoic–Cenozoic uplift history of South Tianshan has been reconstructed in many ways using thermochronological analyses for the rocks from the eastern Kuqa Depression. The main difference in the reconstructions concerns the existence and importance of Early Cretaceous and Paleogene tectonic activities, but the existence of a Cenozoic differential uplift in the Kuqa Depression remains enigmatic. Here, we present new apatite fission-track ages obtained for 12 sandstone samples from the well-exposed Early Triassic to Quaternary sequence of the Kapushaliang section in the western Kuqa Depression. The results reveal that there were four pulses of tectonic exhumation, which occurred during the Early Cretaceous（peak ages of 112 and 105 Ma）, Late Cretaceous（peak age of 67 Ma）, Paleocene–Eocene（peak ages at 60, 53, and 36 Ma）, and early Oligocene to late Miocene（central ages spanning 30–11 Ma and peak ages of 23 and 14 Ma）, respectively. A review of geochronological and geological evidence from both the western and eastern Kuqa Depression is shown as follows.（1） The major exhumation of South Tians Shan during the Early Cretaceous was possibly associated with docking of the Lhasa block with the southern margin of the Eurasian plate.（2） The Late Cretaceous uplift of the range occurred diachronically due to the far-field effects of the Kohistan-Dras Arc and Lhasa block accretion.（3） The Paleogene uplift in South Tianshan initially corresponded to the far-field effects of the India–Eurasia collision.（4） The rapid exhumation in late Cenozoic was driven by the continuous far-field effects of the collision between India and Eurasia plates. The apatite fission-track ages of 14–11 Ma suggest that late Cenozoic exhumation in the western Kuqa Depression prevailed during the middle to late Miocene, markedly later than the late Oligocene to early Miocene activity in the eastern segment. The flathead oak borer Coroebus undatus E （Coleoptera： Buprestidae） is one of the primary pests of cork oak Quercus suber L. in the Mediterranean region causing great economic losses to the cork industry. Very little is known about its biology and behavior and, so far, no control measures have been established. We present the results of a pilot study aimed to develop an efficient trapping method for monitoring this harmful pest. In a 3-year field study, purple-colored prism traps baited with a mixture of green leaf volatiles （GLVs） from the host have been shown the most effective combination to catch C. undatus adults （solely females） compared to other trap and lure types tested. Wavelength and reflectance measurements revealed that purple traps exhibit reflectance peak values similar to those found in the abdominal and elytral cuticle of both sexes, suggesting the involvement of visual cues for mate location in this species. The data presented are the first to demonstrate captures of adults of the genus Coroebus by an attractant-based trapping method.

Hybrid drug delivery systems(DDS) have been prepared by grafting poly(NIPAM-co-MPS) chains on multimodal porous silica nanoparticles having an inner mesoporous structure and an outer thin layer of micropores. The hybrid thermoresponsive DDS were fully characterized and loaded with a model drug. The in vitro drug release tests are carried out at below and above the lower critical solution temperature(LCST) of the copolymer. The results have revealed that due to the presence of small diameter(~1.3 nm) micropores at the periphery of the particles, the collapsed globules of the thermoresponsive copolymer above its LCST hinders the complete release of the drug which resulted in a reverse thermoresponsive drug release profile by the hybrid DDS.

AIM: To evaluate the prevalence and determined the common characteristics of patients diagnosed with non- alcoholic fatty liver disease (NAFLD) at the Philippine General Hospital, Manila, from January 1999 to December 2004. METHODS: NAFLD was diagnosed in 134 from a total of 1102 patients, based on clinical, ultrasonographic and/or histopathological findings. Patients with conditions associated with secondary NAFLD were excluded. Chart review was done to note demographics, co- morbid illnesses, physical characteristics, hepatomegaly, aspartate/alanine aminotransferase (AST/ALT) levels, albumin, lipid levels, alkaline phosphatase, prothrombin time, and partial thromboplastin time. Data obtained were analyzed using the statistical program SPSS version 10. RESULTS: Of the 134 patients included, 71% were female and 29% male. Mean patient age was 42.2 years. Sixty percent of patients were obese, 56% had hepatomegaly, and 69% had diabetes. AST levels were elevated in 45% of subjects and ALT levels in 64%. CONCLUSION: The prevalence of NAFLD at our institution was 12.2%. Patients diagnosed appear to be younger in age in contrast to previous studies. Female sex, obesity, elevated liver enzymes, and diabetes were characteristic features of our NAFLD patients, which is comparable to previous studies from other countries.

Protein evolution proceeds by two distinct processes： 1） individual mutation and selection for adaptive mutations and 2） rearrangement of entire domains within proteins into novel combinations, producing new protein families that combine functional properties in ways that previously did not exist. Domain rearrangement poses a challenge to sequence alignment-based search methods, such as BLAST, in predicting homology since the methodology implicitly assumes that related proteins primarily differ from each other by individual mutations. Moreover, there is ample evidence that the evolutionary process has used （and continues to use） domains as building blocks, therefore, it seems fit to utilize computational, domain-based methods to reconstruct that process. A challenge and opportunity for computational biology is how to use knowledge of evolutionary domain recombination to characterize families of proteins whose evolutionary history includes such recombination, to discover novel proteins, and to infer protein-protein interactions. In this paper we review techniques and databases that exploit our growing knowledge of “horizontal” protein evolution, and suggest possible areas of future development. We illustrate the power of the domain-based methods and the possible directions of future development by a case history in progress aiming at facilitating a particular approach to understanding microbial pathogenicity.
