Regulation of mitochondrial function and endoplasmic reticulum stress by nitric oxide in pluripotent stem cells

mitochondrial dysfunction and endoplasmic reticulum stress(ERS) are global processes that are interrelated and regulated by several stress factors. Nitric oxide(NO) is a multifunctional biomolecule with many varieties of physiological and pathological functions, such as the regulation of cytochrome c inhibition and activation of the immune response, ERS and DNA damage; these actions are dose-dependent. It has been reported that in embryonic stem cells, NO has a dual role, controlling differentiation, survival and pluripotency, but the molecular mechanisms by which it modulates these functions are not yet known. Low levels of NO maintain pluripotency and induce mitochondrial biogenesis. It is well established that NO disrupts the mitochondrial respiratory chain and causes changes in mitochondrial Ca^(2+) flux that induce ERS. Thus, at high concentrations, NO becomes a potential differentiation agent due to the relationship between ERS and the unfolded protein response in many differentiated cell lines. Nevertheless, many studies have demonstratedthe need for physiological levels of NO for a proper ERS response. In this review, we stress the importance of the relationships between NO levels, ERS and mitochondrial dysfunction that control stem cell fate as a new approach to possible cell therapy strategies.

Stem cell pluripotency and differentiation are global processes regulated by several pathways that have been studied intensively over recent years. Nitric oxide(NO) is an important molecule that affects gene expression at the level of transcription and translation and regulates cell survival and proliferation in diverse cell types. In embryonic stem cells NO has a dual role, controlling differentiation and survival, but the molecular mechanisms by which it modulates these functions are not completely defined. NO is a physiological regulator of cell respiration through the inhibition of cytochrome c oxidase. many researchers have been examining the role that NO plays in other aspects of metabolism such as the cellular bioenergetics state, the hypoxia response and the relationship of these areas to stem cell stemness.

mechanical Properties of Terminalia catappa from Ghana

Ghana is rich in forest resources, of whichTerminalia catappa, a wood species of common occurrence is one. Even though sometimes it is used for decorative purposes, however, it grows in the wild. It grows in almost all the regions of the country and does well in the tropics. The parts of the tree such as the leaves, fruits and seeds have been known to be very useful for medicinal and other uses. Some work on the strength properties of the leaves and other parts has also been done, however, strength properties of the wood are yet to be explored. In this study, its mechanical properties such as bending strength, compression parallel to the grain, and shear parallel to the grain properties were determined. The British (BS 373, 1957) and American Society of Testing materials’ specifications (ASTm D143, 1983) using testing methods for small, clear specimens of wood were used in determining the properties. The results showed that at 18% moisture content the wood has a density of 520 kg/m3 with a mean modulus of rupture of 86.04 mpa, compressive strength parallel to the grain of 42.02 mpa, modulus of elasticity of 10,500 mpa, and shear strength parallel to the grain of 16.42 N/mm2. These strength properties are comparable to that of Strombosia glaucescens. Therefore, T. catappa can be used in applications where this species is used.

Overcoming the barrier of nanoparticle production by femtosecond laser ablation in liquids using simultaneous spatial and temporal focusing

There exists an increasing demand of industrial-scale production of high-purity ligand-free nanoparticles due to the continuous development of biomedicine, catalysis, and energy applications. In this contribution, a simultaneous spatial and temporal focusing(SSTF) setup is first proposed for increasing nanoparticle productivity of the eco-friendly pulsed laser ablation in liquids(PLAL) technique. In spite of the fact that femtosecond pulses have proved to achieve higher ablation rates in air than picosecond pulses, in PLAL this is reversed due to the nonlinear energy losses in the liquid. However, thanks to the incorporation of SSTF, the energy delivered to the target is increased up to 70%, which leads to a nanoparticle production increase of a 2.4 factor. This breaks a barrier toward the employment of femtosecond lasers in high-efficiency PLAL.

Quantity and Trends in Streamflows of the malewa River Basin,Kenya

Identification of ompL1 and lipL32 Genes to Diagnosis of Pathogenic Leptospiraspp. Isolated from Cattle

Diagnosis of leptospirosis in Colombia is based on clinical history and serological testing. However, disease symptoms are nonspecific and there is no uniform criteria regarding the qualifications considered positive. Therefore, it is important to identify and characterize genes associated with pathogenicity in native strains for the development of new diagnostic tests and vaccine production. The aim of this study was to identify the ompL1 and lipL32 genes in Leptospira strains isolated from urine samples of cattle. Sixteen strains were obtained from urine samples and, DNA was isolated to perform two Polymerase Chain Reaction (PCR) tests which identified lipL32 and ompL1 genes. As positive control, a reference strain of L. interrogans was used. L. biflexa and Escherichia coli strains were used as a negative control. In 100% of the samples were identified amplicons of 960 bp and 423 bp corresponding to ompL1 and lipL32 genes respectively. Thus, the pathogenic property and conservation of genes in the isolated strains were confirmed. This study is presented as a contribution to the diagnosis of leptospirosis to use these genes as molecular markers of infection. The results of this study might provide clues for future clinical, epidemiological and molecular research leading to implement new diagnostic strategies and to expand knowledge of the pathophysiology of a disease of public health impact on human and animal.

A Comparative Study into the Engineering Properties of Neem Wood from Ghana

A study into the mechanical properties of Neem wood was done to explore the possibility of its usage as a structural material. This paper presents the results of some investigations into the strength properties of Neem (m>Azadirachta indicam> A. Juss.) and compares them with those of Odum (m>milicia excelsam>) and European Oak (m>Quercus m>spp.) which are well known structural wood species in Ghana and Europe, respectively. The properties were determined using British (BS 373, 1957) and American timber testing specifications (ASTm D143, 1983) using testing methods for small, clear specimens of wood. The results showed that at 12% moisture content the wood has a density of 740 kg/m3 with a mean modulus of rupture of 8317 N/mm2, compressive strength parallel to the grain of 318.8 N/mm2, modulus of elasticity of 8644 N/mm2, and shear strength parallel to the grain of 17 N/mm2. The bending strength (mOR) of Neem is about 30% lower than that of Odum and about 2% higher than Oak. The modulus of elasticity of Neem is about 39% lower than Odum and 43% lower than Oak. The strength properties in the present work suggest that the wood of Neem has a potential for utilisation in structural applications.

A qualitative study of childbirth fear and preparation among primigravid women:The blind spot of antenatal care in Lilongwe,malawi

Objectives:This study aimed to explore childbirth fear and childbirth preparation among primigravid women in the late pregnancy from 36 to 40 weeks ***:We purposively recruited 18 primigravid women into in-depth interviews,21 birth companions,and 13 health workers into focus group *** were recruited from two community hospitals'maternity waiting homes in Lilongwe,***-structured interview guides were used to collect data that were analyzed using content ***11 computer software was used to organize the ***:The four categories developed were:"ambivalent pregnancy feelings","dependence on traditional childbirth counseling","inadequate prenatal childbirth instruction"and"inconsistent roles of a birth companion".The findings suggest that primigravid women who were mainly exposed to traditional childbirth mentoring rather than professional care providers,experienced childbirth fear,and lacked proper psychosocial childbirth ***:Childbirth fear among primigravid women emanate from personal;family;ineffective traditional counseling;and inadequate antenatal childbirth *** companions may increase childbirth ***,our findings highlight birth companions as readily available psychosocial support resources among primigravid *** recommend that professional childbirth instruction during antenatal care should be strengthened to surpass traditional childbirth *** and effectiveness of birth companions need to be carefully assessed.

Aflatoxin m1 Contamination of milk and Its Products in Bomet County, Kenya

Aflatoxin m1 (AFm1) is a major carcinogenic compound that may be found in milk and dairy products resulting from ingestion of aflatoxin B1 by dairy animals. The study aimed at determining the level of aflatoxin m1 in milk and milk products from Bomet County. A total of 185 samples (150 raw milk and 35 processed milk and milk products) were randomly collected from milk collection sites and randomly selected milk kiosks respectively. The AFm1 was analyzed using a commercial ELISA kit (Ridascreen, aflatoxin m1 R-Biopharm, Product code, R5812, Darmstadt, Germany). Out of 185 samples investigated, 156 samples were positive for AFm1, an overall contamination rate of 84.32%. The samples with levels higher than the tolerance limit of 0.05 μg/l recommended by Food and Agriculture Organization (FAO) and World Health Organization (WHO) limits were 43.8% mainly contributed by the raw milk compared to processed milk (52.0% versus 8.6%). Processed milk had insignificant level of contamination with aflatoxin m1 (median 0.00 (IQR: 0.00, 0.00 μg/l) with a minimum of 0.00 μg/l and a maximum of 0.69 μg/l. Raw milk showed significant contamination, median 0.09 (IQR: 0.00, 0.50) μg/l with a minimum of 0.00 μg/l and a maximum of 2.93 μg/l. Although there was no significant differences in AFm1 levels with study sites (P = 0.217);the median levels of aflatoxin m1 was high in sites 1, 3, and 7. The sites that had median aflatoxin m1 levels below the WHO/FAO acceptable limits of 0.05 μg/l were sites 2, 4 and 6. Due to high incidence of AFm1 contamination of milk and milk samples in Bomet County, there is need for regular monitoring and regulation of AFm1 contamination in milk and its products in the County.

Intestinal heme absorption in hemochromatosis gene knock-out mice

AIm To investigat the influence of hemochromatosis gene(Hfe) mutation on ^(59)Fe labelled duodenal heme absorption in *** Heme absorption was measured in Hfe wild type and Hfe^((-/-)) mice by the duodenal tied loop and by oral gavage methods. The m RNA expression of heme oxygenase(HO-1), Abcg2 and Flvcr1 genes and levels were determined by quantitative polymerase chain *** Heme absorption was significantly increased in homozygous Hfe^((-/-)) mice despite significant hepatic and splenic iron overload. While duodenal HO-1 mRNA was highly expressed in the wild type and Hfe^((-/-)) heme-treated group following 24 h heme administration, Flvcr1 a mRNA decreased. However, Abcg2 mRNA expression levels in duodenum remained unchanged. CONCLUSION Heme absorption was enhanced in Hfe^((-/-)) mice from both duodenal tied-loop segments and by oral gavage methods. HO-1 m RNA levels were enhanced in mice duodenum after 24 h of heme feeding and may account for enhanced heme absorption in Hfe^((-/-)) mice. Implications for dietary recommendations on heme intake by Hfe subjects to modulate iron loading are important clinical considerations.
