Why endophenotype development requires families

Endophenotypes are heritable quantitative traits that are associated with disease liability, can be measured in both affected and unaffected individuals, and provide much greater power to localize and identify risk genes for mental illness than does affection status alone. Traditionally, endophenotypic markers for psychiatric illnesses include in vivo neuroanatomic and functional magnetic resonance imaging measurements and indices of neurocognitive abilities. However, neurocognitive and neuroimaging measures are by no means the only classes of endophenotypes that could be useful for identifying genes for mental illness. Given the advantages of endophenotype-based strategies for elucidating the genetic underpinnings of psychiatric disorders, it would seem prudent to develop a wide range of putative endophenotypes. In order for a measure to be considered a valid endophenotype, it must meet a number of criteria. Specifically, the trait must (1) have moderate to high heritability, (2) be associated with the illness, (3) be independent of clinical state, and (4) impairment must co-segregate with the illness within a family, with non-affected family members showing impairment relative to the general population. While each of these criteria is critical, the heritability and co-segregation requirements are really what differentiate an endophenotype from a simple biomarker. At this time, one requires an experimental design that includes families to demonstrate both heritability and co-segregation. The assertion that novel endophenotypes can not be fully established without family data does not preclude work in unrelated individuals, rather that unrelated samples will only be able to nominate potential candidate endophenotypes that subsequently need to be confirmed in family-based experiments.

Indicator kriging （IK） is a spatial interpolation technique devised for estimating a conditional cumulative distribution function at an unsampled location. The result is a discrete approximation, and its corresponding estimated probability density function can be viewed as a composition in the simplex. This fact suggested a compositional approach to IK which, by construction, avoids all its standard drawbacks （negative predictions, not-ordered or larger than one）. Here, a simple algorithm to develop the procedure is presented.

The set of probability functions is a convex subset of L1 and it does not have a linear space structure when using ordinary sum and multiplication by real constants. Moreover, difficulties arise when dealing with distances between densities. The crucial point is that usual distances are not invariant under relevant transformations of densities. To overcome these limitations, Aitchison＇s ideas on compositional data analysis are used, generalizing perturbation and power transformation, as well as the Aitchison inner product, to operations on probability density functions with support on a finite interval. With these operations at hand, it is shown that the set of bounded probability density functions on finite intervals is a pre-Hilbert space. A Hilbert space of densities, whose logarithm is square-integrable, is obtained as the natural completion of the pre-Hilbert space.

Malignant melanoma is believed to have the highest rate of cardiac metastasis of any cancer based on autopsy studies,but descriptions of clinical findings,diagnosis,and treatment remain *** particular,the use of immunotherapy has been only sparingly *** present eight cases of malignant melanoma treated at our *** patients displayed symptoms,three were diagnosed on routine surveillance imaging,and one was diagnosed *** of cardiac tumors varied with all four chambers and septum involved between the *** patients underwent genetic profiling:two patients had a CDKN2A variant detected in their cardiac tumor,one patient had a BRAF mutation,and one patient had an NRAS *** patients underwent immunotherapy with either anti-PD-1 and/or anti-CTLA-4 *** report highlights the importance of considering cardiac melanoma in surveillance imaging and the use of immunotherapy for management.
