Oncolytic viruses(OVs)are at the forefront of biologicals for cancer *** represent a diverse landscape of naturally occurring viral strains and genetically modified viruses that,either as single agents or as part of combination therapies,are being evaluated in preclinical and clinical *** the field gains momentum,the research on OVs has been shifting efforts to expand our understanding of the complex interplay between the virus,the tumor and the immune system,with the aim of rationally designing more efficient therapeutic ***,the potential of an OV platform is no longer defined exclusively by the targeted replication and cancer cell killing capacities of the virus,but by its contribution as an immunostimulator,triggering the transformation of the immunosuppressive tumor microenvironment(TME)into a place where innate and adaptive immunity players can efficiently engage and lead the development of tumor-specific long-term memory *** we review the immune mechanisms and host responses induced by ssRNA(-)(negative-sense single-stranded RNA)viruses as OV *** focus on two ssRNA(-)OV candidates:Newcastle disease virus(NDV),an avian paramyxovirus with one of the longest histories of utilization as an OV,and influenza A(IAV)virus,a well-characterized human pathogen with extraordinary immunostimulatory capacities that is steadily advancing as an OV candidate through the development of recombinant IAV attenuated platforms.

Lansoprazole prevents experimental gastric injury induced by non-steroidal anti-inflammatory drugs through a reduction of mucosal oxidative damage

AIM:This study investigated the mechanisms of protection afforded by the proton pump inhibitor lansoprazole against gastric injury induced by different non-steroidal anti-inflammatory drugs (NSAIDs) in rats. METHODS: Male Sprague-Dawley rats were orally treated with indomethacin (100 μmol/kg), diclofenac (60 μmol/kg), piroxicam (150 μmol/kg) or ketoprofen (150 μmol/kg). Thirty minutes before NSAIDs, animals were orally treated with lansoprazole 18 or 90 umol/kg. Four hours after the end of treatments, the following parameters were assessed: gastric mucosal PGE2, malondialdehyde (MDA), myeloperoxidase (MPO) or non-proteic sulfhydryl compounds (GSH) levels; reverse transcription-polymerase chain reaction (RT-PcR) of mucosal cOX-2 mRNA; gastric acid secretion in pylorus-ligated animals; in vitro effects of lansoprazole (1-300 μmol/L) on the oxidation of low density lipoproteins (LDLs) induced by copper sulphate. RESULTS: All NSAIDs elicited mucosal necrotic lesions which were associated with neutrophil infiltration and reduction of PGE2 levels. Increments of MPO and MDA contents, as well as a decrease in GSH levels were detected in the gastric mucosa of indomethacin- or piroxicam-treated animals. Indomethacin enhanced mucosal cyclooxygenase-2 expression, while not affecting cyclooxygenase-1. At the oral dose of 18 μmol/kg lansoprazole partly counteracted diclofenac-induced mucosal damage, whereas at 90 μmol/kg it markedly prevented injuries evoked by all test NSAIDs. Lansoprazole at 90 μmol/kg reversed also the effects of NSAIDs on MPO, MDA and GSH mucosal contents, without interfering with the decrease in PGE2 levels or indomethacin-induced cyclooxygenase-2 expression. However, both lansoprazole doses markedly inhibited acid secretion in pylorus-ligated rats. Estimation of Partition coefficients of Fe-S Protein Models： Se Substitutions in Fe4S4cysn

The ScAP method allows a semiquantitative estimation of the partitioning of many solutes between numerous pairs of solvents. The organic solvent-water partition coefficients P are calculated. For FeaS4cysn, the organic solvent-water partition coefficients for l-octanol Po, cyclohexane Pch and chloroform Pcf decay 4.46, 6.25 and 4.60 log units per cys, respectively. The Po is of the same order of magnitude as cDHI （conformationally dependent hydrophobic index） calculations and Pch-cf as computations performed with a method by Leo and Hansch. The logPo-ch-cf mean relative errors are 17%, 25% and-17%, which represent mean and unsigned relative errors of -3% and 20%. On varying the number of cys, the structures show hydrophobic moments indicative of amphipathic compounds. For Se substitutions in Fe4Seacys,, Po-ch-cf drop 4.52, 6.30 and 4.66 exponents per cys. With the reference calculations, Po-ch-cf decay 4.03, 4.80 and 5.76 exponents per cys. For FeaS4 mSemcys4, the calculated similar partition coefficients and hydrophobic moments, and lower distance in dendrogram suggest a role of Fe-Se clusters in physiological processes. The analysis is extended to lysozyme and its secondary structures.

Binding of Mono, Bi and Tridomain Proteins to Zwitterionic and Anionic Vesicles： Asymmetric Location of Anionic Phospholipids in Mixed Zwitterionic/Anionic Vesicles and cooperative Binding

Lysozyme, myoglobin and BSA were used as models of globular proteins covering a wide range of pl. The purpose is to extend the studies to anionic lipid bilayers. Electrostatics is studied in cationic protein adsorption to zwitterionic Pc and anionic mixed Pc/PG SUVs. Protein adsorption is investigated in SUVs along with changes of fluorescence emission spectra. Partition coefficients and cooperativity parameters are calculated. At pl binding obtains maximum while at lower or higher pHs binding decreases. In Gouy-chapman formalism activity coefficient goes with square charge, which deviations indicate asymmetric location of anionic phospholipid in the inner leaflet, in mixed SUVs for lysozyme- and myoglobin-Pc/PG systems, in agreement with experiments and molecular dynamics simulations. Vesicles bind myoglobin anti-cooperatively while lysozyme-BSA cooperativitivey. A model is proposed for both, which composes two protein sub-layers with different structures and properties. Hill coefficient reflects subunit cooperativity of bi and tridomain proteins.

Lay vs. Professional Perspectives: The Sexual and Reproductive Health of Orphans and Vulnerable children in Uganda

Background/Introduction: Orphans and vulnerable children (OVc) in sub-Saharan Africa have been the focus of much popular and academic writing and the growing body of evidence cannot be overlooked. Existing research focuses on projections of OVc numbers, interventions, outcomes and descriptions of care arrangements. A lot of information exists on their poor sexual and reproductive health (SRH) status—most of it quantitative. The purpose of this paper is to present the voice and perceptions of OVc in regards to their SRH. Methods: This was a mixed-methods qualitative study drawing from phenomenology and it used purposive sampling to recruit 129 participants. Results: OVc perceptions and priorities do not always intersect with those of professionals and this has implications for existing and planned interventions. For example some OVc think favorably about early marriages while others have mechanisms of self-protection again SRH harm although this is not usually acknowledged. conclusions: OVc demonstrate agency;therefore positioning them at the heart of service planning and delivery is an effective strategy in increasing the uptake and impact of interventions seeking to address their sexual and reproductive health needs.
